蛹虫草是一种药食两用真菌,具有与冬虫夏草相似的功能,且富硒能力较强。本研究通过大量的人工栽培试验,旨在探究不同浓度Na_2SeO_4对新疆本地蛹虫草子实体生长的影响。试验表明,质量浓度为20 mg/L的Na_2SeO_4对蛹虫草的生长不产生显著影响,但蛹虫草各项生物学指标均随着培养基中外源Na_2SeO_4浓度的增加而呈下降趋势,说明随着外源Na_2SeO_4浓度的增加会对蛹虫草的生长产生抑制效应,当外源Na_2SeO_4质量浓度达到200 mg/L时,生产的蛹虫草已不具备商品价值。由此可见,20 mg/L的质量浓度是以Na_2SeO_4为硒源进行蛹虫草富硒研究的安全浓度。该研究为富硒产品开发寻找新的硒源开辟了新思路,为新疆地区进一步大规模栽培富硒蛹虫草提供一定的参考,但是对以Na_2SeO_4为硒源的最佳富硒浓度还有待于进一步研究。  相似文献

The aim of this study was to isolate and cultivate protoplasts of the green alga Ulva fasciata Delile and subsequently induce them to form a microthallus suspension for algal seed stock. The protoplasts were covered with secreted mucilage following 6 h of culture when viewed with SEM. The mucilage fused to form thick layers during day 1 of culture. Microfibrillar cell walls were deposited into the thick layers of mucilage on the 5th day of culture. An average of about 10% of the freshly isolated protoplasts began to divide at 6–14 days. These protoplasts subsequently developed varied morphologies, depending on the time of collection during the year. Protoplasts isolated from U. fasciata collected in March to June developed frond thalli or microthalli when they were cultured in low or high densities (cells/area), respectively. The microthallus suspension was cultured for more than two years at 10–40 μ mol·m− 2 ·s− 1 . Frond thalli formed when the suspension was cultivated at 100–160 μ mol·m− 2 ·s− 1 . Therefore, microthallus suspension can serve as a seed stock of U. fasciata .  相似文献

Using the mitochondrial DNA miniprep technique, the identity of sixteen morphologically unusual cultures allocated to Phytophthora nicotianae, Phytophthora mexicana or Phytophthora porri was determined by comparison with a library of mtDNA band patterns obtained from reference cultures. Seven cultures were identified as Phytophthora nicotianae (including those assigned to Phytophthora mexicana and Phytophthora porri), six as strains of Phytophthora palmivora with small, ovoid, weakly caducous sporangia, and one as Phytophthora citrophthora. Some cultures of P. nicotianae had a low percentage of caducous sporangia. Percentage sporangium caducity, but not sporangium L : B ratio, is considered a useful taxonomic criterion for separating species morphologically similar to Phytophthora nicotianae. One culture from tobacco in New Zealand had a highly unusual morphology and a unique DNA band pattern, but was not identifiable. One culture from Acacia mearnsii in South Africa had a unique DNA band pattern which was identical to that of an isolate from Annona squamosa from Australia previously identified as Phytophthora palmivora, the precise identity of which is still unclear. The identity of most isolates from diseased durian was found to be Phytophthora palmivora, confirming its role as the main pathogen, but P. nicotianae was also identified from this host. Lipid classes and fatty acid distribution were analysed in the resting sporangium of Synchytrium endobioticum, the causal agent of the potato wart disease. The sporangium contents were shown to have lipid droplets, the major fatty acids there being C16.0, C18.1, and C19.0. The sporangium wall on the other hand was composed of C18.0, C18.1, C18.2, C20.0, and C20.4 fatty acids. A significantly large portion of the sporangium wall lipids contained wax esters with branched chains.  相似文献

利用透射电镜对模式植物水蕨(Ceratopteris thalictroides)孢子囊的早期发育进行研究.结果表明:水蕨的孢子囊是由叶片表皮的原始细胞发育而来,经过横向和纵向分裂形成外套层原始细胞和内部细胞,此过程中各个细胞内线粒体和叶绿体逐渐变大,变发达;之后外套层原始细胞继续纵分裂形成孢子囊壁细胞,内部细胞分裂形成内外两层绒毡层和孢子母细胞,此过程中电子密集物在分裂最为旺盛的细胞内体积最大,数量最多;最后孢子母细胞减数分裂形成四分孢子,此时可见孢子之间以及孢子与原生质团之间均存在着表面膜.内层绒毡层为周原质团绒毡层,外层绒毡层为腺质型绒毡层.水蕨孢子囊的早期发育属于薄囊蕨型发育.  相似文献

本文利用MTT染色法和点接生物测定法对不同温度干燥处理36h后的葡萄生单轴霉孢子囊存活力和致病力进行了检测,并应用单因素试验和正交试验对其孢子囊进行了MTT染色条件的优化。结果表明:葡萄生单轴霉孢子囊MTT染色的最佳条件为温度36℃、MTT浓度 0.05%、染色48h,孢子囊染色率可达83.0%。20℃恒温干燥处理36h显著提高了孢子囊存活力,葡萄生单轴霉孢子囊的蓝色染色率为79.3%,叶片点接发病率为98.9%,显著高于对照的蓝色染色率52.0%和发病率62.7%。MTT染色得到的孢子囊蓝色染色率与点接生物测定法得出的发病率存在很好的线性关系y=1.276 1x-1.939 1,R2=0.996 1,孢子囊的蓝色染色率与叶片点接发病率呈正相关。本研究表明MTT染色法可以用于葡萄生单轴霉孢子囊存活力的快速和准确检测。  相似文献

Abstract: The tapetum surrounds the developing sporogenous tissue, and therefore is suggested to have an important role during sporogerlesis. In pteridophytes details about tapetum development and function during sporangium development are poorly known. The current article analyses the leptospor-angiate development of the pteridophyte Cyrtomium falcatum in terms of functional compartmentalization related to tapetum function. Morphology, development and localization of different compounds during sporangium development are studied. Dur ing C. falcatum leptosporangiate development phases of initia tion, division, differentiation, and maturation are identified. Functional compartments are distinguished by the presence of a callose culture sac during the division and differentiation phase, and a callose spore sac during the differentiation and ma turation phase.  相似文献
