Squalene synthase (SS) catalyzes the biosynthesis of squalene, the first specific intermediate in the cholesterol biosynthetic pathway. To test the feasibility of lowering plasma cholesterol by inhibiting hepatic SS, we generated mice in which SS is specifically knocked out in the liver (L-SSKO) using Cre-loxP technology. Hepatic SS activity of L-SSKO mice was reduced by >90%. In addition, cholesterol biosynthesis in the liver slices was almost eliminated. Although the hepatic squalene contents were markedly reduced in L-SSKO mice, the hepatic contents of cholesterol and its precursors distal to squalene were indistinguishable from those of control mice, indicating the presence of sufficient centripetal flow of cholesterol and/or its precursors from the extrahepatic tissues. L-SSKO mice showed a transient liver dysfunction with moderate hepatomegaly presumably secondary to increased farnesol production. In a fed state, the plasma total cholesterol and triglyceride were significantly reduced in L-SSKO mice, primarily owing to reduced hepatic VLDL secretion. In a fasted state, the hypolipidemic effect was lost. mRNA expression of liver X receptor α target genes was reduced, while that of sterol-regulatory element binding protein 2 target genes was increased. In conclusion, liver-specific ablation of SS inhibits hepatic cholesterol biosynthesis and induces hypolipidemia without increasing significant mortality.  相似文献

Recent studies have emphasized the important role of microRNA (miRNA) clusters and common target genes in disease progression. Despite the known involvement of the miR‐192/215 family in many human diseases, its biological role in Hirschsprung disease (HSCR) remains undefined. In this study, we explored the role of the miR‐192/215 family in the pathogenesis of HSCR. Quantitative real‐time PCR and western blotting measured relative expression levels of miRNAs, mRNAs, and proteins in 80 HSCR patients and 77 normal colon tissues. Targets were evaluated by dual‐luciferase reporter assays, and the functional effects of miR‐192/215 on human 293T and SH‐SY5Y cells were detected by the Transwell assay, CCK8 assay and flow cytometry. MiR‐192/215 was significantly down‐regulated in HSCR tissue samples, and their knockdown inhibited cell migration and proliferation in the human 293T and SH‐SY5Y cell lines. Nidogen 1 (NID1) was confirmed as a common target gene of miR‐192/215 by dual‐luciferase reporter gene assay and its expression was inversely correlated with that of miR‐192/215 in tissue samples and cell lines. Silencing of NID1 could rescue the extent of the suppressing effects by miR‐192/215 inhibitor. The down‐regulation of miR‐192/215 may contribute to HSCR development by targeting NID1.

Engineered iron oxide nanoparticles (IO-NPs) have been used extensively for environmental remediation. It may cause the release IO-NPs to the environment affecting the functions of ecosystems. Plants are an important component of ecosystems and can be used for the evaluation of overall fate, transport and exposure pathways of IO-NPs in the environment. In this work, the effects of engineered ferrihydrite and hematite NPs on the germination and growth of maize are studied. The germination and growth of maize were done with treatments at different concentrations of hematite and ferrihydrite NPs, namely 1, 2, 4, and 6 g/L. Biological indicators of toxicity or stress in maize seedlings were not observed in treatments with engineered hematite and ferrihydrite NPs. In contrast, the NPs treatments increased the growth of maize and the chlorophyll content, except for hematite NPs at 6 g/L, where non-significant effects were found. The translocation of engineered ferrihydrite and hematite NPs in maize stems was demonstrated using confocal laser scanning microscopy.  相似文献

The social environment modulates gene expression, physiology, behaviour and patterns of inheritance. For more than 50 years, this concept has been investigated using approaches that include partitioning the social component out of behavioural heritability estimates, studying maternal effects on offspring, and analysing dominance hierarchies. Recent advances have formalized this ‘social environment effect’ by providing a more nuanced approach to the study of social influences on behaviour while recognizing evolutionary implications. Yet, in most of these formulations, the dynamics of social interactions are not accounted for. Also, the reciprocity between individual behaviour and group‐level interactions has been largely ignored. Consistent with evolutionary theory, the principles of social interaction are conserved across a broad range of taxa. While noting parallels in diverse organisms, this review uses Drosophila melanogaster as a case study to revisit what is known about social interaction paradigms. We highlight the benefits of integrating the history and pattern of interactions among individuals for dissecting molecular mechanisms that underlie social modulation of behaviour.  相似文献

Herbivory is a fundamental process that controls primary producer abundance and regulates energy and nutrient flows to higher trophic levels. Despite the recent proliferation of small‐scale studies on herbivore effects on aquatic plants, there remains limited understanding of the factors that control consumer regulation of vascular plants in aquatic ecosystems. Our current knowledge of the regulation of primary producers has hindered efforts to understand the structure and functioning of aquatic ecosystems, and to manage such ecosystems effectively. We conducted a global meta‐analysis of the outcomes of plant–herbivore interactions using a data set comprised of 326 values from 163 studies, in order to test two mechanistic hypotheses: first, that greater negative changes in plant abundance would be associated with higher herbivore biomass densities; second, that the magnitude of changes in plant abundance would vary with herbivore taxonomic identity. We found evidence that plant abundance declined with increased herbivore density, with plants eliminated at high densities. Significant between‐taxa differences in impact were detected, with insects associated with smaller reductions in plant abundance than all other taxa. Similarly, birds caused smaller reductions in plant abundance than echinoderms, fish, or molluscs. Furthermore, larger reductions in plant abundance were detected for fish relative to crustaceans. We found a positive relationship between herbivore species richness and change in plant abundance, with the strongest reductions in plant abundance reported for low herbivore species richness, suggesting that greater herbivore diversity may protect against large reductions in plant abundance. Finally, we found that herbivore–plant nativeness was a key factor affecting the magnitude of herbivore impacts on plant abundance across a wide range of species assemblages. Assemblages comprised of invasive herbivores and native plant assemblages were associated with greater reductions in plant abundance compared with invasive herbivores and invasive plants, native herbivores and invasive plants, native herbivores and mixed‐nativeness plants, and native herbivores and native plants. By contrast, assemblages comprised of native herbivores and invasive plants were associated with lower reductions in plant abundance compared with both mixed‐nativeness herbivores and native plants, and native herbivores and native plants. However, the effects of herbivore–plant nativeness on changes in plant abundance were reduced at high herbivore densities. Our mean reductions in aquatic plant abundance are greater than those reported in the literature for terrestrial plants, but lower than aquatic algae. Our findings highlight the need for a substantial shift in how biologists incorporate plant–herbivore interactions into theories of aquatic ecosystem structure and functioning. Currently, the failure to incorporate top‐down effects continues to hinder our capacity to understand and manage the ecological dynamics of habitats that contain aquatic plants.  相似文献

目的:观察唑来膦酸联合钙尔奇D治疗糖尿病性骨质疏松症的临床疗效。方法:将120例糖尿病性骨质疏松患者随机分为实验组和对照组,实验组60例予唑来膦酸联合钙尔奇D治疗,对照组60例予钙尔奇D治疗。连续治疗12个月后,比较两组患者的骨密度、疼痛评分及不良反应的发生情况。结果:治疗12个月后,实验组患者的股骨颈、股骨粗隆和腰椎正位的骨密度均较对照组明显升高,差异有统计学意义(P0.05);VAS疼痛评分较对照组显著降低(P0.05)。治疗过程中,两组均无严重的不良反应发生。结论:唑来膦酸联合钙尔奇D治疗糖尿病性骨质疏松临床效果优于钙尔奇D单药治疗。  相似文献

目的:探讨脓毒血症患者血清外周白细胞中的微小RNA(mi RNA)表达水平的变化及其在脓毒症患者中的表达意义以及免疫调控的关系。方法:采用流式细胞仪检测外周血CD4+CD25+Treg细胞表达,采用实时定量PCR(RT-PCR)方法检测110例脓毒症患者以及100例正常对照外周血白细胞中mi RNA以及Foxp3 m RNA表达量,酶联免疫吸附法测定TNF-α和IL-10浓度,序贯器官衰竭估计(SOFA)评分系统评价脓毒症患者的严重程度。对mi RNA与白细胞总数、TNF-α、IL-10和SOFA评分之间的相关性进行分析。结果:实验组mi RNA表达水平较对照组显著降低(P0.01),WBC、IL-10水平显著升高(P0.01)。实验组mi RNA表达水平以及SOFA评分、血清TNF-α和IL-10之间呈负相关关系(r值分别为-0.512、-0.623、-0.432,P0.05);与WBC无显著相关性(r=0.215,P0.05)。脓毒症患者外周血Treg表达率和Foxp3m RNA均显著高于对照组(P0.01);随病情严重而升高,轻、中、重度实验组间两两比较差异均有统计学意义(P0.01)。死亡均在重度脓毒症患者中,死亡组Treg、Foxp3 m RNA及IL-10均显著高于存活组(P0.01);mi RNA低于存活组(P0.01)。结论:脓毒症患者外周血的mi RNA表达量显著降低,表达水平在一定程度上可以反应机体的炎症反应情况,同时还可以判断疾病的严重度,且mi RNA参与对Treg细胞增殖的调节,在脓毒症免疫失衡机制中发挥一定的作用。  相似文献

EU expenditures for Rural Development, having increased from 2.9% of the total Common Agricultural Policy (CAP) budget in the 1990s to 12.3% in the beginning of the 2010s, reflect the growing strategic and societal values attached to this policy in addressing the new global challenges for rural areas in the enlarged EU. The analysis of the recent and ongoing reforms shows that Rural Development Programmes (RDP) made substantial progress towards sectoral integration and thematic broadening, and, at the same time, provided continuity of well-established and important measures. Yet, under growing budget constraints, better targeting of funds becomes even more crucial (European Court of Auditors, 2011). Comparing the design and allocation practices of Regional RDPs, however, reveals significant differences in targeting strategies, all too often based on the implementation history and experience of previous periods. Better availability, coordination and digital connectivity of expenditure data and other data sources across the EU encouraged the adoption of the Common Monitoring and Evaluation Framework (CMEF), aimed at more clearly depicting intervention needs and effects at the regional scale (NUTS2–NUTS3) (NUTS (Nomenclature des unités territoriales statistiques) is a EU geocode standard for referencing spatial subunits for statistical purposes). The EU project SPARD (Spatial Analysis of Rural Development Measures) took those data as a starting point to find out more about the causal relationships between RD measure implementation, and their determining factors and impacts with a specific look at their spatial dimension: in which rural development measures is success determined by neighbourhood conditions, and at what scale? SPARD developed and applied new methodological approaches, particularly spatial econometrics, to evaluate selected measures that contribute to improved competitiveness, environmental performance and rural viability. The results are presented in this special issue in four thematic foci:
  • •Analysing RDP performance by applying spatial econometric modelling (theory, procedures, key results) on RDP payments at the national and European scale.
  • •Effectiveness and efficiency of RDP participation towards impacts at the regional scale: an in-depth view of European case studies, specific measures, indicators and shortcomings of the CMEF.
  • •Learning about spatial and non-spatial determinants of participation in RDP.
  • •Experiences and requirements related to the CAP 2020 and the improvement of the CMEF.
Overall, the results emphasise the difficulty of evaluating of RDPs, even when using much more sophisticated instruments than those used in the current evaluation practices. While a lack of appropriate information remains an issue, the experience of SPARD also underscores the challenge of matching the quest for generalised approaches and the need to consider ad hoc local determinants, as well as the trade-off between the benefits of higher precision and the costs of implied information burden.  相似文献
