There is a diverse range of microbiological challenges facing the food, healthcare and clinical sectors. The increasing and pervasive resistance to broad‐spectrum antibiotics and health‐related concerns with many biocidal agents drives research for novel and complementary antimicrobial approaches. Biofilms display increased mechanical and antimicrobial stability and are the subject of extensive research. Cold plasmas (CP) have rapidly evolved as a technology for microbial decontamination, wound healing and cancer treatment, owing to the chemical and bio‐active radicals generated known collectively as reactive oxygen and nitrogen species. This review outlines the basics of CP technology and discusses the interactions with a range of microbiological targets. Advances in mechanistic insights are presented and applications to food and clinical issues are discussed. The possibility of tailoring CP to control specific microbiological challenges is apparent. This review focuses on microbiological issues in relation to food‐ and healthcare‐associated human infections, the role of CP in their elimination and the current status of plasma mechanisms of action.  相似文献

Organisms often face a higher risk of local extinction in fragmented than in continuous habitat. However, whether populations are affected by reduced size and connectivity of the habitat or by changes in habitat quality in fragmented landscapes remains poorly investigated. We studied the regional distribution and microhabitat selection of the lacertid lizard Psammodromus algirus in a fragmented landscape where the existence of deciduous and evergreen woodlands brought about variation in habitat quality. Lizards never occupied any fragment smaller than 0.5 ha. However, above that limit fragment size no longer predicted lizard occurrence, which was explained by woodland type instead, with lizards being more frequently found in deciduous than in evergreen woodlands. Lizards selected microhabitats that had structural features favouring thermoregulation, foraging and predator avoidance, and we identified better conditions for thermoregulation and food acquisition in deciduous than in evergreen woodlands. Our results support the idea that variation in habitat quality can sometimes override the effect of habitat fragmentation on animal populations. We consider the implications of our study for the conservation of Mediterranean lizards, discussing our results in a broader context framed by previous studies conducted in nearby areas.  相似文献

闽江河口红树林土壤微生物群落对互花米草入侵的响应   总被引:2,自引:0,他引:2  
采用磷脂脂肪酸标记法(PLFA)研究外来入侵植物互花米草对闽江河口湿地红树林土壤微生物群落结构的影响,并探讨其主要影响因素。结果表明:从3种不同植被群落土壤(红树林群落MC、红树林-互花米草混生群落MS、互花米草群落SC)共检测到22种PLFA生物标记,MS土壤微生物PLFA生物标记总量明显高于其他植被群落,3种植被群落土壤理化性质和酶活性的变化趋势为:MCMSSC,表明互花米草入侵后土壤微生物量增加,而理化性质和酶活性均有明显下降,红树林湿地土壤质量发生了明显退化。3种植被群落土壤中含量最高的PLFA生物标记是16:0,16:1w7c,9Me15:0w,18:1w12c。土壤中特征微生物相对生物量存在明显差异,细菌分布量最大,其次是真菌和放线菌,原生动物分布量最小。群落多样性指数呈相似规律,MS土壤微生物类群多样性指数均小于MC,表明互花米草入侵后土壤微生物群落多样性指数均有下降。通过主成分分析,基本能区分出3种不同植被群落微生物群落的特征。土壤理化性质、酶活性间存在相关性,有机碳、全氮、蔗糖酶、过氧化氢酶与革兰氏阴性菌、放线菌呈显著或极显著正相关。研究结果表明互花米草入侵在一定程度上具有影响红树林群落土壤营养代谢循环的潜力,特别是关于碳、氮、磷等的循环及酶活性,改变部分有利于自身生长的土壤环境相关的微生物类群含量,竞争有利环境,迅速扩张实现入侵。  相似文献

萘作为土壤动物化学抑制剂已在土壤动物生态功能的研究中广泛使用,但其非目标效应使其应用仍存在很大的不确定性。为了解在亚高山森林土壤应用萘抑制土壤动物群落的非目标效应,以川西亚高山森林土壤为研究对象,采用微缩实验研究了土壤微生物生物量、丰度和磷脂脂肪酸对萘胁迫的短期响应。结果表明,萘处理和对照的土壤微生物生物量碳(MBC)、真菌丰度以及细菌、真菌、革兰氏阳性菌(G~+)和革兰氏阴性菌(G~-)PLFAs含量在整个培养期间表现为降低的变化趋势,二者的土壤微生物生物量碳和G~+PLFAs含量以培养52d最低,细菌、真菌和G~-PLFAs含量以培养的45d最低。萘处理和对照的微生物生物量氮(MBN)含量表现出先升高后降低的动态,微生物生物量碳氮比(MBC/MBN)则表现为相反趋势。对照的真菌/细菌PLFAs比值呈现先升高后降低的动态,以培养的17d最高,但萘处理的真菌/细菌PLFAs比值无明显变化规律;萘处理的G~+/G~-PLFAs比值表现为降低的变化趋势,对照的G~+/G~-PLFAs比值表现为先降低后升高的趋势。萘处理仅显著影响了G~+/G~-PLFAs比值,但萘处理和采样时间的交互作用显著影响MBC/MBN、细菌丰度、真菌/细菌丰度比以及细菌、真菌的PLFAs含量、真菌/细菌PLFAs比值、G~+/G~-PLFAs比值。萘作为土壤动物抑制剂对川西亚高山森林土壤微生物群落的非目标效应具有时间变异性。  相似文献

Protected cultivation of ornamental flowers, as a commercial venture, becomes less profitable with excessive use of fertilizers. The present study examined the influence of microbial biofilm inoculants (AnabaenaAzotobacter, AnabaenaTrichoderma and TrichodermaAzotobacter) on the availability of soil nutrients and structure of rhizosphere microbial communities in three varieties of chrysanthemum (var. White Star, Thai Chen Queen and Zembla). Varietal-specific responses in growth, enzyme activities, flower yield of plants and availability of soil nutrients were recorded. Dehydrogenase activity was highest in var. White Star treated with the AnabaenaTrichoderma biofilm inoculants. The AnabaenaAzotobacter inoculant enhanced the availability of nitrogen, phosphorus and micronutrients in the soil, besides 40–50% increase in soil organic carbon, as compared to carrier alone or no inoculation. PCR-DGGE profiling of the cyanobacterial communities and qPCR quantification of 16S rRNA abundance of bacteria, archaea and cyanobacteria in the rhizosphere soils, revealed the stronger influences of these inoculants, especially in var. Zembla. Principal Component Analysis (PCA) helped to illustrate that the enhanced microbe-mediated availability of soil macro-and micronutrients, except iron content (Fe), was the most influential factor facilitating improved plant growth and yield parameters. The AnabaenaAzotobacter, and Anabaena–Trichoderma biofilm inoculants, proved superior in all three chrysanthemum varieties.  相似文献

A hybrid anaerobic solid-liquid bioreactor for food waste digestion   总被引:5,自引:0,他引:5  
A hybrid anaerobic solid-liquid (HASL) bioreactor is an enhanced two-phase anaerobic system, that consists of a solid waste reactor as the acidification reactor and a wastewater reactor, i.e. an upflow anaerobic sludge blanket (UASB) reactor as the methanogenic reactor. Food waste digestion in HASL bioreactors with pre-acidification and HASL operation stages was investigated in two separate runs. After 8 days of pre-acidification in Run A and 4 days in Run B, total volatile fatty acid (TVFA) and chemical oxygen demand (COD) concentrations in the leachates of both acidification reactors were similar. During HASL operation stage, TVFA and COD removal in the methanogenic phase were 77–100% and 75–95%, respectively. Some 99% of the total methane generated was from the methanogenic phase with a content of 68–70% methane. At the end of operation, about 59–60% of the added volatile solids (VS) were removed with a methane yield of 0.25 l g–1 VS.  相似文献

In order to calculate the dietary fumonisin intake of the German consumer, a large survey was carried out on a variety of potentially contaminated products in the period between December 1998 and July 2001. A total of 1960 food samples comprising all known relevant groups of products were analysed for fumonisins. Furthermore, 272 of these samples were also analysed for hydrolysed fumonisins (HFB). For routine analysis enzyme immunoassay was used, confirmatory and control analyses were performed using HPLC-FLD after precolumn derivatisation, or by LC-MS/MS. Daily intake of fumonisins was calculated by combining fumonisin contamination data obtained in this study with available food consumption data for Germany. In a “mean case” scenario, median fumonisin levels in foods and mean food intake values were used. To generate a “bad case” scenario, the 90th percentile of fumonisin levels in foods and mean food intake values were combined. The overall daily fumonisin intake by the German consumer was 1.1 μg in the “mean case” scenario, and 21 μg in the “bad case” scenario. It was concluded that in general there is no increased risk for the German consumer in aspects of exceeding the recommended tolerable daily intake of fumonisins (2 μg/kg body weight). However, certain products (and certain brands of products) were repeatedly found to contain elevated fumonisin levels, which in a “worst case” scenario (“high” food intake of maize-based products) could pose a potential risk for the consumer, in particular concerning foods for infants and young children. High fumonisin levels were found in infant foods in 1999, but contamination levels decreased strongly in the following years. HFBs (mostly HFB1) were frequently found in processed cereals such as corn flakes, but in relatively low concentrations. According to our findings, the new European Union maximum levels for fumonisins are suitable to eliminate peak contamination levels of fumonisins in foods, but would lead to a regular excess of the TDI for infants and young children if these maximum levels would indeed be exhausted. Nonthermal disinfection technologies are gaining increasing interest in the field of minimally processed food in order to improve the microbial safety or to extend the shelf life. Especially fresh‐cut produce or meat and fish products are vulnerable to microbial spoilage, but, due to their sensitivity, they require gentle preservation measures. The application of intense light pulses of a broad spectral range comprising ultraviolet, visible and near infrared irradiation is currently investigated as a potentially suitable technology to reduce microbial loads on different food surfaces or in beverages. Considerable research has been performed within the last two decades, in which the impact of various process parameters or microbial responses as well as the suitability of pulsed light (PL) for food applications has been examined. This review summarizes the outcome of the latest studies dealing with the treatment of various foods including the impact of PL on food properties as well as recent findings about the microbicidal action and relevant process parameters.  相似文献

Bacterial communities are known to play important roles during the developmental stages of insects, but current knowledge of bacteria associated with the midgut of Apis dorsata, the giant Asian honeybee, is limited. Using polymerase chain reaction‐denaturing gradient gel electrophoresis analysis (PCR‐DGGE) and 16S rRNA sequencing, the aim of this study was to determine the dynamics of bacterial community structure across four A. dorsata life stages in different geographical locations. The results reveal that bacterial diversity increased as the bee progressed through larval stage to newly emerged worker and old worker. However, in the pupal stage, no bands identified as bacteria could be observed. Overall, 2 bacterial phyla (Proteobacteria and Firmicutes) and 4 classes (Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, and Bacilli) were identified, but the frequency varied among the different stages and locations. The classes of Gammaproteobacteria and Bacilli dominated among larval, newly emerged worker and old worker developmental stages.  相似文献
