前哨淋巴结(sentinel lymph node,SLN)是肿瘤淋巴结转移的第一站,SLN活检肿瘤阳性的患者需要做系统性淋巴结清扫;SLN活检阴性的患者,不需要做系统性淋巴结清扫,可以缩短手术时间,降低手术费用,减少手术并发症;目前识别SLN的方法包括生物活性染料示踪法,放射性核素示踪法,联合示踪法,纳米炭(carbon nanoparticles,CNP)标记前哨淋巴结活检技术以及吲哚菁绿(Indocyanine Green,ICG)荧光标记法。SLN活检技术在乳腺癌、甲状腺癌、胃癌、恶性黑色素瘤、宫颈癌、子宫内膜癌等肿瘤中皆有不同程度的研究。本文通过复习文献,对前哨淋巴结检测方法予以归纳及其在常见肿瘤中的研究进展予以综述,旨在为恶性肿瘤临床治疗提供参考。  相似文献

Identification of specific nucleic acid sequences mediated by gold nanoparticles derivatized thiol-modified oligonucleotides (Au–nanoprobes) has been proven to be a useful tool in molecular diagnostics. Here, we demonstrate that, on optimization, detection may be simplified via the use of a single Au–nanoprobe to detect a single nucleotide polymorphism (SNP) in homo- or heterozygote condition. We validated this non-cross-linking approach through the analysis of 20 clinical samples using a single specific Au–nanoprobe for an SNP in the FTO (fat mass and obesity-associated) gene against direct DNA sequencing. Sensitivity, specificity, and limit of detection (LOD) were determined, and statistical differences were calculated by one-way analysis of variance (ANOVA) and a post hoc Tukey’s test to ascertain whether there were any differences between Au–nanoprobe genotyped groups. For the first time, we show that the use of a single Au–nanoprobe can detect SNP for each genetic status (wild type, heterozygous, or mutant) with high degrees of sensitivity (87.50%) and specificity (91.67%).  相似文献

We characterized fifteen microsatellite markers for the butterfly Maniola jurtina. For the six studied populations (96 samples) the total number of alleles per locus ranged from 3 to 55 and mean overall expected heterozygosity across all loci was 0.74. In spite of a high frequency of null alleles detected in part of the loci, a recurrent phenomenon in Lepidopteron, the estimation of pairwise FST seems rather insensitive to the presence of these null alleles as shown by the high correlation between FST calculated after correction for the presence of null alleles and non-corrected FST, indicating that the loci may be usable in population genetics, more specifically for the study of populations genetics structure.  相似文献

Autophagy is an important catabolic program to respond to a variety of cellular stresses by forming a double membrane vesicle, autophagosome. Autophagy plays key roles in various cellular functions. Accordingly, dysregulation of autophagy is closely associated with diseases such as diabetes, neurodegenerative diseases, cardiomyopathy, and cancer. In this sense, autophagy is emerging as an important therapeutic target for disease control. Among the autophagy machineries, PIK3C3/VPS34 complex functions as an autophagy-triggering kinase to recruit the subsequent autophagy protein machineries on the phagophore membrane. Accumulating evidence showing that inhibition of PIK3C3/VPS34 complex successfully inhibits autophagy makes the complex an attractive target for developing autophagy inhibitors. However, one concern about PIK3C3/VPS34 complex is that many different PIK3C3/VPS34 complexes have distinct cellular functions. In this study, we have developed an in vitro PIK3C3/VPS34 complex monitoring assay for autophagy inhibitor screening in a high-throughput assay format instead of targeting the catalytic activity of the PIK3C3/VPS34 complex, which shuts down all PIK3C3/VPS34 complexes. We performed in vitro reconstitution of an essential autophagy-promoting PIK3C3/VPS34 complex, Vps34–Beclin1–ATG14L complex, in a microwell plate (96-well format) and successfully monitored the complex formation in many different conditions. This PIK3C3/VPS34 complex protein assay would provide a reliable tool for the screening of autophagy-specific inhibitors.  相似文献

The aim of this work is to show a quick and simple procedure able to identify the geometrical parameters of the intervertebral disc that strongly affect the behavior of the FEM model. First, we allocated a selection criterion for the minimum number of geometrical parameters that describe, with a good degree of approximation, a healthy human vertebra. Next, we carried out a sensitivity analysis using the ‘Taguchi orthogonal array’ to arrive at a quick identification of the parameters that strongly affect the behavior of the Fem model.  相似文献

The cytotoxic effect of dehydroabietic acid (DHAA), a resin acid found in rosin, was studied on human polymorphonuclear leukocytes using leakage of 51Cr from prelabeled cells, supravital staining, and transmission electron microscopy. DHAA caused a strong dose-related release of 51Cr, a high uptake of trypan blue, and total cell necrosis, as seen in transmission electron microscopy. Albumin slightly reduced the toxic effects, whereas the addition of zinc in various forms strongly inhibited these toxic effects of DHAA in the concentration range of 10-500 micrograms/mL. In the presence of albumin, zinc oxide as a suspension inhibited the damage of the cell membranes more than a filtrate of zinc oxide, indicating a subsequent slow release of zinc from the zinc oxide.  相似文献

作者建立了在厌氧条件下两种微量而敏感的抑菌试验,可用于鉴定蛋白质或多肽类抑菌物质。(1)琼脂糖弥散法:可检测抗菌蛋白抑菌活性,(2)电泳凝胶弥散法:可直接确定存在于PAGE凝胶中抗菌蛋白条带。应用这两种方法,作者首次鉴定出血链球菌培养上清液中存在抑制牙周可疑致病菌的抗菌蛋白。  相似文献

甲真菌病是皮肤科的常见病,是由皮肤癣菌、酵母菌和非皮肤癣菌性霉菌侵犯甲板和(或)甲床所致的病变,其中由皮肤癣菌感染引起的甲真菌病称为甲癣。人甲板中角蛋白硬、厚且致密,从而形成一个完美屏障。针对甲板的特殊解剖结构,需要局部治疗药物具备穿透指甲屏障,体现高活性抗皮肤癣菌的特质。目前,甲癣的局部用药体外药效学研究没有统一的标准,缺乏指导依据。本文查阅近年来文献报道,综述甲癣局部治疗药物的体外药敏实验、药物与甲板内角蛋白亲和力实验以及药物在甲板内的渗透等,并阐述上述实验方法的模型选择、方式方法及方法间的比较,以推荐适合化合物筛选的方法。  相似文献

Stereoselective and efficient synthesis of hydroxymethyl-substituted rac-quercitols (1315) was achieved, starting from cis-furan (Kobayashi, 2008) with photooxygenation reaction, which is readily available by the reduction of cis-phtalic anhydride. α- and β-Glucosidase enzyme activity of the target molecules was evaluated and good inhibitor activity was seen. One- and two-dimensional NMR spectroscopy, IR spectroscopy and X-ray crystallography were utilized in the structure characterization of products.  相似文献
