The aim of this work was to develop a photoperiod-sensitive male sterile rice with stable sterility. We developed Changguang S, an indica rice strain, by using a short critical day length. Differences in the fertility responses of Changguang S strain pollen to temperature and photoperiod under natural and controlled conditions were studied. The results showed that Changguang S strain exhibited stable sterility under long-day and low-temperature conditions (22°C, 15 days). The stability of sterility was significantly higher than that of other such rice strains, Nongken 58S and 7001S. The critical photoperiod for inducing male sterility in Changguang S was 13 h or shorter, and its duration was significantly shorter than that required for rice strains Nongken 58S and 7001S. It is suggested that Changguang S is a typical photoperiod-sensitive male sterile rice strain with a shorter critical day length and a lower critical temperature. It is promising to apply this strain to two-line hybrid rice production.  相似文献

The sterile insect technique (SIT) has been used successfully for the control of fruit flies. The efficiency of this technique can be significantly reduced when sterile released insects are exposed to adverse conditions and predators, as a great number of sterile insects die before reaching sexual maturity and thus fail to mate with wild females. Treatments with juvenile hormone (JH) analogues such as methoprene (M) significantly reduce the time to reach sexual maturity by sterile Anastrepha ludens (Loew) (Diptera: Tephritidae) males. In this study, we compared the sexual performance of non‐treated sexually mature males with young males that had been sexually accelerated with M. Furthermore, we compared the ability of M‐fed males in inhibiting female remating compared with sexually mature males. Results showed that at 5 days M‐fed males had lower mating success than mature males; however, 6‐day‐old (0.1%) M‐fed males had the same amount of matings as mature 13‐day‐old males. Young 5‐ to 10‐day‐old M‐fed males also had similar number of matings as mature non‐treated 12‐ to 17‐day‐old males. There were no differences in copula duration between treatments. Moreover, there were no differences between the fertility, fecundity or refractory period of females mated with either young male fed M or normal sexually mature males. These results indicated that young males that were sexually accelerated with M have the same sexual performance as non‐treated sexually mature males. Implications of using M as a pre‐release treatment for A. ludens controlled through SIT are discussed.  相似文献

Recovery time after experience of a given minimum temperature below torpor threshold is related to the value of that minimum, the length of time spent at that minimum, and the temperature prevailing during the recovery period above torpor threshold. A model can predict recovery time for flies experiencing a given temperature fluctuation if the length of time spent at the minimum is expressed as a proportion of LE50 at that minimum.The model has applications in defining the optimal protocol for chilling insects for use in the Sterile Insect Release Method. The model was confirmed by experiments showing that it is likely that flies will recover from non-lethal frosts before ant predators become active.
Résumé Le temps de récupération après avoir subi une température minimal située au-dessous du seuil d'engourdissement dépend de la valeur de ce minimum, du temps passé à ce minimum, et de la température au-dessus du seuil d'engourdissement pendant la période de récupération. Un modèle mathématique permet d'estimer le temps de récupération après avoir subi une chute de température déterminée, en fonction du temps passé au minimum thermique exprimé comme une fraction du LE50 (temps nécessaire pour tuer 50% des mouches) à ce minimum.Ce modèle s'est trouvé étayé par des observations montrant qu'il est probable que les mouches se remettent des gelées sublétales avant la reprise d'activité des fourmis prédatrices. Ce modèle peut être utilisé pour définir les conditions optimales de refroidissement des insectes utilisés lors de la libération d'individus stériles.

Recent evidence suggests that the nutritional state of male Mediterranean fruit flies, Ceratitis capitata (Wied.) (medfly), is an important influence on various components of their reproductive biology, including mating success. The objective of the present study was to examine experimentally the effect of temporary starvation on the mating success of wild male C. capitata. Males were maintained on protein–sugar or sugar-only diets, and for each diet we compared the mating success of continuously fed males versus males starved for 18 or 24 h immediately before testing. In trials conducted on field-caged, host trees, males starved for 24 h obtained only about half as many matings as fed males for both diets. However, when the starvation period was 18 h, starved males reared on the protein–sugar diet mated significantly less frequently than fed males, whereas starved males reared on sugar mated as often as fed males. Measurements of male pheromone calling and female attraction revealed that reduced mating success likely reflected the decreased signaling activity of starved males.  相似文献

The golden‐headed lion tamarin (Leontopithecus chrysomelas) is an endangered species endemic to Brazil's Atlantic Forest, a shrinking biodiversity hotspot. As in other Neotropical primates, its semen characteristics and freezability are poorly studied. Hence, reproductive technologies for callitrichids would greatly benefit from reliable methods of semen analysis. In a bid to promote reproductive research in tamarins, we validated simple and inexpensive sperm function tests that can be used to monitor sperm‐egg binding, plasma membrane and acrosome integrity, mitochondrial activity, and DNA fragmentation. Ejaculates from adult males were individually diluted and divided into control and damage‐induced aliquots, and then samples comprising assorted amounts of damaged spermatozoa were examined by organelle‐specific tests. Our findings showed that sperm‐binding in chicken egg perivitelline membrane (EPM) positively correlated with the number of spermatozoa injured by snap‐freezing. Eosin‐nigrosin (EN) and propidium iodide readings were correlated with each other, and both provided robust measurements of plasma membrane integrity. A high correlation between expected and measured amounts of acrosome‐intact spermatozoa was found using Fast Green‐Rose Bengal (FG‐RB), Coomassie Blue (CB), and FITC‐PSA stains, and all three methods exhibited comparable results. Likewise, different percentages of UV‐irradiated spermatozoa were accurately assessed for DNA integrity by Toluidine Blue (TB) and sperm chromatin dispersion (SCD) tests. Comparisons between 3,3′‐diaminobenzidine (DAB) and JC‐1 stains also indicated the reliability of the former assay to ascertain gradual increases in spermatozoa with greater mitochondrial function. These data confirmed that different parts of the tamarin spermatozoa can be simply and consistently evaluated by EPM, EN, FG‐RB, CB, TB, and DAB protocols.  相似文献

Summary An assay is described whereby Eco RI restriction fragment length polymorphisms of mitochondrial and chloroplast DNAs can definitively identify cytoplasms of interest in Brassica crop development. Restrictable mitochondrial and chloroplast DNA is extracted from as little as 2–3 g and 0.5 g leaf tissue, respectively, and the donor plants are able to continue to develop in a normal manner. An unknown cytoplasm can be identified in three days, which is a considerable saving in time and labor compared to the several years required by traditional methods. The assay is very inexpensive and should be established as a routine procedure in laboratories involved in sexual or somatic Brassica hybrid production.  相似文献
