以中国东海鲸鲨软骨为原料,通过盐抽提、丙酮分级沉淀、离子交换层折、分子筛层析、高效液相色谱等步骤,获得鲨鱼软骨血管生成抑制因子-I(shark cartilgae angiogenesis inhibitory factor-I,SCAIF-I),对其分子量、抑制血管生成及抑制肿瘤生长活性进行了研究。结果显示SCAIF-I分子量18kD,在细胞和整体水平上显著抑制新血管生成,显著抑制小鼠肿瘤的生长  相似文献

单细胞凝胶电泳(single cell gel electrophoresis assay,SCGE)也叫彗星试验(comet assay),是一种快速、敏感、简便、廉价的检测单个哺乳动物细胞DNA断裂的技术,目前已用于检测氧化、紫外线和电离辐射引起的损伤,以及三氯乙烷、丙烯酰胺等化学物及老化、吸烟所致损害的研究.文章介绍SCGE的发展、检测分析方法、原理及其在DNA损伤与修复、生物监测、遗传毒理研究、肿瘤治疗方案优化和疗效研究方面的应用前景.  相似文献

从土壤中筛选到一株活性较高的肝素酶产生菌株Corynebacterium sp.。培养及产酶最佳条件表明,最适培养基组成(g/L):胰蛋白胨20,氯化钠1,磷酸氢二钾25,硫酸镁05,麦芽糖20,pH65。最适生长温度27℃,最佳产酶温度31℃。在500mL三角瓶中装40mL培养基,在30℃,200r/min摇床上培养24 h,每升发酵液可产酶1700u。  相似文献

Abstract: Polysaccharide lyases are the products of various microorganisms, bacteriophage and some eukaryotes. All such enzymes cleave a hexose-1,4-α- or β-uronic acid sequence by β-elimination. They are in some examples, the only known type of enzymes degrading their polyanionic substrates. Although only a small number of these enzymes have been exhaustively studied, the pectin lyases of bacterial origin have proved to be of interesting crystal structure containing a parallel β-helix domain. Alginate and heparin lyases may yield products with biotechnological potential.  相似文献

用100μg/ml藻红蛋白联合激光处理8113人口腔上皮癌细胞,MTT比色法测定癌细胞存活率。实验表明,先用藻红蛋白处理再经波长为488nm,25.6J/cm2氩离子激光辐照,细胞存活率为25%;而单用藻红蛋白,单用激光组分别为43%和107%。结果证明,藻红蛋白的光敏作用对体外培养的肿瘤细胞具有较强的杀伤作用。  相似文献

肝素类分子是一类结构异常复杂的高度硫酸化的糖胺聚糖,是临床上的一种主要的抗凝剂。除了抗凝血及其相关的抗栓活性以外,肝素还具有多种其他生物学功能,如抗平滑肌细胞及肾小球系膜细胞的增殖^[1]、抗炎症^[2]和阻止肿瘤生长及转移的作用^[3]等。因而肝素可用于防治球囊扩充血管成型术后的血管再狭窄、肾小球系膜细胞增生性肾炎、病理性炎症及肿瘤。但由于完整肝素的抗凝血活性,会引起出血及血小板减少综合症等负作用,限制了肝素在这些方面的临床应用。研究表明,肝素的抗凝血活性依赖于一种独特的肝素五糖序列,约占完整肝素链的1/3,除了五糖序列以外,还需要附近的至少13糖残基,因而不含五糖序列或含五糖序列小于18糖的肝素其抗凝活性大大降低^[4]。而六糖以上的肝素片段就具有抗平滑肌细胞增生、抗炎症及抗肿瘤的活性,并且这些活性与抗凝活性无关。因此,可利用特异性的肝素酶降解肝素长链,产生一系列不同结构及大小的肝素片段,并从中筛选出具有不同生物活性的片段。  相似文献

The aim of the present work is to evaluate the effect of a heparin derivative, low molecular weight heparin (LMWH) on the biochemical changes, tissue peroxidative damage and abnormal antioxidant levels in adriamycin (ADR) induced cardiac and hepatic toxicity. Male Wistar rats (140 +/- 10 g) were divided into four groups: untreated control (group I), ADR group (a single dose intravenous injection of 7.5 mg/kg ADR--group II), LMWH control (300 microg/day per rat s.c. for 1 week--group III) and ADR plus LMWH group (7.5 mg/kg ADR on day 1 of study period followed by LMWH treatment, 300 microg/day per rat commencing on day 8 and continued for a week. At the end of the 2-week experimental period, all animals were terminated. Cellular damage was assessed in terms of serum and tissue lactate dehydrogenase (LDH), aminotransferases and alkaline phosphatase (ALP) activities. Creatine phosphokinase (CPK) was assessed in the serum and heart tissue. The role of LMWH in altering the oxidative stress in ADR-induced toxicity was evaluated on the basis of its influence on cardiac and hepatic lipid peroxidation and antioxidant status (enzymatic and non-enzymatic)--superoxide dismutase (SOD), catalase and glutathione peroxidase (GPx), reduced glutathione (GSH), alpha-tocopherol (Vitamin E) and ascorbate (Vitamin C). LMWH administration to ADR-induced rats prevented the rise in serum and tissue levels of LDH, aminotransferases and ALP, while these parameters were significantly elevated in the ADR group in comparison with the control group. Cardiotoxicity indicated by rise in serum CPK in the ADR group was attenuated by LMWH treatment in group IV. LMWH decreased the cardiac and hepatic lipid peroxidation induced by ADR. Histologic examination revealed that the ADR-induced deleterious changes in the heart and liver tissues were offset by LMWH treatment. Restoration of cellular normalcy accredits LMWH with cytoprotective role in adriamycin-induced cardiac and hepatic toxicity.  相似文献

Does tumor growth follow a "universal law"?   总被引:4,自引:0,他引:4  
A general model for the ontogenetic growth of living organisms has been recently proposed. Here we investigate the extension of this model to the growth of solid malignant tumors. A variety of in vitro and in vivo data are analysed and compared with the prediction of a "universal" law, relating properly rescaled tumor masses and tumor growth times. The results support the notion that tumor growth follows such a universal law. Several important implications of this finding are discussed, including its relevance for tumor metastasis and recurrence, cell turnover rates, angiogenesis and invasion.  相似文献
