以围封保护和自由放牧油蒿草场为研究对象,通过野外调查与室内分析,研究了围封和放牧条件下沙地草场生物量和植被-土壤碳密度。结果表明:(1)自由放牧使油蒿群落中植物种类增加,但降低了植物群落盖度。自由放牧不仅导致油蒿草场地上、地下总生物量降低,也使得油蒿地上、地下生物量占群落地上、地下总生物量的比例减小。生长季自由放牧样地凋落物生物量显著大于围封保护样地(P0.05);(2)围封保护样地植被碳密度大于自由放牧样地,土壤碳密度却小于自由放牧样地,但两个样地间差异不显著(P0.05);(3)油蒿草场90%以上的碳储存于土壤中,围封保护样地和自由放牧样地油蒿草场土壤碳密度占植被-土壤系统碳密度的91%、93%;(4)围封保护油蒿草场碳密度为2.29 kg/m2,自由放牧油蒿草场碳密度为2.68 kg/m2,两个样地间差异不显著,自由放牧对油蒿草场碳密度影响不大。  相似文献

为了探究植物与固碳相关属性在不同功能群、器官和物种间的差异, 以及这些属性对不同土地利用方式的响应, 2012年8月对内蒙古大针茅草原的4个样地(长期无干扰样地、长期自由放牧样地、4年围封样地和4年围封割草样地)进行了群落调查, 并采集样地中的常见植物, 测定了与植物固碳相关的属性, 包括全碳含量、全氮含量、碳氮比、纤维素含量、木质素含量和酸性洗涤纤维含量等。以常见植物为对象, 在功能群水平研究了各土地利用方式下全碳含量、全氮含量和碳氮比的差异; 针对各样地的共有种——糙隐子草(Cleistogenes squarrosa)、大针茅(Stipa grandis)、冷蒿(Artemisia frigida)、羊草(Leymus chinensis)和猪毛菜(Salsola collina), 从物种和器官水平分析了不同土地利用方式下植物的固碳相关属性。结果表明: 大针茅草原植物不同功能群、物种和器官间的固碳相关属性存在极显著差异, 不同土地利用方式下大针茅草原不同功能型、物种和器官的固碳相关属性有显著差异。与其他利用方式相比, 4年围封割草对植物与固碳相关属性的影响最为明显, 功能群、器官和物种水平的植物氮含量均有下降。糙隐子草和猪毛菜的这些属性对长期自由放牧的响应敏感, 且方向相反。  相似文献

In two-component regulatory systems, covalent phosphorylation typically activates the response regulator signaling protein, and hydrolysis of the phosphoryl group reestablishes the inactive state. Despite highly conserved three-dimensional structures and active-site features, the rates of catalytic autodephosphorylation for different response regulators vary by a factor of almost 106. Previous studies identified two variable active-site residues, corresponding to Escherichia coli CheY residues 59 and 89, that modulate response regulator autodephosphorylation rates about 100-fold. Here, a set of five CheY mutants, which match other “model” response regulators (ArcA, CusR, DctD, FixJ, PhoB, or Spo0F) at variable active-site positions corresponding to CheY residues 14, 59, and 89, were characterized functionally and structurally in an attempt to identify mechanisms that modulate autodephosphorylation rate. As expected, the autodephosphorylation rates of the CheY mutants were reduced 6- to 40-fold relative to wild-type CheY, but all still autodephosphorylated 12- to 80-fold faster than their respective model response regulators. Comparison of X-ray crystal structures of the five CheY mutants (complexed with the phosphoryl group analogue BeF3) to wild-type CheY or corresponding model response regulator structures gave strong evidence for steric obstruction of the phosphoryl group from the attacking water molecule as one mechanism to enhance phosphoryl group stability. Structural data also suggested that impeding the change of a response regulator from the active to the inactive conformation might retard the autodephosphorylation reaction if the two processes are coupled, and that the residue at position ‘58’ may contribute to rate modulation. A given combination of amino acids at positions ‘14’, ‘59’, and ‘89’ adopted similar conformations regardless of protein context (CheY or model response regulator), suggesting that knowledge of residue identity may be sufficient to predict autodephosphorylation rate, and hence the kinetics of the signaling response, in the response regulator family of proteins.  相似文献

半干旱区封禁草地凋落物的积累与分解   总被引:19,自引:0,他引:19  
针对我国西部典型草原地带,退化草地封禁后凋落物的积累、分解与水分变化过程,进行了为期20a(1982~2002年)的定位试验研究.结果表明:本氏针茅、百里香、铁杆蒿和大针茅草地群落的凋落物积累与厚度的变化趋势,拟合曲线符合指数方程,无论在植物年生长的初期还是末期,其相关性极为显著.随着气温的升高和降雨量的增加,凋落物的分解速率加快,本氏针茅和大针茅群落凋落物的分解率达到峰值需150d,百里香群落需180d,铁杆蒿群落需210d;凋落物在积累与分解过程中,具有吸水和保水的重要功能,可截留大量天然降水,促进土壤水分的缓慢入渗,通常在植物生长的初期和末期,凋落物在自然状态下饱和含水量,本氏针茅群落为112.30%~124.02%;百里香群落为116.61%~134.09%;铁杆蒿群落为124.76%~144.32%.但草地适宜封禁年限为11~15a,有利于草地自然更新和凋落物的积累.  相似文献

The fast growth of smart electronics requires novel solutions to power them sustainably. Portable power sources capable of harvesting biomechanical energy are a promising modern approach to reduce battery dependency. Herein, a novel elastic impact‐based nonresonant hybridized generator (EINR‐HG) is reported to effectively harvest biomechanical energy from diverse human activities outdoors. Through the rational integration of a nonlinear electromagnetic generator with two contact‐mode triboelectric nanogenerators, the proposed EINR‐HG generates hybrid electrical output simultaneously under the same mechanical excitations. By introducing a flux‐concentrator with a nanowire‐nanofiber surface modification, significant improvement in the energy harvesting efficiency of the EINR‐HG is achieved. After optimizing using simulations and vibration tests, the as‐fabricated EINR‐HG delivers an outstanding normalized power density of 3.13 mW cm?3 g?2 across a matching resistance of 1.5 kΩ at 6 Hz under 1 g acceleration. Under human motion testing, the EINR‐HG generates an optimal output power of 131.4 mW with horizontal handshaking. With a customized power management circuit, the EINR‐HG serves as a universal power source that successfully drives commercial smart electronics, including smart bands and smartphones. This work shows the massive potential of biomechanical energy‐driven hybridized generators for powering personal electronics and portable healthcare monitoring devices.  相似文献

应用数量生态学中的梯度分析方法研究了云雾山天然草地群落类型、物种分布以及与环境因子的关系.研究结果表明,作为重要的人为管理措施,封育通过改善草地的水分、养分等环境条件,显著影响了云雾山草地的物种分布和群落类型.在封育草地,植被类型为本氏针茅群落,未封育草地为铁杆蒿群落.本氏针茅、百里香、大针茅、直茎点地梅、阿尔泰狗哇花等中旱生植物在封育草地中占优势,铁杆蒿、硬质早熟禾、星毛委菱菜等在退化草地中占优势.除封育措施以外,20~100cm 的土壤含水量对物种分布影响显著.同时,养分作为一个重要的环境因素对物种的分布也具有重要影响,特别是速效钾和NO-3-N.海拔、坡位、坡向等地形因素对植物分布的影响不明显.  相似文献

The present study was carried out to assess the possible role of mitogen-activated protein kinase (MAPK) in the meiosis-inducing action of the AMP-activated protein kinase (AMPK) activator, 5-aminoimidazole-4-carboxamide 1-beta-ribofuranoside (AICAR). Cumulus cell-enclosed oocytes (CEO) or denuded oocytes (DO) from immature, eCG-primed mice were cultured 4 hr in Eagle's minimum essential medium containing dbcAMP plus increasing concentrations of AICAR or okadaic acid (OA). OA is a phosphatase inhibitor known to stimulate both meiotic maturation and MAPK activation and served as a positive control. Both OA and AICAR were potent inducers of meiotic resumption in mouse oocytes and brought about the phosphorylation (and thus, activation) of MAPK, but by different kinetics: MAPK phosphorylation preceded GVB in OA-treated oocytes, while that resulting from AICAR treatment appeared only after GVB. The MEK inhibitors, PD98059 and U0126, blocked the meiotic resumption induced by AICAR but not that induced by OA. Although the MEK inhibitors suppressed MAPK phosphorylation in both OA- and AICAR-treated oocytes, meiotic resumption was not causally linked to MAPK phosphorylation in either group. Furthermore, AICAR-induced meiotic resumption in Mos-null oocytes (which are unable to stimulate MAPK) was also abrogated by PD98059 treatment. A non-specific effect of the MEK inhibitors on AICAR accessibility to the oocyte was discounted by showing that they failed to suppress either nucleoside uptake or AICAR-stimulated phosphorylation of acetyl CoA carboxylase (ACC), a substrate of AMPK. The suppression of AICAR-induced maturation by MEK inhibitors must, therefore, be occurring by actions unrelated to MEK stimulation of MAPK; consequently, it would be prudent to consider this possible non-specific action of the inhibitors when they are used to block MAPK activation in mouse oocytes.  相似文献
