As a solution to the problems of mass transfer limitation in submerged cultures and scale up of solid-state/liquid-surface cultures, an alternating liquid phase–air phase bioreactor was developed. It consisted of a bioreactor equipped with a siphon system and a reservoir. Aspergillus awamori was immobilized in loofa sponge inside the bioreactor and culture broth was pumped from the reservoir into the bioreactor. Each time the culture broth level reached a critical level, the broth automatically siphoned back into the reservoir. Thus the immobilized cells were alternatingly submerged and exposed to air. The duration of each phase was controlled by the pumping rate and with an on-off timer. During amylase production from soluble starch and raw cassava starch, the optima ratios of the liquid to air phases were 12 h : 12 h and 3 h : 6 h respectively. Saccharomyces cerevisiae IR2 was immobilized in the reservoir and the system was used for simultaneous amylase production, hydrolysis and ethanol production from raw cassava starch. The process was very stable for more than 7 batches with high ethanol yield of 0.46 g-ethanol/g-starch and productivity of 1.73 g-ethanol/L/h. These values are high, the system can be scaled up, and thus it has many potential applications.  相似文献

Populations of the phytoseiid predator Amblyseius(=Neoseiulus) idaeus (Denmark & Muma) from northeastern Brazil, have been successfully introduced into Benin, West Africa, as part of a classical biological control campaign to control the exotic cassava green mite Mononychellus tanajoa (Bondar). Monthly follow‐up surveys revealed the presence of A. idaeus in most release sites. Some populations have persisted for at least 18 months, including two cycles of potentially limiting wet and dry season conditions. In some sites A. idaeus has been the numerically dominant phytoseiid predator on cassava Manihot esculenta, where it is associated with the tetranychids M. tanajoa and Oligonychus gossypii Zacher. During periods of low M. tanajoa densities A. idaeus disappeared from cassava, but were found on weeds with O. gossypii until prey densities on cassava increased.  相似文献

The cyanogenic potentials and residual cyanide contents of local cassava parenchyma and their locally processed food products in southeastern Nigeria were studied. Seven species of cassava locally grown and four main food products from them were analyzed colorimetrically for their cyanide contents. Results of the analyses indicated that five of the species contain cyanide potentials between 50 and 100 mg HCN/kg fresh weight while only one contains cyanogens level greater than 100 mg HCN/kg fresh weight. Of the cassava products analyzed, two contained cyanide above the level recommended by the WHO/FAO (10 mg HCN/kg). The result raises concern as these cassava products constitute about 80–90% of the diet of the local people and the facts known about cyanide poisoning from intake of high cyanide containing food.  相似文献

The economic importance of Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) is mainly due to its wide host range, variable kinds of damage, and great intraspecific variation. The delineation of two African biotypes of this pest has been carried by molecular, ecological, and host‐affiliation approaches, with largely consistent results. However, an understanding of its intricate host–pest interaction is necessary as a basis of its sustainable integrated control. This study investigated the host preference and suitability of cassava and okra biotypes of B. tabaci, based on multiple‐choice landing and oviposition preference assays and stage‐specific survival on eight common whitefly hosts. The cassava biotype significantly preferred cassava, Manihot esculenta, for landing and oviposition, but did not oviposit on okra, Abelmoschus esculentus. The okra biotype preferred okra, oviposited on eggplant, Solanum melongena, tomato, Lycopersicon esculentum, garden egg, Solanum integrifolium, and cowpea, Vigna unguiculata, but did not oviposit on cassava. The okra biotype developed on all hosts except cassava, but only survived marginally on cabbage, Brassica oleracea, and pepper, Capsicum annum var. grosum, while the cassava biotype did not develop on okra, cabbage, or pepper. Thus the observed host acceptance of the two biotypes is wider than earlier reported by host transfer experiments and molecular genetic surveys. Mortality was highest in the first instar nymphal stage, during which total mortality occurred on non‐hosts. Development time was slightly longer on marginal hosts than on the preferred hosts. Cowpea, garden egg, and tomato are additional common hosts of the two biotypes, whose role as reservoir hosts and biotype interbreeding grounds should be investigated further.  相似文献

Summary The aim of this research was to develop methods to use low-cost carbon compounds for rhizobial inoculant production. Five raw starch materials; steamed cassava, sticky rice, fresh corn, dry corn and sorghum were tested for sugar production by an amylase-producing fungus. Streamed cassava produced the highest amount of reducing sugar after fermentation. Bradyrhizobium japonicum USDA110, Azorhizobium caulinodans IRBG23, Rhizobium phaseoli TAL1383, Sinorhizobium fredii HH103, and Mesorhizobium ciceri USDA2429 were tested on minimal medium supplemented with reducing sugar obtained from cassava fermentation. All strains, except B. japonicum USDA110, could grow in medium containing cassava sugar derived from 100 g steamed cassava per litre, and the growth rates for these strains were similar to those in medium containing 0.5 (w/v) mannitol. The sugar derived from steamed cassava was further used for production of glycerol using yeast. After 1 day of yeast fermentation, the culture containing glycerol and heat-killed yeast cells, was used to formulate media for culturing bradyrhizobia. A formulation medium, FM4, with a glycerol concentration of 0.6 g/l and yeast cells (OD600 = 0.1) supported growth of B. japonicum USDA110 up to 3.61 × 109 c.f.u./ml in 7 days. These results demonstrate that steamed cassava could be used to provide cheap and effective carbon sources for rhizobial inoculant production.  相似文献

Three field trials were conducted at Namulonge Agricultural and Animal Production Research Institute to investigate the population dynamics of Bemisia tabaci Gennadius and its associated aphelinid parasitoids in Uganda. Results showed that more whitefly occurred on the cassava mosaic disease-resistant variety, (Nase 4) compared to the susceptible variety, (Ebwanatereka). Two species of aphelinid parasitoids, Eretmocerus mundus Mercet and Encarsia sophia Girault and Dodd, were identified during the study. Overall percent parasitism did not differ significantly (P>0.05) between varieties in all the trials, but significant differences occurred at 13, 15 and 21 weeks after planting during late season (2000), 18 weeks after planting during early season (2001), and 8 weeks after planting during late season (2001). The trends in the build up in numbers of both parasitoids species and apparent parasitism were similar, differed significantly on certain dates. It was, however, noted that percent parasitism decreased with nymph number. The significance of this phenomenon on the potential use of these aphelinid parasitoids as biocontrol agents of the cassava whitefly is discussed.  相似文献

The consequence of harvesting young leaves of cassava as vegetable on the vulnerability of the crop to cassava mosaic disease (CMD) and on storage root yield was investigated using 30 cassava genotypes planted in IITA fields located in the humid forest (Port Harcourt?:?Onne), forest-savannah transition (Ibadan), southern guinea savannah (Mokwa) and northern guinea savannah (Zaria) agroecologies in Nigeria. Tender apical leaves and shoots of the cassava genotypes were removed from forty plants per cassava genotype with the same number of plants considered as control. Whitefly infestation, disease incidence (DI) and symptom severity (ISS) of the disease were assessed at monthly interval for six months and also at the ninth month after planting (MAP). Yield reduction due to this treatment was calculated as percentage harvest index (HI). Whitefly population fluctuated throughout the period of observation at all locations with higher population obtained generally for treated plants compared to control plants. Sprouting leaves of some treated genotypes were observed with severe mosaic symptoms, while corresponding control showed no mosaic symptoms. Contrarily, no remarkable difference was observed in Zaria between the mean ISS of treated and control cassava genotypes. There was a highly significant difference (P?<?0.01) in DI and ISS among cassava genotypes across all locations. Also, there was a highly significant interaction (P?<?0.01) in symptom severity between location (loc) and genotype, genotype and treatment (trt), loc and trt. Interaction between loc, genotypes and trt with regard to DI was highly significant at 2, 3 and 4 MAP, while with ISS, the interaction was highly significant all through the counting period. There was a positive relationship between DI and ISS on plants of genotypes 96/1039 and ISU. The percentage HI (27.4) of treated plants of genotype 95/0166 in Ibadan was remarkably lower than the value obtained for corresponding control (41.9) plants. Also, sharp distinction in% HI of treated (39.5) and control (43.8) ISU was observed in Onne with their respective ISS values as 3.7 and 3.2. Therefore, harvesting tender apical leaves and shoots of cassava as vegetables should be discouraged as it increases the severity of CMD infection in the regenerating shoots of cassava with attendant storage root yield reduction.  相似文献

Bacterial blight disease caused by Xanthomonas axonopodis pv. manihotis (Berthet-Bondar) Dye was assessed in 11 artificially inoculated cassava genotypes in a screenhouse. Disease progress was estimated at intervals of 3 days by measuring the length of necrotic lesions on stems and leaves, as well as estimating the average disease score and area under disease progress curve (AUDPC). Based on the average disease scores, cassava genotypes 30572, TME 1, TME 7 and TME 9 were classified as resistant to bacterial blight, genotypes 4(2)1425, TME 2, TME 4 and TME 12 were tolerant while cassava genotypes 30001, TME 3, and TME 28 were susceptible. Direct correlations, statistically significant at p < 0.05, were obtained between stem necrosis, leaf necrosis, average disease scores and AUDPC in the 11 cassava genotypes. Screenhouse experiments afford rapid assessment of resistance status of cassava genotypes to bacterial blight in Nigeria.  相似文献

Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae), one of the most economically important agricultural pests worldwide, is the vector of cassava mosaic geminiviruses that cause cassava mosaic disease (CMD). In East and Central Africa, a severe CMD pandemic that spread from Uganda in the late 1980s still continues to devastate cassava crops. To assess the association of distinct B. tabaci genetic groups with the CMD pandemic, mitochondrial cytochrome oxidase I gene sequences were analysed from whiteflies collected during surveys conducted from 2010 to 2013 in Tanzania. Four genetic groups – Sub‐Saharan Africa 1 (SSA1), Mediterranean, Indian Ocean and East Africa 1, and a group of unknown whitefly species were identified. SSA1 comprised four subgroups: SSA1‐SG1, SSA1‐SG2, SSA1‐SG1/2 and SSA1‐SG3. SSA1‐SG1 was confined to the pandemic‐affected north‐western parts of Tanzania whilst SSA1‐SG2 and SSA1‐SG3 were found in the central and eastern parts not yet affected by the pandemic. The CMD pandemic front was estimated to lie in Geita Region, north‐western Tanzania, and to be spreading south‐east at a rate of ca 26 km/year. The pandemic‐associated B. tabaci SSA1‐SG1 predominated up to 180 km ahead of the CMD front indicating that changes in whitefly population characteristics precede changes in disease characteristics.  相似文献
