Summary. Stato- or otoliths are calcified structures in the organ of balance and equilibrium of vertebrates, the inner ear, where they enhance its sensitivity to gravity. The compact otoliths of fish are composed of the calcium carbonate polymorph aragonite and a small fraction of organic molecules. The latter form a protein skeleton which determines the morphology of an otolith as well as its crystal lattice structure. This short review addresses findings according to which the brain obviously plays a prominent role in regulating the mineralisation of fish otoliths and depends on the gravity vector. Overall, otolith mineralisation has thus been identified to be a unique, neuronally guided biomineralisation process. The following is a hypothetical model for regulation of calcification by efferent vestibular neurons: (1) release of calcium at tight junctions in the macular epithelia, (2) macular carbonic anhydrase activity (which in turn is responsible for carbonate deposition), (3) chemical composition of matrix proteins. The rationale and evidence that support this model are discussed. The function of sensory hair cells of the cochlea and vestibular organs depends on an influx of K+ through apical mechanosensitive ion channels and its subsequent removal over their basolateral membrane. The KCNQ4 (Kv7.4) K+ channel, which is mutated in DFNA2 human hearing loss, is expressed in the basal membrane of cochlear outer hair cells where it may mediate K+ efflux. Like the related K+ channel KCNQ5 (Kv7.5), KCNQ4 is also found at calyx terminals ensheathing type I vestibular hair cells where it may be localized pre- or postsynaptically. Making use of Kcnq4−/− mice lacking KCNQ4, as well as Kcnq4dn/dn and Kcnq5dn/dn mice expressing dominant negative channel mutants, we now show unambiguously that in adult mice both channels reside in postsynaptic calyx-forming neurons, but cannot be detected in the innervated hair cells. Accordingly, whole cell currents of vestibular hair cells did not differ between genotypes. Neither Kcnq4−/−, Kcnq5dn/dn nor Kcnq4−/−/Kcnq5dn/dn double mutant mice displayed circling behavior found with severe vestibular impairment. However, a milder form of vestibular dysfunction was apparent from altered vestibulo-ocular reflexes in Kcnq4−/−/Kcnq5dn/dn and Kcnq4−/− mice. The larger impact of KCNQ4 may result from its preferential expression in central zones of maculae and cristae, which are innervated by phasic neurons that are more sensitive than the tonic neurons present predominantly in the surrounding peripheral zones where KCNQ5 is found. The impact of postsynaptic KCNQ4 on vestibular function may be related to K+ removal and modulation of synaptic transmission.  相似文献

The presence of calcium within the utricle of larval cichlid fish Oreochromis mossambicus was analysed by means of energy-filtering transmission electron microscopy. Electron-spectroscopic imaging and electron energy loss spectra revealed discrete calcium precipitations that were more numerous in the proximal endolymph than in the distal endolymph, clearly indicating a decreasing proximo-distal gradient. This decreasing proximo-distal gradient was also present within the proximal endolymph between the sensory epithelium and the otolith. Further calcium particles covered the peripheral proteinaceous layer of the otolith. They were especially pronounced at the proximal surface of the otolith indicating that otolithic calcium incorporation takes place here. Other calcium precipitates accumulated at the macular junctions clearly supporting an earlier assumption according to which the endolymph is supplied with calcium via a paracellular pathway. The present results clearly show that the apical region of the macular epithelium is involved in the release of calcium and that the calcium supply of the otoliths takes place via the proximal endolymph.This work was financially supported by the German Aerospace Center (DLR) e.V. Differentiation in the size and number of seeds among populations or particular individuals of a given species may depend on genetic features and environmental conditions. The objective of our study was to answer two questions: whether any differences exist in the size and shape of utricles among Carex spicata populations growing in several plant communities and whether the hypothesized differences remain constant in ‘common garden’ conditions (i.e. if the sedges grown in different plant communities are evolving distinctly separate ecotypes). We studied utricle morphological traits (projected area, width, length, W/L ratio and projected perimeter) and number of utricles per spike collected from plants grown in five different plant communities (natural sites) and from plants transferred to common garden conditions. C. spicata utricles showed significant variability of morphological traits that depended on the plant community of origin. Among five plant communities, the largest utricles were found in Agropyron repens–Urtica dioica and Convolvulo arvensis–Agropyretum repentis communities, while the smallest ones were in plants from the Lolio-Plantaginetum community. The similarity of the analyzed populations regarding sedge utricle traits corresponded to the similarity of plant communities where the populations had grown. Moreover, the differences in utricle traits collected from natural conditions remained stable in common garden conditions. This indicated that differences in traits among utricles had a physiological dependence on different biotope conditions or different interspecific interactions prevailing in the particular plant communities. The relationship between the similarities in the diaspores of the populations studied and the similarities in the ecological conditions of the communities may also indicate that the variability of utricles is important for evolution and adaptation.The results support the hypothesis that C. spicata populations in different communities are producing separate ecotypes, i.e. specific species forms with genetically fixed traits adapted to narrowly determined habitat conditions.  相似文献

The cellular distribution of parvalbumin immunoreactivity in the vestibular peripheral system of mouse, rat, and guinea pig was investigated by light and electron microscopy. Parvalbumin was found in all neurons of the vestibular ganglia of these species but in the sensory epithelia immunoreactivity was restricted to type I hair cells localized exclusively in the central areas. The very intense staining pattern was similar in the cristae ampullares and utricles of all three species but a faint immunoreaction was also detectable in sensory cells of peripheral areas of rat cristae. The parvalbumin-immunoreactive type I sensory cells are connected by nerve fibres of the calyx unit type which are known selectively to contain calretinin.  相似文献

摘要 目的:探讨缺氧对豚鼠前庭耳石器椭圆囊毛细胞内兴奋性神经递质谷氨酸的合成及释放的影响。方法:分离4周龄豚鼠双侧椭圆囊进行体外培养,分别置于常氧(21% O2)和低氧(1% O2)环境下处理30 min,利用Western blotting、酶活性检测等方法检测前庭外周感受器椭圆囊斑毛细胞中谷氨酰胺酶的活性和表达水平,评价谷氨酸合成变化情况;利用ELISA、免疫荧光等方法检测椭圆囊组织中谷氨酸含量和毛细胞内谷氨酸分布情况。结果:在给予缺氧处理后,豚鼠椭圆囊毛细胞中谷氨酸合成关键酶谷氨酰胺酶的表达、酶活性均未发生显著性变化(P>0.05);ELISA结果提示,相较于对照组,低氧组椭圆囊组织中谷氨酸水平降低5.6%(对照组vs.缺氧组:15.86±2.19 vs.13.02±1.21,P<0.05);免疫荧光结果提示,相较于对照组,低氧组椭圆囊毛细胞内的谷氨酸更加广泛的分布于毛细胞基底部,而在纤毛一侧分布较少。结论:在体外培养的椭圆囊组织中,低氧处理并未影响毛细胞谷氨酸的合成过程,但会显著增加毛细胞内兴奋性神经递质谷氨酸的释放。  相似文献

The resting potassium current (I KI ) in gerbil dissociated type I vestibular hair cells has been characterized under various ionic conditions in whole cell voltage-clamp. When all K+ in the patch electrode solution was replaced with Na+, (Na+) in or Cs+, (Cs+) in , large inward currents were evoked in response to voltage steps between −90 and −50 mV. Activation of these currents could be described by a Hodgkin-Huxley-type kinetic scheme, the order of best fit increasing with depolarization. Above ∼−40 mV currents became outward and inactivated with a monoexponential time course. Membrane resistance was inversely correlated with external K+ concentration. With (Na+) in , currents were eliminated when K+ was removed from the external solution or following extracellular perfusion of 4-aminopyridine, indicating that currents flowed through I KI channels. Also, reduction of K+ entry through manipulation of membrane potential reduced the magnitude of the outward current. Under symmetrical Cs+, 0 K+ conditions I KI is highly permeable to Cs+. However, inward currents were reduced when small amounts of external K+ were added. Higher concentrations of K+ resulted in larger currents indicating an anomalous mole fraction effect in mixtures of external Cs+ and K+. Received: 23 June 1999/Revised: 27 September 1999  相似文献

The mechanosensory hair cells of the inner ear have emerged as one of the primary models for studying the development of planar polarity in vertebrates. Planar polarity is the polarized organization of cells or cellular structures in the plane of an epithelium. For hair cells, planar polarity is manifest at the subcellular level in the polarized organization of the stereociliary bundle and at the cellular level in the coordinated orientation of stereociliary bundles between adjacent cells. This latter organization is commonly called Planar Cell Polarity and has been described in the greatest detail for auditory hair cells of the cochlea. A third level of planar polarity, referred to as tissue polarity, occurs in the utricular and saccular maculae; two inner ear sensory organs that use hair cells to detect linear acceleration and gravity. In the utricle and saccule hair cells are divided between two groups that have opposite stereociliary bundle polarities and, as a result, are able to detect movements in opposite directions. Thus vestibular hair cells are a unique model system for studying planar polarity because polarization develops at three different anatomical scales in the same sensory organ. Moreover the system has the potential to be used to dissect functional interactions between molecules regulating planar polarity at each of the three levels. Here the significance of planar polarity on vestibular system function will be discussed, and the molecular mechanisms associated with development of planar polarity at each anatomical level will be reviewed. Additional aspects of planar polarity that are unique to the vestibular maculae will also be introduced.  相似文献
