The enigmatic arrow worms (Chaetognatha) are marine carnivores and among the most abundant planktonic organisms. Their phylogenetic
position has been heavily debated for a long time. Most recent molecular studies still provide a diverging picture and suggest
arrow worms to be some kind of basal protostomes. In an effort to understand the organization of the nervous system in this
clade for a broad comparison with other Metazoa we analysed the ultrastructure of the ventral nerve centre in Spadella cephaloptera by transmission electron microscopy. We were able to identify six different types of neurons in the bilateral somata clusters
by means of the cytoplasmic composition (regarding the structure of the neurite and soma including the shape and eu-/heterochromatin
ratio within the nucleus) as well as the size and position of these neurons. Furthermore, our study provides new insights
into the neuropil composition of the ventral nerve centre and several other fine structural features. Our second goal was
to examine if individually identifiable neurons are present in the ventral nerve centres of four chaetognath species, Sagitta setosa, Sagitta enflata, Pterosagitta draco, and Spadella cephaloptera. For that purpose, we processed whole mount specimens of these species for immunolocalization of RFamide-related neuropeptides
and analysed them with confocal laser-scanning microscopy. Our experiments provide evidence for the interspecific homology
of individual neurons in the ventral nerve centres of these four chaetognath species suggesting that the potential to generate
serially arranged neurons with individual identities is part of their ground pattern. 相似文献

Summary Synaptogenesis has been studied in the electric organ of embryonic Torpedo marmorata by use of two antisera directed against components of synaptic vesicles (anti-SV) and presynaptic plasma membranes (ap-anti-TSM), respectively. The anti-SV serum was previously shown to recognize a proteoglycan specific for synaptic vesicles. The ap-anti-TSM serum was raised to plasma membranes of synaptosomes derived from the electromotor nerve terminals and affinity-purified on electric-organ gangliosides. The vesicular antigen was first detectable at the 81-mm stage of development, which is 1–2 weeks earlier than the formation of morphologically mature presynaptic terminals, but is coincident with a rise in choline acetyltransferase levels and the ability of the electric organ to generate discharges. The gangliosidic antigen recognized by the ap-anti-TSM was first detectable on the ventral electrocyte surface at the 93-mm stage of development. This indicates that specific carbohydrate epitopes, not present on the growth cones, are expressed during maturation of the nerve terminal. The nerve terminal components recognized by these sera arose pari passu with neurite coverage of the ventral surface of the electrocyte, reaching a maximum in the adult. In contrast, postsynaptic aggregates of acetylcholine receptor, rendered visible with rhodamine-labeled -bungarotoxin, arose previous to the presynaptic antigens, reaching a maximum surface density at 110 mm and then declining in the adult. 相似文献

Summary During embryogenesis and planula development of the colonial hydroidHydractinia echinata cell proliferation decreases in a distinct spatio-temporal pattern. Arrest in S-phase activity appears first in cells localized at the posterior and then subsequently at the anterior pole of the elongating embryo. These areas do not resume S-phase activity, even during the metamorphosis of the planula larva into the primary polyp. Tissue containing the quiescent cells gives rise to the terminal structures of the polyp. The posterior area of the larva becomes the hypostome and tentacles, while the anterior part of the larva develops into the basal plate and stolon tips. In mature planulae only a very few cells continue to proliferate. These cells are found in the middle part of the larva. Labelling experiments indicate that the prospective material of the postmetamorphic tentacles and stolon tips originates from cells which have exited from the cell cycle in embryogenesis or early in planula development. Precursor cells of the nematocytes which appear in the tentacles of the polyp following metamorphosis appear to have ceased cycling before the 38th hour of embryonic development. The vast majority of the cells that constitute the stolon tips of the primary polyp leave the cell cycle not later than 58 h after the beginning of development. We also report the identification of a cell type which differentiates in the polyp without passing through a post-metamorphic S-phase. Recent research findings suggest neuro-modulation of tumors. Finding new modifiable prognostic factors paves the way for additional treatments, which is crucial in advanced cancer, particularly pancreatic cancer. This study examined the relationship between vagal nerve activity, indexed by heart rate variability (HRV), and overall survival (OS) in patients (N = 272) with advanced pancreatic cancer. A “historical prospective” design was employed, where vagal activity and other confounders were retroactively obtained from medical charts at diagnosis, and subsequent OS was examined. HRV was obtained from 10 sec ECGs near diagnosis. Levels of C-reactive protein (CRP) were measured as an inflammatory marker. OS and survival date were obtained from medical charts and the Belgian national registry. Patients with high HRV (>20 msec) survived on average more than double the days (133.5) than those with low HRV (64.0). In a multivariate cox regression, higher initial HRV was significantly correlated with lower risk of death, independent of confounders including age and cancer treatments. This relationship was statistically mediated (accounted for) by CRP levels. Importantly, in patients who lived up to one month from diagnosis only, HRV was unrelated to CRP, while in patients surviving longer, HRV was significantly inversely related to CRP (r = −0.20, p < 0.05). These results are in line with possible vagal nerve protection in a fatal cancer, and propose that the mechanism may involve neuroimmuno-modulation. Future studies must test whether vagal nerve activation may help patients with advanced cancers. 相似文献

Primary afferent sprouting in the spinal cord was evaluated by comparing the central projection of horseradish peroxidase (HRP)-labeled sciatic nerve afferent axons in nonlesioned control rats, and in rats subjected to acute or chronic partial spinal hemisections as adults. The lesions were performed at various levels from T10 to L3, and removed supraspinal and varying amounts of descending propriospinal afferents to lumbar segments receiving the maximal sciatic projection. The hemisections typically involved all but the dorsal column, although in some cases a portion of the dorsal column, including the corticospinal tract, was also transected.The distribution pattern and density of spinal HRP reaction product was not significantly different in experimental and control preparations in any segment below the lesion, regardless of the quantity of denervation, or the density of the normal sciatic projection in a given terminal region. These results, together with our previous finding concerning an absence of primary afferent sprouting following long-term dorsal root ganglionectomies, suggest that current concepts concerning collateral sprouting as a factor in functional plasticity in the mature mammalian spinal cord warrant re-evaluation. 相似文献

Activity of neck-muscle motoneurones during eye cleaning behaviour in the cricket Gryllus campestris
The activity of neck-muscle motoneurones which control head movements during eye cleaning behaviour was recorded from motor nerves with chronically implanted electrodes in unrestrained crickets. We show that motoneurones of the dorso-ventral muscles displayed strong activity differences between both sides of the neck, with higher discharge frequencies either ipsi- or contralateral to the direction of the head movement. Motoneurones innervating dorsal-longitudinal muscles were equally active on both sides. A single excitatory motoneurone of one dorso-ventral muscle showed a discharge pattern unequivocally related to eye cleaning. Lesions of connectives revealed that this motoneurone is monitored by interneuronal pathways from the suboesophageal ganglion although the primary sensory axons eliciting eye cleaning, project into the prothoracic ganglion. 相似文献

Optical coherence tomography angiography (OCTA) is a widely applied tool to image microvascular networks with high spatial resolution and sensitivity. Due to limited imaging speed, the artifacts caused by tissue motion can severely compromise visualization of the microvascular networks and quantification of OCTA images. In this article, we propose a deep-learning-based framework to effectively correct motion artifacts and retrieve microvascular architectures. This method comprised two deep neural networks in which the first subnet was applied to distinguish motion corrupted B-scan images from a volumetric dataset. Based on the classification results, the artifacts could be removed from the en face maximum-intensity-projection (MIP) OCTA image. To restore the disturbed vasculature induced by artifact removal, the second subnet, an inpainting neural network, was utilized to reconnect the broken vascular networks. We applied the method to postprocess OCTA images of the microvascular networks in mouse cortex in vivo. Both image comparison and quantitative analysis show that the proposed method can significantly improve OCTA image by efficiently recovering microvasculature from the overwhelming motion artifacts. 相似文献

Age-related changes in amounts of myelin proteins from rat sciatic nerve or spinal root were analyzed by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE). In the aged peripheral nerve myelin, the relative amounts of band 105K and proteins X and Y increased, whereas those of proteins P0 and P1 and band 190K decreased. Band 105K purified by preparative SDS-PAGE exhibited three bands of 105K, 28K, and 21K at the second electrophoresis. A repeated SDS-PAGE did not improve the purity of bank 105K, but increased the ratio of 21K to 28K. Compared with P0 protein, band 105K has a very similar peptide map pattern and amino acid composition, as well as the identical NH2 terminal residue, isoleucine. These findings suggest that band 105K is an aggregate form of P0 protein and its fragment, 21K. The 21K protein is a distinct entity from X protein. The quantitative and qualitative alterations in myelin proteins, as we report here, may reflect continuing demyelination and remyelination in aged peripheral nerves. 相似文献
