Phenotypic diversity of five Jordanian populations of cyst nematodes, Heterodera spp. collected from five regions from Jordan (Ar-Ramtha, Madaba, Dana, Al-Karak, and Jerash) was investigated. Soil samples were collected from one representative field in each region. Morphological and morphometrical characteristics revealed that Heterodera latipons is dominated in cereal fields at Ar-Ramtha, Madaba, Dana and Al-Karak regions and Heterodera schachtii in Jerash. Cysts populations from all cereal fields had bifenestrate vulval cone and a strong underbridge. Wherever, cysts of the cabbage population had ambifenestrate vulval cone with long vulval slit. The bullae were absent in Ar-Ramtha, Madaba and Dana populations, but present in Al-Karak and Jerash. Based on 12 morphometrical characters, the first three functions in canonical discriminant analysis accounted 99.3% of the total variation. Distance from dorsal gland duct opening to stylet base, underbridge length, a = L/W (body length/midbody width) and length of hyaline tail tip had strong and significant contributions in the first function. While the second function was strongly influenced by length of hyaline tail, fenestral length, fenestral width and tail length. However, the third canonical discriminate function was found to be influenced by stylet length, fenestral length, a = L/W (body length/midbody width) and underbridge width. The graphical representation of the distribution of the samples showed that the first canonical discriminant function clearly separated H. schachtii from Jerash from other populations. Whereas, H. latipons collected from Madaba and Dana were clearly separated in the second function. The results indicated that differences at morphological and morphometrical levels revealed diverse populations of Heterodera spp. in Jordan.  相似文献

We present measurements of the effective spontaneous curvature of fluid lipid bilayers as a function of trans-bilayer asymmetry. Experiments are performed on micrometer-scale vesicles in sugar solutions with varying species across the membrane. There are two effects leading to a preferred curvature of such a vesicle. The spontaneous curvatures of the two monolayers as well as their area difference combine into an effective spontaneous curvature of the membrane. Our technique for measuring this parameter allows us to use vesicle morphology as a probe for general membrane-solute interactions affecting elasticity. QTL for stem sugar-related and other agronomic traits were identified in a converted sweet (R9188) × grain (R9403463-2-1) sorghum population. QTL analyses were conducted using phenotypic data for 11 traits measured in two field experiments and a genetic map comprising 228 SSR and AFLP markers grouped into 16 linkage groups, of which 11 could be assigned to the 10 sorghum chromosomes (SBI-01 to SBI-10). QTL were identified for all traits and were generally co-located to five locations (SBI-01, SBI-03, SBI-05, SBI-06 and SBI-10). QTL alleles from R9188 were detected for increased sucrose content and sugar content on SBI-01, SBI-05 and SBI-06. R9188 also contributed QTL alleles for increased Brix on SBI-05 and SBI-06, and increased sugar content on SBI-03. QTL alleles from R9403463-2-1 were found for increased sucrose content and sucrose yield on SBI-10, and increased glucose content on SBI-07. QTL alleles for increased height, later flowering and greater total dry matter yield were located on SBI-01 of R9403463-2-1, and SBI-06 of R9188. QTL alleles for increased grain yield from both R9403463-2-1 and R9188 were found on SBI-03. As an increase in stem sugars is an important objective in sweet sorghum breeding, the QTL identified in this study could be further investigated for use in marker-assisted selection of sweet sorghum.  相似文献

Abstract The complete nucleotide sequence of the gene encoding the Corynebacterium glutamicum mannose enzyme II (EIIMan) was determined. The gene consisted of 2052 base pairs encoding a protein of 683 amino acid residues; the molecular mass of the protein subunit was calculated to be 72570 Da. The N-terminal hydrophilic domain of EIIMan showed 39.7% homology with a C-terminal hydrophilic domain of Escherichia coli glucose-specific enzyme II (EIIGlc). Similar homology was shown between the C-terminal sequence of EIIMan and the E. coli glucose-specific enzyme III (EIIIGlc), or the EIII-like domain of Streptococcus mutans sucrose-specific enzyme II. Sequence comparison with other EIIs showed that EIIMan contained residues His-602 and Cys-28 which were homologous to the potential phosphorylation sites of EIIIGlc, or EIII-like domains, and hydrophilic domains (IIB) of several EIIs, respectively.  相似文献

Riboflavin-binding proteins (RBP) purified from chicken egg white, yolk and the serum of laying hens differ in their carbohydrate compositions reflecting tissue-specific modifications of a single gene product. All three are complex glycoproteins having more than twice as many N-acetylglucosamine residues (>12) as mannose residues (approx. 6). Egg white RBP is distinctive in having only one sialic acid and two galactose residues. Serum RBP contains approx. five sialic acid and seven galactose residues. In addition there is one residue of fucose. The carbohydrate composition of yolk RBP indicates the hydrolysis, respectively, of one, one, two and 3 residues of sialic acid, fucose, galactose, and N-acetylglucosamine from its precursor, serum RBP. The effect of these differing levels of glycosylation on plasma clearance, ovarian uptake and tissue distribution of 125I-labeled riboflavin-binding proteins in laying hens were compared. 2 h after intravenous injection, 19% of the egg white RBP, 29% of the yolk RBP, and 37% of the serum RBP remained in circulation. The kinetics of plasma clearance was distinctly biphasic for each of the radioiodinated proteins. The initial rapid-turnover component (t12=13 min) ranged from 27% of the serum RBP sample to 48% of the egg white RBP sample. The remaining slow-turnover components were cleared with half-lives fo 81 min (egg white RBP), 101 min (yokl RBP), and 121 min (serum RBP). 16 h after injection, only 4% of the egg white RBP was deposited in the yolk of developing oocytes while about 12% of the serum RBP and yolk RBP was deposited. This higly significant difference is apparently due to preferential, carbohydrate-dependent clearance of egg white RBP by the liver rather than preferential uptake of serum and yolk RBP by the ovarian follicle. We find no evidence for carbohydrate-directed uptake of riboflavin-binding protein by the ovarian follicle.  相似文献

Changes in the composition of a Fagus-Acer (Beech-Sugar maple) forest in southeastern Wisconsin over a 16-yr period from 1971 to 1987 are analyzed in relation to a severe glaze (ice) storm disturbance occurring within the census period. Landscape topography created ‘windward’ and ‘leeward’ forest aspects with respect to storm severity, which resulted in greater canopy opening on the windward aspect. In the tree stratum, most species remained stable in density and most of the common species increased in basal area into larger size classes. However, Fagus grandifolia, Ulmus rubra, and the small tree Ostrya virginiana suffered net losses that suggest synergistic effects between glaze storm disturbance and other factors upon tree mortality. In the sapling stratum, canopy opening strongly promoted release of shade-to levant Acer sac-charum. On the windward forest aspect, sapling densities of less shade-tolerant species also increased, in contrast to the absence of such increases on the leeward forest aspect. In the shrub (regeneration) stratum, species responses were heterogeneous. Regeneration of most species increased over the 16-yr period, and some less shade-tolerant species showed increased regeneration differentially on the windward forest aspect. Overall, disturbance appears to have accelerated forest succession toward increased dominance by A. saccharum and persistence of both Fagus and Tilia americana through their capacities for root sprouting. However, forest succession was retarded somewhat on the windward aspect through increased recruitment of less-shade tolerant species. These results parallel those of other studies of glaze storm disturbance, and they illustrate how spatially heterogeneous disturbance intensity may contribute to maintenance of forest diversity.  相似文献

Selection reduces additive genetic variation by generating gametic-phase disequilibrium, a phenomenon largely ignored when predicting response in plant breeding programs. The development of gametic-phase disequilibrium is here taken into account when predicting the response to selection for various schemes of recurrent selection applicable to plant populations. A general program permits prediction of response to selection from schemes of recurrent selection in which two or more rounds of selection occur in each cycle. An example from Sugar Beet, with alternate rounds of half-sib and S1 family selection, is illustrated. It is shown that failure to take into account the effects of gametic-phase disequilibrium can result in substantial overestimation of the response to selection as well as to changes in rank of the merits of alternative breeding schemes. For a given scheme, ignoring gametic-phase disequilibrium has only small effects on defining the optimum allocation of plots and the numbers of families tested.  相似文献

In the frog intestine, both in vitro and in vivo, experiments were carried out in order to increase knowledge of the mechanism of sugar exit across the basolateral membrane of the enterocyte. The frog intestine was chosen because it lacks crypt cells and, consequently, any external fluid circuit mechanism during sugar transport can be avoided. Therefore, the sugar concentration in the absorbate collected on the serosal side is likely to be similar to that present underneath the basolateral membrane of the enterocyte. Under this condition, cell and absorbate sugar concentrations are similar; yet there is a concomitant net transintestinal sugar transport. Moreover, in in vivo experiments a net transintestinal sugar transport takes place even against a concentration difference. These results suggest that sugar exit across the basolateral membrane is not simply due to a chemically facilitated diffusion.  相似文献

(1) The effects of vanadate of hexose transport, 45Ca-exchange and (Na+, K+)-contents have been characterized in isolated adipose tissue and skeletal muscles of the rat. (2) In whole epididymal fat pads, vanadate (0.5–5.0 mM) markedly stimulated the uptake of 2-deoxyl[14C]glucose as well as the efflux of 3-O-[14C]methylglucose. (3) Within the same concentration range, vanadate induced an early increase in 45Ca-washout from preloaded fat pads. The maximum increases in the fractional losses of 3-O-[14C]methylglucose and 45Ca were significantly correlated (P < 0.001, r = 0.98). (4) In extensor digitorum longus and soleus muscles, vanadate (0.5–5.0 mM) stimulated the efflux of 3-O-[14C]methylglucose and this effect was preceded by a rise in the washout of 45Ca. The maximum increases in the fractional losses of 3-O-[14C]methyglucose and 45Ca were significantly correlated (P < 0.005, r = 0.98). (5) In extensor digitorum longus and soleus muscles, vanadate increased K+-contents and decreased Na+ contents. (6) The stimulation of 45Ca-washout presumably reflects an increase in the cytoplasmic Ca2+ level, brought about by an inhibitory effect of vanadate on the Ca2+-sensitive ATPase of the sarcoplasmic or the endoplasmic reticulum. As demonstrated for most other insulin-like agents (Sørensen, S.S., Christensen, F. and Clausen, T. (1980) Biochim. Biophys. Acta 602, 433–445), the stimulating effect of vanadate on glucose transport appears to be associated with or mediated by a rise in the cytoplasmic Ca2+ level.  相似文献
