Septins are a highly conserved family of GTP‐binding proteins that contribute to many cellular and metabolic functions, including cell polarity, cytokinesis, cell morphogenesis and pathogenesis. In this study, we characterized the septins FaCdc3 and FaCdc12 in the filamentous fungus Fusarium asiaticum. The functions of FaCdc3 and FaCdc12 were evaluated by constructing deletion mutants of FaCdc3 and FaCdc12, designated ΔFaCdc3‐5 and ΔFaCdc12‐71, respectively. The deletion mutants exhibited a reduced rate of mycelial growth, increased aerial hyphae formation, irregularly shaped hyphae, reduced conidiation and a lack of sexual reproduction in wheat kernels. Histochemical analysis revealed that the conidia and hyphae of ΔFaCdc3‐5 and ΔFaCdc12‐71 formed large lipid droplets (LDs). ΔFaCdc3‐5 and ΔFaCdc12‐71 also exhibited increased resistance to agents that induce osmotic stress and damage the cell membrane and cell wall. In addition, the hyphae and conidia of the two mutants formed fewer septa than those of the wild‐type and exhibited aberrant nuclear distribution. Pathogenicity assays showed that ΔFaCdc3‐5 and ΔFaCdc12‐71 exhibited reduced virulence on wheat spikelets, which was indirectly correlated with a reduced level of deoxynivalenol accumulation. All of these defects were restored by genetic complementation of the two mutants with the parental FaCdc3 and FaCdc12. These results indicate that FaCdc3 and FaCdc12 play a critical role in various cellular processes in F. asiaticum.  相似文献

对光皮木瓜果实发育期间主要成分含量的变化进行了分析.结果显示:(1)光皮木瓜果实发育期间,可溶性固形物含量总体呈上升趋势,花后151 d达到最大值14.8%;干物质、粗纤维和多酚含量的变化均呈单峰型,分别于花后95、67和95 d达到峰值(26.49%、14.96%、81.843 1 mg/gDW),并分别于花后165、179和179 d降至最低值(18.3%、6.47%、42.014 6 mg/gDW);黄酮、多糖、齐墩果酸含量均呈逐渐增加趋势,在果实发育后期略有下降,其中多糖和齐墩果酸含量均于花后137 d达到最大值,分别为110.431 2 mg/gDW和10.312 1 mg/gDW,黄酮含量在花后151 d达到最大值3.201 1 mg/gDW.(2)多酚的单果产量在果实发育过程中的变化趋势与多酚含量一致,于花后95 d多酚单果产量达到最高值(5.448 6 g);齐墩果酸、多糖及黄酮的单果产量在果实发育期间逐渐增加,花后165 d多糖的单果产量达到最高值(8.508 1 g),花后179 d黄酮和齐墩果酸的单果产量均达到最高值(0.261 2 g和0.747 4 g).研究表明,花后137 d(8月25日)至151 d(9月8日)木瓜果实青绿,齐墩果酸、多糖、黄酮含量最高,为加工木瓜饮片等的最佳采收期;花后165 d(9月22日)至179 d(10月6日)时,木瓜果实基本转黄,其中齐墩果酸、多糖、黄酮的单果产量最高,且多酚和粗纤维含量最低,是药用有效成分提取和加工果酒、果脯、果醋等木瓜食品的最佳采收期.  相似文献

Many insects undergo diapause to survive adverse seasons. Although the mechanism of diapause induction is the subject of extensive study, that of diapause termination remains poorly understood. In the present study, we show the endocrine processes leading to the termination of pupal diapause in Mamestra brassicae. Diapause of this insect is terminated if the pupae are exposed to a low temperature for several weeks. During this period, the prothoracic glands (PGs) of pupae acquire the potential to secrete sufficient ecdysteroids necessary for inducing adult development. The main endocrine changes observed under the low temperature conditions are: (i) the increase in activity of the PGs in two steps; (ii) the increase in responsiveness of the glands to prothoracicotropic hormone (PTTH); and (iii) two‐step increase in PTTH gene expression in the brain. The timing of the first and second increases in PG activity roughly coincides with that of the two steps of increase in PTTH gene expression, and the timing of the increase in the responsiveness of the PGs to PTTH coincides with the second, larger increase in PTTH gene expression. The ablation of the PGs prior to cooling pupae does not affect the increase in PTTH gene expression, whereas brain removal results in a failure to increase PG activity, strongly suggesting that PTTH is the master regulator of diapause development and termination.  相似文献

Background and Aims

Experimental evidence challenges the approximation, central in crop models, that developmental events follow a fixed thermal time schedule, and indicates that leaf emergence events play a role in the timing of development. The objective of this study was to build a structural development model of maize (Zea mays) based on a set of coordination rules at organ level that regulate duration of elongation, and to show how the distribution of leaf sizes emerges from this.

Methods

A model of maize development was constructed based on three coordination rules between leaf emergence events and the dynamics of organ extension. The model was parameterized with data from maize grown at a low plant population density and tested using data from maize grown at high population density.

Key Results

The model gave a good account of the timing and duration of organ extension. By using initial conditions associated with high population density, the model reproduced well the increase in blade elongation duration and the delay in sheath extension in high-density populations compared with low-density populations. Predictions of the sizes of sheaths at high density were accurate, whereas predictions of the dynamics of blade length were accurate up to rank 9; moderate overestimation of blade length occurred at higher ranks.

Conclusions

A set of simple rules for coordinated growth of organs is sufficient to simulate the development of maize plant structure without taking into account any regulation by assimilates. In this model, whole-plant architecture is shaped through initial conditions that feed a cascade of coordination events.  相似文献

黄土丘陵区生物土壤结皮表面糙度特征及影响因素   总被引:1,自引:0,他引:1  
地表糙度是影响地表径流和侵蚀过程的重要属性.生物结皮在干旱半干旱区广泛分布,是地表糙度的影响因子之一.本文采用链条法测定了黄土丘陵区不同发育阶段生物结皮表面糙度特征,分析了不同发育阶段生物结皮表面糙度对坡向、土壤含水量和冻融作用的响应及其与各理化性质的相关性,初步探索了生物结皮对地表糙度的影响及其相关因素.结果表明: 生物结皮显著改变地表糙度,随着生物结皮从藻结皮向藓结皮演替,其糙度先降低后增加,生物结皮发育形成10年以后,其表面糙度基本趋于稳定;研究区早期形成的藻结皮表面糙度较裸土降低47.0%,深色藻结皮(藓类盖度<20%)较裸土降低20.4%,混生结皮(藓类盖度为20%~60%)和苔藓结皮(藓类盖度>70%)表面糙度与深色藻结皮基本一致;坡向对发育10年以上的生物结皮表面糙度的影响不显著;土壤含水量影响地表糙度特征.研究区浅色藻结皮表面糙度随水分变化较为剧烈;随着生物结皮发育,深色藻结皮、混生结皮和苔藓结皮表面糙度随水分的变化趋于平缓.冻融增加了生物结皮表面糙度.浅色藻结皮经两次冻融后表面糙度增加29.7%;深色藻结皮、混生结皮和藓结皮表面糙度的影响需经过反复冻融才有所体现.生物结皮表面糙度与藓结皮盖度呈显著正相关(P<0.1).  相似文献

Fragments of several lichens were grown in the laboratory and developmental steps of growth and regeneration could be described. Different degrees of determination could be observed. The stages of podetial development inCladonia are easily distinguished and are reached after comparably short time. As they occur highly synchronized the results encourage further morphogenetic experiments.Dedicated to Prof. DrO. L.
