目的:吴茱萸次碱(Rutaecarpa RUT)是芸香科植物吴茱萸的主要成分,它可激活辣椒素受体促进降钙素基因相关肽(CGRP) 等神经递质的释放来发挥药理作用。本实验的目的为观察吴茱萸次碱软膏对小鼠银屑病模型治疗作用。方法:通过药剂学方法制 成了不同浓度(2%, 5%, 10%)RUT 软膏,在小鼠尾部与背部银屑病模型给药14 天,观察小鼠尾部颗粒层形成的变化,给药结束后 取小鼠背部皮肤组织匀浆后采用放射免疫方法检测CGRP水平变化,并且取小鼠血浆检测CGRP水平变化。结果:不同浓度的吴 茱萸次碱软膏均能促进小鼠尾部颗粒层形成(P<0.05),并且浓度为5%与10%的RUT 软膏能显著降低小鼠背部CGRP 含量(P<0. 05),但是其对小鼠血浆的CGRP 没有影响。结论:吴茱萸次碱外用对小鼠银屑病模型有一定的治疗作用,这种作用与其促进小鼠 皮肤CGRP 释放而导致其含量降低有关。  相似文献

【背景】从海南热带海区中分离得到一株微藻,其生长速度快、适应力强,经鉴定该微藻为普通小球藻。【目的】提高热带普通小球藻的生长速率。【方法】以"宁波大学3#微藻培养液配方"为基础培养液,分别添加有机碳(C6H12O6和CH3COONa)对热带普通小球藻进行自养、兼养及异养培养,获得促进热带普通小球藻快速生长的培养方式。在"宁波大学3#微藻培养液配方"的基础上对热带普通小球藻的兼养培养基配方进行优化,并用优化兼养培养基与"宁波大学3#微藻培养基"对比培养热带普通小球藻。【结果】添加6 g/L CH3COONa的兼养模式促进热带普通小球藻生长效果最好;优化的兼养培养基配方为:6 g/L CH3COONa,20 mg/L(NH4)2SO4-N,5 mg/L Na H2PO4-P,3 mg/L Fe SO4-Fe,1 mg/L Vitamin B1和0.000 5 mg/L Vitamin B12。对比培养实验结果显示,培养第6天,兼养培养液收获的生物量(细胞密度)达4.20×107 cells/m L,是"宁波大学3#配方微藻培养液"的2.30倍。【结论】兼养培养模式为热带普通小球藻的最佳培养模式,优化的兼养培养基极显著地提高了热带普通小球藻的生物量(P0.01)。  相似文献

目的:报道一组以肢端出现对称性、棕褐色、角化过度性斑片病例,探讨发病原因和治疗方法。方法:系统收集自2012年10月至2016年6月期间诊疗的以对称性肢端角化性皮损为表现的9例患者的临床资料,包括年龄、性别、职业、家族成员情况、临床表现、皮损组织病理学改变、治疗、治疗转归等。结果:9例患者中男8例,女1例,平均年龄27.4岁(13~43岁),病程2个月至15年,平均(4.5±4.3)年。有1例男性患者有一同胞姐姐有类似病史。临床主要表现为红褐色角化性斑片,每年7、8月开始发病,10月左右开始缓解并渐完全消退。部分手部有轻度瘙痒不适。皮肤组织病理学检查表现为明显的网篮状角化过度,轻度乳头瘤样增生,真皮浅层血管周围稀疏淋巴细胞浸润。1例用异维A酸治疗短期有效。结论:对称性肢端角化病主要以肢端出现对称性角化斑为主,常伴随寻常型鱼鳞病;遗传因素和环境因素在发病中的作用有待阐述;异维A酸可作为一种治疗选择。  相似文献

以小球藻及莱茵衣藻原生质体为受体细胞,利用电击法将质粒p CAMBIA1301转入小球藻和莱茵衣藻,摸索电击转化条件并进行分子检测。结果表明:两类藻都对潮霉素敏感,小球藻及莱茵衣藻分别在含25 mg/L和100 mg/L潮霉素的固体培养基上的生长被完全抑制;小球藻和莱茵衣藻原生质体电击转化的最佳电击场强分别为0.8 k V/cm和0.6 k V/cm,最佳脉冲时间均为10 ms;制备原生质体和通过2-脱氧-D-葡萄糖处理可明显提高转化效率;分子检测说明GUS报告基因成功转入两种藻并可稳定遗传。  相似文献

Abstract. Activation spectra of photochemical reactions were measured by a flash spectrophotometer in leaves having varying chlorophyll contents at different stages of greening. The increase of chlorophyll concentration up to 30 nmol cm-2 elevated the rates of photochemical reactions at all wavelengths of light used, and was found to be produced by an increase in the amounts of reaction centres. Further accumulation of chlorophyll up to 40 nmol cm-2 was associated with an increase in light-harvesting chlorophyll, an improved rate of photochemical reactions around 600 nm and at 700 nm, and self-absorption and screening effects where chlorophyll absorbed maximally (400–450 nm and around 680 nm).  相似文献

The kinetics of NADH oxidation by the outer membrane electron transport system of intact beetroot (Beta vulgaris L.) mitochondria were investigated. Very different values for Vmax and the Km for NADH were obtained when either antimycin A-insensitive NADH-cytochrome c activity (Vmax= 31 ± 2.5 nmol cytochrome c (mg protein)?1 min?1; Km= 3.1 ± 0.8 μM) or antimycin A-insensitive NADH-ferricyanide activity (Vmax= 1.7 ± 0.7 μmol ferricyanide (mg protein)?1 min?1; Km= 83 ± 20 μM) were measured. As ferricyanide is believed to accept electrons closer to the NADH binding site than cytochrome c, it was concluded that 83 ± 20 μM NADH represented a more accurate estimate of the binding affinity of the outer membrane dehydrogenase for NADH. The low Km determined with NADH-cytochrome c activity may be due to a limitation in electron flow through the components of the outer membrane electron transport chain. The Km for NADH of the externally-facing inner membrane NADH dehydrogenase of pea leaf (Pisum sativum L. cv. Massey Gem) mitochondria was 26.7 ± 4.3 μM when oxygen was the electron acceptor. At an NADH concentration at which the inner membrane dehydrogenase should predominate, the Ca2+ chelator, ethyleneglycol-(β-aminoethylether)-N,N,-tetraacetic acid (EGTA), inhibited the oxidation of NADH through to oxygen and to the ubiquinone-10 analogues, duroquinone and ubiquinone-1, but had no effect on the antimycin A-insensitive ferricyanide reduction. It is concluded that the site of action of Ca2+ involves the interaction of the enzyme with ubiquinone and not with NADH.  相似文献

A field experiment was carried out to determine whether group-foragingstarlings (Sturnus vulgaris) use public information to helpthem estimate the quality of an artificial resource patch anddepart accordingly. Three kinds of information are potentiallyavailable in a group: patch-sample information, pre-harvestinformation, and public information. These three types of informationcan be combined into four patch assessment strategies: (1) patch-samplealone; (2) patch-sample and pre-harvest; (3) patch-sample andpublic; and (4) patch-sample, pre-harvest, and public. Dependingon the foraging environment we presented to the starlings, eachassessment strategy made a unique set of predictions concerningthe patch departure decisions of pairs of birds based on differencesin their foraging success. The environment was manipulated intwo ways: by altering the variability in patch quality and bychanging compatibility, the ease with which individual birdscould simultaneously acquire both patch-sample and public information.Our observations on patch persistence and departure order demonstratethat the starlings used a combination of patch-sample and publicinformation, but not pre-harvest information, to estimate thequality of the experimental patch. Moreover, our results suggestthat starlings use public information only when it is easilyavailable and ignore it under incompatible conditions. Thisstudy provides the first evidence of public information usein a patch assessment problem.  相似文献

目的:探索夏枯草对人甲状腺乳头状癌细胞(K1)增殖和诱导K1细胞凋亡的影响及其可能的作用机制。方法:采用MTT比色法测定2~200 mg/mL夏枯草作用24 h对K1细胞增殖的影响;采用Hoechst染色法和流式细胞术(Annexin V-FITC/PI联合标记)观察0.3、0.6、1.2、2.4、4.8 mg/mL夏枯草作用24 h K1细胞凋亡的影响;采用蛋白质免疫印迹法(Western blotting)测定1.2、2.4、4.8 mg/mL夏枯草作用24h对K1细胞凋亡相关蛋白表达的影响。结果:在2~200 mg/mL的浓度范围内夏枯草作用24 h对K1细胞增殖有明显抑制作用(P0.05),IC50值为2.427 mg/mL。流式细胞术检测结果显示夏枯草可诱导K1细胞凋亡,2.4、4.8 mg/mL组K1细胞总凋亡率分别为(11.35±0.92)%、(44.57±3.07)%,与对照组相比明显升高(P0.05);与对照组相比,1.2、2.4、4.8 mg/mL夏枯草作用24 h胞浆内凋亡蛋白Bax、细胞色素C(Cyto C)、Caspase-9、活化的Caspase-3(Cleaved Caspase-3)表达增多。结论:夏枯草能抑制人甲状腺癌K1细胞增殖并诱导其凋亡,其机制可能与活化线粒体凋亡通路有关。  相似文献

Electron spin resonance (ESR) spectroscopy was used to demonstrate that free radicals are formed in O3-fumigated plant leaves prior to the formation of visible leaf injury. ESR signals with a g-value of 2.0037 to 2.0043, were observed in pea ( Pisum sativum L. cv. Feltham first) and bean ( Phaseolus vulgaris L. cv. Pinto) plants that had been fumigated for 4 h with 70–300 nl l−1 of ozone after they had been treated with the spin-trap N- t -butyl-α-phenylnitrone (PBN). The size of the ESR signals increased with the concentration of ozone used but the nature of the trapped radicals could not be identified. However, further experiments using an inhibitor of ethylene biosynthesis, arninoethoxyvinyl glycine (AVG), showed that the reaction between ozone and ethylene is the cause for ozone toxicity.  相似文献

True bugs (Hemiptera) are an important pest complex not controlled by Bt‐transgenic crops. An alternative source of resistance includes inhibitors of digestive enzymes, such as protease or amylase inhibitors. αAI‐1, an α‐amylase inhibitor from the common bean, inhibits gut‐associated α‐amylases of bruchid pests of grain legumes. Here we quantify the in vitro activity of α‐amylases of 12 hemipteran species from different taxonomic and functional groups and the in vitro inhibition of those α‐amylases by αAI‐1. α‐Amylase activity was detected in all species tested. However, susceptibility to αAI‐1 varied among the different groups. α‐Amylases of species in the Lygaeidae, Miridae and Nabidae were highly susceptible, whereas those in the Auchenorrhyncha (Cicadellidae, Membracidae) had a moderate susceptibility, and those in the Pentatomidae seemed to be tolerant to αAI‐1. The species with αAI‐1 susceptible α‐amylases represented families which include both important pest species but also predatory species. These findings suggest that αAI‐1‐expressing crops have potential to control true bugs in vivo.  相似文献
