The Chlamydomonas reinhardtii truncated light-harvesting antenna 4 (tla4) DNA transposon mutant has a pale green phenotype, a lower chlorophyll (Chl) per cell and a higher Chl a/b ratio in comparison with the wild type. It required a higher light intensity for the saturation of photosynthesis and displayed a greater per chlorophyll light-saturated rate of oxygen evolution than the wild type. The Chl antenna size of the photosystems in the tla4 mutant was only about 65% of that measured in the wild type. Molecular genetic analysis revealed that a single plasmid DNA insertion disrupted two genes on chromosome 11 of the mutant. A complementation study identified the “chloroplast signal recognition particle 54” gene (CpSRP54), as the lesion causing the tla4 phenotype. Disruption of this gene resulted in partial failure to assemble and, therefore, lower levels of light-harvesting Chl-binding proteins in the C. reinhardtii thylakoids. A comparative in silico 3-D structure-modeling analysis revealed that the M-domain of the CpSRP54 of C. reinhardtii possesses a more extended finger loop structure, due to different amino acid composition, as compared to that of the Arabidopsis CpSRP54. The work demonstrated that CpSRP54 deletion in microalgae can serve to generate tla mutants with a markedly smaller photosystem Chl antenna size, improved solar energy conversion efficiency, and photosynthetic productivity in high-density cultures under bright sunlight conditions.  相似文献

Abstract. The Rothamsted Park Grass Experiment was established in 1856, with experimental plots subjected to annual applications of fertilizer and twice-yearly cutting of hay. There were two major responses to fertilizer, one reflecting high ammonium-nitrogen and increased acidity and the other reflecting high herbage yield without increased acidity. We calculated mean Ellenberg indicator values for N (nitrogen) and R (soil reaction) for the hay harvested between 1948 and 1975, using both unweighted and abundance-weighted means. Plot Ellenberg values were compared with herbage yield and with fertilizer application rates and published soil data. Annual yield of hay varied from 1.5 to 7.4 t/ha and was well predicted by the unweighted mean Ellenberg N-values (r = 0.91). Relatively large negative residuals from the relationship were found in plots whose soil combined low K and low pH. Soil pH was poorly predicted by the unweighted mean R-value, but showed a moderately good relation with weighted mean R (r = 0.73). The fact that Ellenberg N-values correlated better with yield than with applied nitrogen suggests that they might rather be called productivity values.  相似文献

The addition of nutrients has been shown to decrease the species richness of plant communities. Herbivores feed on dominant plant species and should release subdominant species from competitive exclusion at high levels of nutrient availability with a severe competitive regime. Therefore, the effects of nutrients and invertebrate herbivory on the structure and diversity of plant communities should interact. To test this hypothesis, we used artificial plant communities in microcosms with different levels of productivity (applying fertilizer) and herbivory (adding different numbers of the snail, Cepaea hortensis, and the grasshopper, Chorthippus parallelus). For analyses, we assigned species to three functional groups: grasses, legumes and (non-leguminous) herbs. With the addition of nutrients aboveground biomass increased and species richness of plants decreased. Along the nutrient gradient, species composition shifted from a legume-dominated community to a community dominated by fast-growing annuals. But only legumes showed a consistent negative response to nutrients, while species of grasses and herbs showed idiosyncratic patterns. Herbivory had only minor effects, and bottom–up control was more important than top–down control. With increasing herbivory the biomass of the dominant plant species decreased and evenness increased. We found no interaction between nutrient availability and invertebrate herbivory. Again, species within functional groups showed no consistent responses to herbivory. Overall, the use of the functional groups grasses, legumes and non-leguminous herbs was of limited value to interpret the effects of nutrients and herbivory during our experiments.  相似文献

Recent studies on global climate change report that increase in seawater temperature leads to coastal ecosystem change, including coral bleaching in the tropic. In order to assess the effect of increased seawater temperature on a temperate coastal ecosystem, we studied the inter-annual variation in productivity of Laminaria japonica using long-term oceanographic observations for the Uwa Sea, southern Japan. The annual productivity estimates for L. japonica were 2.7 ± 2.5 (mean ± SD) kg wet wt. m−1 (length of rope) (2003/2004), 1.0 ± 0.6 kg wet wt. m−1 (2004/2005) and 12.1 ± 12.5 kg wet wt. m−1 (2005/2006). Our previous study using the same methodology at the same locality reported that the productivity was estimated for the 2001/2002 (33.3 ± 15.2 kg wet wt. m−1) and 2002/2003 (34.0 ± 8.7 kg wet wt. m−1) seasons. Productivity in 2003/2004 and 2004/2005 was significantly lower than in years 2001/2002, 2002/2003 and 2005/2006. A comparison of oceanographic conditions among the 5 years revealed the presence of threshold seawater temperature effects. When the average seawater temperature during the first 45 days of each experiment exceeded 15.5°C, productivity was reduced to about 10 % of that in cooler years. Moreover the analysis of growth and erosion rates indicates that when the seawater temperature was over 17.5°C, erosion rate exceeded growth rate. Thus, an increase of seawater temperature of just 1°C during winter drastically reduces the productivity of L. japonica in the Uwa Sea.  相似文献

For analyzing sustainability of algal biofuels, we identify 16 environmental indicators that fall into six categories: soil quality, water quality and quantity, air quality, greenhouse gas emissions, biodiversity, and productivity. Indicators are selected to be practical, widely applicable, predictable in response, anticipatory of future changes, independent of scale, and responsive to management. Major differences between algae and terrestrial plant feedstocks, as well as their supply chains for biofuel, are highlighted, for they influence the choice of appropriate sustainability indicators. Algae strain selection characteristics do not generally affect which indicators are selected. The use of water instead of soil as the growth medium for algae determines the higher priority of water- over soil-related indicators. The proposed set of environmental indicators provides an initial checklist for measures of algal biofuel sustainability but may need to be modified for particular contexts depending on data availability, goals of stakeholders, and financial constraints. Use of these indicators entails defining sustainability goals and targets in relation to stakeholder values in a particular context and can lead to improved management practices.  相似文献

 许多有关物种多样性-生态系统功能关系的观察、理论和实验研究都表明, 在局域尺度范围内, 植物种多样性对生态系统生产力存在正效应。 然而, 对于促成这种关系的潜在生态学机制却缺乏足够的了解。 该实验利用9种一年生栽培牧草, 采用各物种单播及混播的方法, 构建不同多样性梯度的实验群落, 对物种多样性与生态系统生产力的关系及资源互补效应对系统生产力的影响进行了研究。 结果表明, 在一年生植物群落内,植物种多样性在一定程度内对系统生产力存在正效应, 物种多样性与生产力呈二次函数关系, 关系式为y = -98.449x2 + 1 039.2 x - 42.407, (R2 = 0.423 1)。 各物种在资源利用、生长速度和竞争能力等功能特征方面存在较大差异, 最高产物种和最低产物种间产量相差5.8倍。 在同一多样性梯度内, 不同物种组合的群落间生产力和互补效应也存在较大差异, 说明物种的成分对生态系统生产力也有重要影响。 同时,在混播群落中程度不同地存在着资源的互补性利用, 说明物种多样性对系统生产力有增强作用, 但相关分析表明, 互补效应和物种多样性间不存在显著相关关系。互补效应的4种计算方法所反映的资源互补程度有所不同, 每种方法各有利弊, 在对系统的多样性效应作用机制进行评价时, 应根据具体情况, 同时采用几种方法, 以利于对资源互补效应做出恰当的估测。  相似文献

1987年发生在大兴安岭北坡的特大火灾造成了森林资源的巨大损失,森林景观恢复一直是人们关注的热点.本研究选取与森林生产力密切相关的郁闭度因子作为研究对象,以ArcView、ArcGIS等地理信息系统软件为研究平台,采用1987年和2000年两期森林资源二类调查数据,对郁闭度、火烧强度、抚育类型和地形因子等进行分级,利用Kendall等级相关分析、相似性分析等方法,探讨了森林郁闭度格局的恢复状况以及火烧强度、更新类型、地形因子对郁闭度恢复的影响.结果表明,2000年郁闭度等级构成与1987年火前相比发生了明显变化,无林地以及高郁闭度等级比重明显下降,较低郁闭度等级比重显著上升;火烧强度对火后恢复的影响最为关键,火烧强度与郁闭度等级呈负相关;更新措施短期内对郁闭度恢复影响不显著,但可以缩短森林演替的周期,对未来针叶林群落生产力恢复具有重要的促进作用;地形因子中坡度对郁闭度恢复影响最为明显,其次为坡位,坡向影响最弱.  相似文献

Question: What is the value of using Rhinanthus minor in grassland restoration and can restrictions on its establishment be overcome? Location: England (United Kingdom). Methods: Two experiments were established to determine the efficacy of inoculating R. minor on a suite of four agriculturally improved grasslands and the efficacy of using R. minor in grassland restoration. In Experiment 1, the effect of herbicide gap creation on the establishment and persistence of R. minor in grasslands ranging in productivity was investigated with respect to sward management. In Exp. 2, R. minor was sown at 1000 seeds/m2 in conjunction with a standard meadow mix over a randomized plot design into Lolium perenne grassland of moderate productivity. The treatment of scarification was investigated as a treatment to promote R. minor. Results: Gap size had a significant role in the establishment and performance of R. minor, especially the 30 cm diameter gaps (Exp. 1). However, R. minor failed to establish long‐term persistent populations in all of the agriculturally improved grasslands. In Exp. 2, establishment of R. minor was increased by scarification and its presence was associated with a significant increase in Shannon diversity and the number of sown and unsown species. Values of grass above‐ground biomass were significantly lower in plots sown with R. minor, but values of total above‐ground biomass (including R. minor) and forb biomass (not including R. minor) were not affected. Conclusions: The value of introducing R. minor into species‐poor grassland to increase diversity has been demonstrated, but successful establishment was dependent on grassland type. The scope for using R. minor in grassland restoration schemes is therefore conditional, although establishment can be enhanced through disturbance such as sward scarification.  相似文献

Bioinformatics tools have facilitated the reconstruction and analysis of cellular metabolism of various organisms based on information encoded in their genomes. Characterization of cellular metabolism is useful to understand the phenotypic capabilities of these organisms. It has been done quantitatively through the analysis of pathway operations. There are several in silico approaches for analyzing metabolic networks, including structural and stoichiometric analysis, metabolic flux analysis, metabolic control analysis, and several kinetic modeling based analyses. They can serve as a virtual laboratory to give insights into basic principles of cellular functions. This article summarizes the progress and advances in software and algorithm development for metabolic network analysis, along with their applications relevant to cellular physiology, and metabolic engineering with an emphasis on microbial strain optimization. Moreover, it provides a detailed comparative analysis of existing approaches under different categories.  相似文献
