《Peptides》2015
A new peptide with 61 amino acids cross-linked by 4 disulfide bridges, with molecular weight of 6938.12 Da, and an amidated C-terminal amino acid residue was purified and characterized. The primary structure was obtained by direct Edman degradation and sequencing its gene. The peptide is lethal to mammals and was shown to be similar (95% identity) to toxin Ts1 (gamma toxin) from the Brazilian scorpion Tityus serrulatus; it was named Tt1g (from T. trivittatus toxin 1 gamma-like). Tt1g was assayed on several sub-types of Na+-channels showing displacement of the currents to more negative voltages, being the hNav1.3 the most affected channel. This toxin displays characteristics typical to the β-type sodium scorpion toxins. Lethality tests and physiological assays indicate that this peptide is probably the most important toxic component of this species of scorpion, known for causing human fatalities in the South American continent. 相似文献

Degeneration of mesencephalic dopaminergic (mesDA) neurons is the pathological hallmark of Parkinson’s diseae. Study of the biological processes involved in physiological functions and vulnerability and death of these neurons is imparative to understanding the underlying causes and unraveling the cure for this common neurodegenerative disorder. Primary cultures of mesDA neurons provide a tool for investigation of the molecular, biochemical and electrophysiological properties, in order to understand the development, long-term survival and degeneration of these neurons during the course of disease. Here we present a detailed method for the isolation, culturing and maintenance of midbrain dopaminergic neurons from E12.5 mouse (or E14.5 rat) embryos. Optimized cell culture conditions in this protocol result in presence of axonal and dendritic projections, synaptic connections and other neuronal morphological properties, which make the cultures suitable for study of the physiological, cell biological and molecular characteristics of this neuronal population. 相似文献

Fire can cause severe nitrogen (N) losses from grassland, chaparral, and temperate and boreal forest ecosystems. Paradoxically,
soil ammonium levels are markedly increased by fire, resulting in high rates of primary production in re-establishing plant
communities. In a manipulative experiment, we examined the influence of wild-fire ash residues on soil, microbial and plant
N pools in a recently burned Californian bishop pine (Pinus muricata D. Don) forest. Ash stimulated post-fire primary production and ecosystem N retention through direct N inputs from ash to
soils, as well as indirect ash effects on soil N availability to plants. These results suggest that redistribution of surface
ash after fire by wind or water may cause substantial heterogeneity in soil N availability to plants, and could be an important
mechanism contributing to vegetation patchiness in fire-prone ecosystems. In addition, we investigated the impact of fire
on ecosystem N cycling by comparing 15N natural abundance values from recently burned and nearby unburned P. muricata forest communities. At the burned site, 15N natural abundance in recolonising species was similar to that in bulk soil organic matter. By contrast, there was a marked
15N depletion in the same species relative to the total soil N pool at the unburned site. These results suggest that plant uptake
of nitrate (which tends to be strongly depleted in 15N because of fractionation during nitrification) is low in recently burned forest communities but could be an important component
of eco- system N cycling in mature conifer stands.

The effects of implementing Directive 91/271/EEC of 21 May 1991 (Waste Water Treatment Plan Directive) and Directive 91/676/EEC
of 12 December (Nitrates Directive) are analysed in 7 Portuguese estuaries (Minho, Lima, Douro, Mondego, Tagus, Sado and Guadiana)
and two coastal lagoons (Ria de Aveiro and Ria Formosa), with a modelling approach. MOHID Water Modelling System was used
to perform simulations with three nitrogen load scenarios for each system: a reference scenario, a 50% nitrate removal by
agriculture scenario and another with a 100% nutrients removal by waste water treatment plants (WWTP). It is shown that the
interaction between hydrodynamic and ecological processes is an important feature to study trophic problems in estuaries.
Ecological processes such as primary production only occur inside the system if the residence time of water is high enough
to enable organismal activity and if the adequate conditions are found (e.g. light, nutrients, temperature). From the model
results it is possible to conclude: (i) in systems with short residence time a reduction in nutrient load will only produce
a decrease in nutrient transit and will not affect the system’s global ecological status (e.g. Douro Estuary); (ii) in systems
with long residence time the effects will range from significant, when primary production is mostly limited by nutrients (e.g.
Ria de Aveiro), to non-significant, when primary production in the system is light-limited (e.g. Tagus Estuary). 相似文献

Abstract A diatom biofilm was grown in a chamber developed for culture of biofilms in chemical gradients. The diatoms grew on a polycarbonate membrane filter which separated a sterile reservoir, with added phosphate, from a reservoir without phosphate. Within 3 weeks of inoculation, a thick biofilm developed on the surface of the filter. The biofilms were homogeneous and therefore suitable for calculations of O2 diffusion fluxes from concentration profiles of O2 . Profiles of O2 , pH, and gross photosynthesis at different light intensities and liquid medium concentrations of dissolved inorganic carbon and O2 were measured with microelectrodes. Respiratory activity in a layer of the biofilm was determined as the difference between gross photosynthesis and outflux of O2 from that layer. The photosynthetic activity in a well-developed biofilm grown at 360 μEinst m−2 s−1 and 2.4 mM HCO3 − was limited by the supply of inorganic carbon. Exposure to light above 360 μEinst m−2 s−1 stimulated gross photosynthesis as well as respiratory processes without affecting net outflux of O2 . Higher concentrations of inorganic carbon, on the other hand, enhanced gross photosynthesis without concurrent increase in respiratory rate, resulting in an increased outflux of O2 . High concentrations of O2 in the liquid medium decreased the net outflux of O2 with little effect on the gross photosynthesis. The effects of inorganic carbon and O2 on the metabolic activities of the biofilm were consistent with the presence of photorespiratory activity. 相似文献

Summary . We propose a fully inferential model-based approach to the problem of comparing the firing patterns of a neuron recorded under two distinct experimental conditions. The methodology is based on nonhomogeneous Poisson process models for the firing times of each condition with flexible nonparametric mixture prior models for the corresponding intensity functions. We demonstrate posterior inferences from a global analysis, which may be used to compare the two conditions over the entire experimental time window, as well as from a pointwise analysis at selected time points to detect local deviations of firing patterns from one condition to another. We apply our method on two neurons recorded from the primary motor cortex area of a monkey's brain while performing a sequence of reaching tasks. 相似文献

AbstractNumber of naturally occurring primary sequences of proteins is an infinitesimally small subset of the possible number of primary sequences that can be synthesized using 20 amino acids. Prevailing views ascribe this to slow and incremental mutational/selection evolutionary mechanisms. However, considering the large number of avenues available in form of diversity of emerging/evolving and/or disappearing living systems for exploring the primary sequence space over the evolutionary time scale of ~3.5 billion years, this remains a conjecture. Therefore, to investigate primary sequence space limitations, we carried out a systematic study for finding primary sequences absent in nature. We report the discovery of the smallest peptide sequence “Cysteine-Glutamine-Tryptophan-Tryptophan” that is not found in over half-a-million curated protein sequences in the Uniprot (Swiss-Prot) database. Additionally, we report a library of 83605 pentapeptides that are not found in any of the known protein sequences. Compositional analyses of these absent primary sequences yield a remarkably strong power relationship between the percentage occurrence of individual amino acids in all known protein sequences and their respective frequency of occurrence in the absent peptides, regardless of their specific position in the sequences. If random evolutionary mechanisms were responsible for limitations to the primary sequence space, then one would not expect any relationship between compositions of available and absent primary sequences. Thus, we conclusively show that stoichiometric constraints on amino acids limit the primary sequence space of proteins in nature. 目的:探讨血流阻断的缺血预处理技术在肝癌切除术中的临床应用价值。方法:选取2010年4月至2013年7月我院收治的96名原发性肝癌并采用肝脏部分切除术进行治疗的患者,将患者随机分为观察组和对照组,每组各48例,观察组在肝脏部分切除术阻断肝门血流前先分别给予1个5 min缺血和再灌注的处理。对照组不采取任何干预措施。术前、术后1天、3天、7天时分别进行生化检查,并于术前及术后1h对Fas-mRNA表达、Caspasc-3活性及AI进行测定,观察记录患者术后的并发症情况、手术时间、术中出血量以及住院时间。结果:术后1天、3天、7天时两组间的AST、ALT、TBIL等生化指标的含量情况相比,观察组均显著优于对照组(P0.05);术后l d,两组患者ALB均有不同程度的降低,对照组低于观察组(P0.05);术后住院时间观察组为13.28±3.85天,对照组为19.48±4.92天,观察组明显低于对照组(P0.05);术后1h,两组患者的Fas-mRNA表达、Caspasc-3活性相比于阻断前均显著提高,但观察组提高幅度明显低于对照组,差异有统计学意义(P0.05);两组阻断前均未见肝细胞凋亡,术后l h时,两组组均可见肝细胞凋亡,且对照组明显高于观察组组(P0.05)。结论:血流阻断的缺血预处理技术具有操作简便、副作用小的重要特点,应用于肝癌切除术中在保护肝功能方面具有显著的优势。 相似文献

In recent years, immunodeficiency condition has experienced a rise among children, who are at risk of invasive fungal infections (IFI) due to their health condition. Cancer, non-malignant hematological diseases, as primary immunodeficiencies, hematopoietic stem cell transplantation (HSCT), extreme prematurity, or critically ill condition in Pediatric Intensive Care Unit (PICU) are some immunosuppressive situations in children. The use of oncologic therapies, including immunotherapy and monoclonal antibodies, for the treatment of the aforementioned health conditions has led to an increase in morbidity and mortality rates of IFI in children.The underlying diseases and their management, comorbidities, the diagnostic tests used (both molecular and imaging), as well as the treatment used can be significantly different between adult patients and children admitted to PICU or with cancer. In pediatrics, the treatment of IFI is based primarily on pharmacokinetic studies performed in adults. In higher risk patients prophylaxis should be considered and, in the case of an IFI diagnosis, an antifungal treatment should be administered as early as possible, supported by the reversion of the immune dysfunction and surgery when appropriate. 相似文献

Reconstruction mapping of the natural (primary) vegetation of intensively cultivated land is based on: (1) classification of actually existing remains of natural or near-natural plant communities as mapping units; (2) delimitation of their habitat types; (3) detection of correlations between vegetation units and habitat types. Natural plant communities thus serve as indicators of abiotic habitat conditions. Reconstruction mapping is based on the extrapolation of the potential distribution of individual vegetation units to sites of similar habitat types where the natural vegetation does not exist any more. The same procedure is used for mapping the potential natural vegetation. Both types of natural vegetation maps are identical on sites where the abiotic natural habitat conditions (relief, geological substratum, climate, water regime, soils) remain practically unchanged. On sites where the natural habitat conditions have been considerably changed by man, e.g. in areas with superficial coal mining (complete destruction of the landscape, removal of soil cover, creation of large slag heaps) or in towns, no natural (primary) vegetation exists. This causes difficulties in the hypothetical concept of the potential natural vegetation and its definition. In contrast, in such sites reconstruction vegetation mapping uses the extrapolation of mapping units of the primary vegetation to the original natural habitat conditions. 相似文献
