This study examined whether increased K supply in conjunction with BAPcould increase lupin seed yield and harvest index by enlarging sink volume (podnumber), increasing assimilate and improving assimilate partitioning to filltheadditional pods induced by BAP treatment. Narrow-leafed lupin(Lupinusangustifolius, cv. Danja abs mutant) was grown inaglasshouse, in pots containing sandy soil with four K treatments (0, 15, 60 and120 mg K/kg soil). BAP (2 mM) was applied daily toallmain stem flowers throughout the life of each flower from opening to senesced.BAP application did not affect assimilate production (as measured by totalabove-ground biomass), but changed assimilate partitioning. On BAP-treatedplants, there were greater proportions of seed to pod wall dry weight on themain stem but smaller proportions on the branches, and an increased weightratioof seed to pod wall overall which meant more assimilate was used for seedgrowthrather than pod wall growth. BAP increased the number of pods per plant by35% and this more than compensated for the decreases in seeds per podandseed weight. Therefore, there was an increased harvest index (+11%)and seed yield per plant (+13%) in BAP-treated plants. BAP alsoincreased the number of pods with filled seeds (146%) on the main stemand main stem seed K+ concentration (from 0.81% to0.87%). Added K increased biomass but only slightly affected assimilatepartitioning. As applied K increased, relatively more assimilate was used forpod wall growth rather than seed growth. Added K increased seed yield per plantby about 14% due to increases in seed weight and the number of pods onthe main stem. Moreover, K+ concentration in seeds and shootsincreased with increasing level of applied K. Seed yield was enhanced more byBAP when K was supplied at high levels. Increasing K supply interactedpositively with added BAP by increasing narrow-leaf lupin seed yield andharvestindex through increases in assimilate supply and its partitioning into seeds.  相似文献

The gibberellin (GA) economy of young pea (Pisum sativum L.) fruits was investigated using a range of mutants with altered GA biosynthesis or deactivation. The synthesis mutation lh-2 substantially reduced the content of both GA4 and GA1 in young seeds. Among the other synthesis mutations, ls-1, le-1 and le-3, the largest reduction in seed GA1 content was only 1.7-fold (le-1), while GA4 was not reduced in these mutants, and in fact accumulated in some experiments (compared with the wild type). Mutation sln appeared to block the step GA20 to GA29 in young pods and seeds, but not as strongly as in older seeds. Mutations ls-1, le-1 and le-3 markedly reduced pod GA1 levels, but pod elongation was not affected. After feeds of [13C,3H]GA20 to leaves, the pods contained 13C,3H-labelled GA20, GA1, GA29 and GA81, and the seeds, [13C,3H]GA20 and [13C,3H]GA29. Summary The response of groundnut cotyledons to the presence of various growth regulators in concentrations from 0.1 to 5 mg/l has been studied in detail using several genotypes of groundnut on two different media. Cotyledons with embryo axis, cultured on Blaydes' medium with cytokinins, produced shoots, in the axils of which 2–7 flower buds could be seen. The frequency of flower bud induction in general increased with increasing concentrations of cytokinins, the optimal levels being 3 mg/l of KN or 4 mg/l of BAP. Cotyledons without embryo axis, cultured on Blaydes' medium with BAP (0.5 mg/l), produced a cluster of flower buds directly, ranging in number from 8–28, without any vegetative growth. Excised embryo axes cultured on the same medium gave plantlets without flower buds. The growth regulators IAA, NAA, GA3 and ABA failed to induce flower buds in independent treatments. However, lower concentrations of IAA and NAA in combination with cytokinins exerted a positive influence on flowering. The blooming of the flower buds was facilitated on media supplemented with low concentrations of cytokinins. Six percent of the induced flowers resulted in gynophore development and ultimately formed pods when cultured under complete dark conditions in modified MS medium supplemented with kinetin.  相似文献

Summary Experiments were conducted to determine if changes in the accumulation and partitioning of dry matter (DM) and nitrogen (N) in soybean [Glycine max (L.) Merr.] were associated with agronomic improvements and to assess the degree of genetic variation present for these traits. Fifteen maturity group II soybean genotypes including three ancestral cultivars, three modern cultivars, and nine agronomically superior plant introductions (PI's) were grown in replicated tests at four locations in the eastern U.S. The DM and N of stems, pod walls, and seeds were determined at maturity, and the apparent harvest indices (HI) and the apparent nitrogen harvest indices (NHI) were calculated. Pod DM partitioning was calculated as the ratio of seed DM to total pod DM and pod N partitioning was the ratio of seed N to total pod N. The mean DM accumulation of the modern cultivars was significantly greater than that of the ancestral cultivars and PI's. The apparent HI and the pod DM partitioning of both the modern and ancestral cultivars were significantly higher than that of the PI's. The three modern cultivars demonstrated the highest N accumulation. As a group, the modern cultivars consistently showed maximal accumulation and partitioning of DM and N suggesting that these physiological traits are associated with agronomic improvement. No individual PI was found to possess DM or N accumulation or partitioning which significantly exceeded the best modern cultivar or ancestral cultivar, indicating that genotypes with accumulation or partitioning characteristics which exceed available germplasm may be difficult to identify. Seed yield was correlated (P<0.05) with both DM (r=0.61) and N (r=0.57) accumulation.  相似文献

Glomus mosseae and the two pod rot pathogens Fusarium solani and Rhizoctonia solani and subsequent effects on growth and yield of peanut (Arachis hypogaea L.) plants were investigated in a greenhouse over a 5-month period. At plant maturity, inoculation with F. solani and/or R. solani significantly reduced shoot and root dry weights, pegs and pod number and seed weight of peanut plants. In contrast, the growth response and biomass of peanut plants inoculated with G. mosseae was significantly higher than that of non-mycorrhizal plants, both in the presence and absence of the pathogens. Plants inoculated with G. mosseae had a lower incidence of root rot, decayed pods, and death than non-mycorrhizal ones. The pathogens either alone or in combination reduced root colonization by the mycorrhizal fungus. Propagule numbers of each pathogen isolated from pod shell, seed, carpophore, lower stem and root were significantly lower in mycorrhizal plants than in the non-mycorrhizal plants. Pod storage protein (PSP) accumulated in developing pods of French bean (Phaseolus vulgaris L.) plants, and increasing the PSP mRNA level by pod removal resulted in the enhancement of PSP accumulation in pods that formed later. Pod storage protein was detected in flowers, young leaves and young stem internodes in addition to pods. Accumulation of PSP and its mRNA was induced by sink-removal in an organ-specific manner. In addition, wounding induced PSP accumulation systemically in leaves. Methyl jasmonate did not induce PSP synthesis but enhanced the synthesis that was induced by wounding. In senescing pods, PSP was degraded, and degradation products with molecular masses of 20 and 17 kDa were detected in the pods. The auxin 4-chlorophenoxyacetic acid (4-CPA) has been found to delay the maturation and dehiscence of the pods of oil seed rape. Dipping pods in solutions of 500 gl–1, 4-CPA, 36 and 39 days after anthesis delayed significantly the autolysis of the thin-walled parenchyma of the dehiscence zones, the senescence of the pod walls and the loss of water from the pod walls and seeds. Increases in both the fresh and dry weights of both pod walls and seeds were recorded in the treated pods as compared with controls.  相似文献

To evaluate the molecular mechanisms during pod and seed formation in legumes, starting with the development of reproductive organs, we constructed two cDNA libraries from developing flowers (MtFLOW) and pods including seeds (MtPOSE) of the model plant Medicago truncatula Gaertner. A total of 2,516 expressed sequence tags (ESTs) clustered into 1,776 nonredundant sequences (2k-set), which were annotated and assigned to functional classes. While about 30% of the ESTs encoded proteins of yet unknown function, typical annotations pointed to seed storage proteins, LTPs and lipoxygenases. The 2k-set was used to upgrade Mt6k-RIT microarrays (Küster et al. in J Biotechnol 108: 95, 2004) to Mt8k versions representing approximately 6,300 nonredundant M. truncatula genes. These were used to perform time course expression profiling studies based on hybridizations of samples that covered eight different developmental stages from flower buds to almost mature pods versus leaves as a common reference. About 180 up- and 70 downregulated genes were typically found for each stage and in total, 782 genes were either twofold up- or downregulated in at least one of the eight stages investigated. Based on this set, a combination of self-organizing map and hierarchical clustering revealed genes displaying expression regulation during characteristic stages of M. truncatula flower and pod development.
