The aim of this work was to study the effects of colchicine application on chromosome doubling and androgenic response in
anther and microspore culture of different bread wheat genotypes. Colchicine was applied during a mannitol stress pretreatment
or during the first 48 h of culture at concentrations of 0, 150 and 300 mg l−1. When colchicine was applied during stress pretreatment, the percentage of doubling depended on genotype and concentration.
A significant increase in doubling was observed with 300 mg l−1 in the low androgenic responding cv. Caramba. Colchicine incorporation during the first hours of culture improved percentage
of doubling in all genotypes, in both anther and microspore culture. Application of 300 mg l−1 colchicine improved the percentage of doubling in the two low responding genotypes, to 118% of control in DH24033, and 75%
in Caramba in microspore and anther culture, respectively. Concerning the androgenic response, the effect of colchicine on
embryo formation and percentage of green plants depended on the genotype and on the culture method. In cv. Pavon, a 2- and
a 3-fold increase in percentage of embryogenesis and green plants, respectively, were obtained with 300 mg l−1 colchicine in microspore culture. However, no significant differences in these two variables were observed in anther culture.
The number of green doubled haploid (DH) plants reflects the index of success of the procedure. Regardless of the culture
method, when colchicine was incorporated during the first hours of culture, the number of green DH plants increased significantly
in three of four genotypes. These results confirm the usefulness of colchicine application during the first hours of culture

Arundo micrantha Lam., a reed species from southern Mediterranean area, has found in Sardinia. Its presence represents the first record for Italy and the third one for western Europe. Investigations on distribution and ecology of the Sardinian population have been carried out, with special focusing on synecology of the species. A. micrantha has found in 40 localities mainly distributed along temporary streams and permanent rivers in the central and southern part of the island. Most of Sardinian populations occurs on alluvial soils in Thermomediterranean bioclimatic context. The autonomy of Sardinian phytocoenoses was characterized with the definition of exclusive floristic taxa, biological and chorological elements, and syndinamic relationships with other assemblages. Comparing the community with other reed beds from the Mediterranean basin, a marked floristic differentiation arose between the phytocoenoses characterized by autochthonous Arundo species and those dominated by the invasive Arundo donax. Finally, management activities are proposed to ensure the conservation of this threatened autochthonous plant community in freshwater habitat. 相似文献

A comparative study is presented of the pollen emissions of Urticaceae, Plantaginaceae and Poaceae, collected during 1995
with Hirst samplers (Burkard or Lanzoni) at five sites in western Spain: two Mediterranean sites located in the south (Huelva
and Seville) and three Atlantic sites in the north (Orense, Vigo and Santiago). The annual pollen of Poaceae and Plantaginaceae
collected in the Atlantic cities was found to be twice that in the Mediterranean sites, and the total amount of Urticaceae
was higher at sites with an urban environment and subject to sea influence (Vigo, Huelva and Seville). At all the sites, the
start of the main pollination periods (MPP) took place in the following order: Urticaceae, Plantaginaceae and Poaceae. It
was also observed that the MPP of these three pollen types began earlier in Huelva and Seville, where the mean temperatures
necessary for the beginning of pollen emissions are recorded very early. Regarding the variation in pollen concentrations
throughout the year, Urticaceae presented peaks of maximum concentration in March (Huelva, Seville, Vigo and Orense) and June
(Santiago); Plantaginaceae in March (south) and June (north); and Poaceae in May (south) and June–July (north). At northern
sites, pollen emissions of Urticaceae and Plantaginaceae continued throughout the summer, while in the south they decreased
considerably from May onwards. From the allergenic point of view, the indices of reactivity described for Urticaceae and Poaceae
were exceeded more often at northern sites, in particular at Vigo. The meteorological conditions associated with periods of
highest pollen emission of these three herbaceous types are a rise in mean temperature, light or absent rainfall, and abundant
sunshine. The statistical correlations between pollen emissions and meteorological factors were not well-defined, either for
the stations or for all the taxa, although they were clearer for the Atlantic cities and for Urticaceae. 相似文献

Genetic mapping identifies a rice naringenin O-glucosyltransferase that influences insect resistance

Naringenin, the biochemical precursor for predominant flavonoids in grasses, provides protection against UV damage, pathogen infection and insect feeding. To identify previously unknown loci influencing naringenin accumulation in rice (Oryza sativa), recombinant inbred lines derived from the Nipponbare and IR64 cultivars were used to map a quantitative trait locus (QTL) for naringenin abundance to a region of 50 genes on rice chromosome 7. Examination of candidate genes in the QTL confidence interval identified four predicted uridine diphosphate-dependent glucosyltransferases (Os07g31960, Os07g32010, Os07g32020 and Os07g32060). In vitro assays demonstrated that one of these genes, Os07g32020 (UGT707A3), encodes a glucosyltransferase that converts naringenin and uridine diphosphate-glucose to naringenin-7-O-β-d -glucoside. The function of Os07g32020 was verified with CRISPR/Cas9 mutant lines, which accumulated more naringenin and less naringenin-7-O-β-d -glucoside and apigenin-7-O-β-d -glucoside than wild-type Nipponbare. Expression of Os12g13800, which encodes a naringenin 7-O-methyltransferase that produces sakuranetin, was elevated in the mutant lines after treatment with methyl jasmonate and insect pests, Spodoptera litura (cotton leafworm), Oxya hyla intricata (rice grasshopper) and Nilaparvata lugens (brown planthopper), leading to a higher accumulation of sakuranetin. Feeding damage from O. hyla intricata and N. lugens was reduced on the Os07g32020 mutant lines relative to Nipponbare. Modification of the Os07g32020 gene could be used to increase the production of naringenin and sakuranetin rice flavonoids in a more targeted manner. The majority of angiosperms produce hermaphrodite flowers, while a lesser number (20–30%) produce unisexual flowers. Little is known about the molecular biology of sex-determination in angiosperms, however, a few sex-determining genes have been cloned from the model system Zea mays. One of these genes is Tasselseed2 (Ts2) which has been shown to be involved in the arrest of developing pistils in male flowers. In this study, we sequenced a putative homologue of Ts2 in species of Bouteloua, a genus in the grass subfamily Chloridoideae. We found significant genetic variation at Ts2 in Bouteloua relative to other developmental genes characterized in maize and other grass species. We also found that in Bouteluoua, Ts2 is evolving non-neutrally in the hermaphrodite-flowered Bouteloua hirsuta while no difference from neutral expectation was detected at Ts2 in the monoecious/dioecious Bouteloua dimorpha. The putatively neutral gene Alcohol Dehydrogenase1 (Adh1) was also examined for the same species of Bouteloua, and no departure from neutral expectation was detected. Our results suggest that purifying selection may be acting on Ts2 in the hermaphrodite-flowered B. hirsuta while no evidence of selection was detected at Ts2 in the monoecious/dioecious B. dimorpha. 相似文献

Many asexual animal populations comprise a mixture of genetically different lineages, but to what degree this genetic diversity leads to ecological differences remains often unknown. Here, we test whether genetically different clonal lineages of Aptinothrips grass thrips differ in performance on a range of plants used as hosts in natural populations. We find a clear clone‐by‐plant species interactive effect on reproductive output, meaning that clonal lineages perform differently on different plant species and thus are characterized by disparate ecological niches. This implies that local clonal diversities can be driven and maintained by frequency‐dependent selection and that resource heterogeneity can generate diverse clone assemblies. 相似文献
