云南水稻地方品种月亮谷的群体多样性分析 总被引:2,自引:0,他引:2
月亮谷是云南元阳梯田种植历史悠久、种植面积最大的优良水稻地方品种之一,当地少数民族具有引种或换种的稻作习惯。为揭示这种稻作习惯对月亮谷群体遗传多样性的影响,本研究对该品种群体内和群体间进行了遗传多样性的比较和分析,目的是为更好地了解月亮谷的群体遗传结构,为持久利用地方品种提供理论依据。首先采用分层随机取样的策略从元阳梯田不同海拔获得24个原位栽培群体,采用形态指数分类法和抗病性测定对24个群体共720个单株样品的月亮谷进行形态学分类和稻瘟病抗性鉴定,并分析了这些单株材料在48个SSR位点的遗传多样性。研究结果表明,形态上月亮谷属于栽培稻的籼稻类型,其群体对稻瘟病具中抗水平,但无论是在群体内还是群体间,均普遍表现出明显差异,说明不同来源的月亮谷存在抗病功能表型上的变异;遗传多样性分析显示,48对SSR引物共检测出91个多态位点,多态性位点百分率为77.08%,Nei多样性指数平均值为0.064,变幅为0~0.4302。24个群体之间的遗传相似系数在0.9753~0.9866之间,群体内个体之间的遗传相似系数在0.86~1.00之间;AMOVA分析显示,以地理村寨作为自然居群单位,居群间的变异为3.36%,居群内群体间的变异为33.15%,居群内的变异为63.49%;聚类分析显示,村寨群体间的遗传多样性与村寨间的地理空间距离有一定相关性。 相似文献

Rice ( Oryza sativa L. cv. Sasanishiki) coleoptiles grown under water achieved greater length than those grown either in air or under water with constant air bubbling. The extensibility of cell walls in coleoptiles grown under water was larger than that in the other treatments. Per unit length of the coleoptile, the content of ferulic and diferulic acids ester-linked to hemicelluloses was higher in air and bubbling type coleoptiles than in water type ones. The extensibility of the coleoptile cell walls correlated with the content of diferulic acids per unit length and per hemicellulose, suggesting that the enhancement of the formation of diferulic acid bridges in hemicelluloses in air or under water with air bubbling makes the cell walls mechanically rigid; thereby inhibiting cell elongation in rice coleoptiles. In addition, the ratio of diferulic acid to ferulic acid was almost constant irrespective of coleoptile age, zone and growth conditions, suggesting that the feruloylation of hemicelluloses is rate-limiting in the formation of diferulic acid bridges in the cell walls of rice coleoptiles. 相似文献

Background and Aims
The effect of environmental factors on the regulation of aerenchyma formation in rice roots has been discussed for a long time, because aerenchyma is constitutively formed under aerated conditions. To elucidate this problem, a unique method has been developed that enables sensitive detection of differences in the development of aerenchyma under two different environmental conditions. The method is tested to determine whether aerenchyma development in rice roots is affected by osmotic stress.Methods
To examine aerenchyma formation both with and without mannitol treatment in the same root, germinating rice (Oryza sativa) caryopses were sandwiched between two agar slabs, one of which contained 270 mm of mannitol. The roots were grown touching both slabs and were thereby exposed unilaterally to osmotic stress. As a non-invasive approach, refraction contrast X-ray computed tomography (CT) using a third-generation synchrotron facility, SPring-8 (Super photon ring 8 GeV, Japan Synchrotron Radiation Research Institute), was used to visualize the three-dimensional (3-D) intact structure of aerenchyma and its formation in situ in rice roots. The effects of unilateral mannitol treatment on the development of aerenchyma were quantitatively examined using conventional light microscopy.Key Results
Structural continuity of aerenchyma was clearly visualized in 3-D in the primary root of rice and in situ using X-ray CT. Light microscopy and X-ray CT showed that the development of aerenchyma was promoted on the mannitol-treated side of the root. Detailed light microscopic analysis of cross-sections cut along the root axis from the tip to the basal region demonstrated that aerenchyma developed significantly closer to the root tip on the mannitol-treated side of the root.Conclusions
Continuity of the aerenchyma along the rice root axis was morphologically demonstrated using X-ray CT. By using this ‘sandwich’ method it was shown that mannitol promoted aerenchyma formation in the primary roots of rice. 相似文献5.

The effects of aluminum on lipid peroxidation and activities of antioxidative enzymes were investigated in detached rice leaves treated with 0 to 5 mM AlCl3 at pH 4.0 in the light. AlCl3 enhanced the content of malondialdehyde but not the content of H2O2. Superoxide dismutase activity was reduced by AlCl3, while catalase and glutathione reductase activities were increased. Peroxidase and ascorbate peroxidase activities were increased only after prolonged treatment, when toxicity occurred. The results give evidence that Al treatment caused oxidative stress and in turn, it caused lipid peroxidation. 相似文献

The aim of this work was to develop a photoperiod-sensitive male sterile rice with stable sterility. We developed Changguang S, an indica rice strain, by using a short critical day length. Differences in the fertility responses of Changguang S strain pollen to temperature and photoperiod under natural and controlled conditions were studied. The results showed that Changguang S strain exhibited stable sterility under long-day and low-temperature conditions (22°C, 15 days). The stability of sterility was significantly higher than that of other such rice strains, Nongken 58S and 7001S. The critical photoperiod for inducing male sterility in Changguang S was 13 h or shorter, and its duration was significantly shorter than that required for rice strains Nongken 58S and 7001S. It is suggested that Changguang S is a typical photoperiod-sensitive male sterile rice strain with a shorter critical day length and a lower critical temperature. It is promising to apply this strain to two-line hybrid rice production. 相似文献

This research was conducted to assess heavy metal contamination in the environment and within Oryza sativa. The translocation factors (TFs) and bioaccumulation factors (BAFs) for heavy metals in O. sativa and estimated daily intake (EDI) and health risk index (HRI) were measured. The samples were analyzed for heavy metals using inductively coupled plasma optical emission spectrometry (ICP-OES). Pb and Cr concentrations in water samples within and near the electronic-waste dumping area exceeded water quality standards for surface water sources from the Pollution Control Department in the Ministry of Natural Resources and Environment of Thailand (PCD). The Pb concentration in soil samples within the area also exceeded soil quality standards for habitat and agriculture from PCD. Most of the metals were highly concentrated in roots, except for Mn which has the highest concentration in leaves. Pb concentrations in rice grains exceeded the FAO/WHO standard (0.2 mg/kg). The average TF values for heavy metals from the soil to roots, roots to stems, stems to leaves, and stems to grains were Mn > Pb > Ni > Cr, Mn > Cr > Ni > Pb, Ni > Pb > Mn > Cr, and Pb > Ni > Cr > Mn, respectively. The average BAF values in O. sativa were Mn > Ni > Pb > Cr. The EDI for Cr, Pb, Mn, and Ni via O. sativa consumption were 6.19, 6.02, 370.57, and 3.80 µg/kg/day, respectively. The HRI for Cr, Pb, Mn, and Ni via O. sativa consumption were 0.30, 1.50, 2.60, and 0.002, respectively. 相似文献

Data on root parameters and distribution are important for an improved understanding of the factors influencing nutrient uptake
by a crop. Therefore, a study was conducted on a Crowley silt loam at the Rice Research and Extension Center near Stuttgart,
Arkansas to measure root growth and N, P and K uptake by three rice (Oryza sativa L.) cultivars at active tillering (36 days after emergence (DAE)), maximum tillering (41 DAE), 1.25 cm internode elongation
(55 DAE), booting (77 DAE) and heading (88 DAE). Soil-root core samples were taken to a depth of 40 cm after plant samples
were removed, sectioned into 5 cm intervals, roots were washed from soil and root lengths, dry weights and radii were measured.
Root parameters were significantly affected by the soil depth × growth stage interaction. In addition, only root radius was
affected by cultivar. At the 0- to 5-cm soil depth, root length density ranged from 38 to 93 cm cm-3 throughout the growing season and decreased with depth to about 2 cm cm-3 in the 35- to 40-cm depth increment. The increase in root length measured with each succeeding growth stage in each soil
horizon also resulted in increased root surface area, hence providing more exposed area for nutrient uptake. About 90% of
the total root length was found in the 0- to 20-cm soil depth throughout the season. Average root radius measured in the 0-
to 5-cm and 35- to 40-cm depth increments ranged from 0.012 to 0.013 cm and 0.004 to 0.005 cm, respectively throughout the
season. Total nutrient uptake by rice differed among cultivars only during vegetative growth. Differences in total nutrient
uptake among the cultivars in the field appear to be related to absorption kinetics of the cultivars measured in a growth
chamber study.

β-Amylase deficiency in various cultivars of rice was examined at the molecular level. Using an antibody against β-amylase
purified from germinating seeds of rice, we were able to demonstrate the expression and organization of the β-amylase gene
in normal and deficient cultivars. Although β-amylase is a starch-hydrolyzing enzyme, as is α-amylase, the β-amylase protein/gene
is expressed differently from the α-amylase protein/gene; i.e. (1) β-amylase is synthesized only in aleurone cells, (2) the
enzyme production in the embryo-less half-seeds is not under hormonal control. We identified some cultivars of rice that are
deficient for β-amylase activity. We present new evidence that synthesis is blocked at the level of mRNA synthesis in the
deficient cultivars. The usefulness of β-amylase as a crop trait is also discussed.
