This study characterised the permeability of the salmonid posterior intestine in vivo, to two hydrophilic markers of different molecular weight, both in the presence and absence of sodium deoxycholate (SDA), and determined the influence of mucosal secretions. The posterior intestine of chinook salmon was cannulated with a balloon catheter and the lumen infused with a solution of fluorescein and 14C-mannitol. In treated fish, the solution also contained 5.0 mmol · l−1 SDA. Blood samples from the dorsal aorta were taken at regular time intervals over 3 h. Clearances and volumes of distribution were assessed by intravenous administration of the markers to another group of fish. In the absence of SDA, low permeabilities were recorded for both markers; however, permeabilities for both were significantly greater in the treated groups. Both solutes had volumes of distribution similar to values reported elsewhere. Metabolism of fluorescein by the liver resulted in its plasma clearance. In contrast, elimination of mannitol was negligible during the study period, probably due to the lowered glomerular filtration rates observed in sea water adapted fish. Compared to in vitro investigations, in vivo mucus secretions were significantly lower and solute delivery across the epithelium was higher. In the present study, we addressed the question of whether treatment with mannitol, an osmotic diuretic, affects astrogliovascular responses to status epilepticus (SE). In saline-treated animals, astrocytes exhibited reactive astrogliosis in the CA1-3 regions 2-4 days after SE. In the mannitol-treated animals, a large astroglial empty zone was observed in the CA1 region 2 days after SE. This astroglial loss was unrelated to vasogenic edema formation. There was no difference in SE-induced neuronal loss between saline- and mannitol-treated animals. Furthermore, mannitol treatment did not affect astroglial loss and vasogenic edema formation in the dentate gyrus and the piriform cortex. These findings suggest that mannitol treatment induces selective astroglial loss in the CA1 region independent of vasogenic edema formation following SE. These findings support the hypothesis that the susceptibility of astrocytes to SE is most likely due to the distinctive heterogeneity of astrocytes independent of hemodynamics. 目的:比较等渗透剂量的7.5%高渗盐水(hypertonic saline,HTS)和20%甘露醇治疗颅高压的疗效。方法:当患者的颅内压(intracranial pressure,ICP)超过20 mm Hg时,一组患者接受了4 m L/kg 7.5%HTS(HTS组,n=27),另外一组患者接受了0.5 g/kg 20%甘露醇(甘露醇组,n=31)的降颅压治疗,两种药物均经深静脉在30 min内快速滴注完成。用药期间,连续监测患者ICP,平均动脉压(mean arterial pressure,MAP)、脑灌注压(cerebral perfusion pressure,CPP)及中心静脉压(central venous pressure,CVP)。记录有效降颅压持续时间、ICP最大降幅及其时间,用药前及用药后1 h、3 h、6 h抽血查电解质和血浆渗透压等。结果:静脉快速滴注7.5%HTS和20%甘露醇后,两者均可有效降低ICP(P0.05)。两组在控制颅高压的有效率、起效时间及ICP降幅无统计学差异(P0.05),但HTS组的作用持续时间要明显长于甘露醇组(P0.05)。两组渗透压较用药前相比均有显著上升(P0.05)。用药前后,两组MAP、CPP和CVP变化无统计学差异(P0.05)。结论:等渗透剂量的7.5%HTS与20%甘露醇均可有效降低患者的ICP,两者降颅压效果相当,均可作为治疗颅内高压的一线治疗药物。  相似文献

Seaweeds emerge as promising third-generation renewable for sustainable bioproduction. In the present work, we valorized brown seaweed to produce l-lysine, the world's leading feed amino acid, using Corynebacterium glutamicum, which was streamlined by systems metabolic engineering. The mutant C. glutamicum SEA-1 served as a starting point for development because it produced small amounts of l-lysine from mannitol, a major seaweed sugar, because of the deletion of its arabitol repressor AtlR and its engineered l-lysine pathway. Starting from SEA-1, we systematically optimized the microbe to redirect excess NADH, formed on the sugar alcohol, towards NADPH, required for l-lysine synthesis. The mannitol dehydrogenase variant MtlD D75A, inspired by 3D protein homology modelling, partly generated NADPH during the oxidation of mannitol to fructose, leading to a 70% increased l-lysine yield in strain SEA-2C. Several rounds of strain engineering further increased NADPH supply and l-lysine production. The best strain, SEA-7, overexpressed the membrane-bound transhydrogenase pntAB together with codon-optimized gapN, encoding NADPH-dependent glyceraldehyde 3-phosphate dehydrogenase, and mak, encoding fructokinase. In a fed-batch process, SEA-7 produced 76 g L−1 l-lysine from mannitol at a yield of 0.26 mol mol−1 and a maximum productivity of 2.1 g L−1 h−1. Finally, SEA-7 was integrated into seaweed valorization cascades. Aqua-cultured Laminaria digitata, a major seaweed for commercial alginate, was extracted and hydrolyzed enzymatically, followed by recovery and clean-up of pure alginate gum. The residual sugar-based mixture was converted to l-lysine at a yield of 0.27 C-mol C-mol−1 using SEA-7. Second, stems of the wild-harvested seaweed Durvillaea antarctica, obtained as waste during commercial processing of the blades for human consumption, were extracted using acid treatment. Fermentation of the hydrolysate using SEA-7 provided l-lysine at a yield of 0.40 C-mol C-mol−1. Our findings enable improvement of the efficiency of seaweed biorefineries using tailor-made C. glutamicum strains.  相似文献

In a continued chemosystematic investigation of the water-soluble compounds in Veronica sect. Hebe, four additional species were investigated. In comparison to other, Northern Hemisphere (NH) species of Veronica, those belonging to the New Zealand species in sect. Hebe are apparently more variable in chemical content. In addition to the compounds characteristic for NH Veronica, namely mannitol, aucubin, catalpol and 6-O-esters of catalpol as well as some caffeoyl phenylethanoid glucosides (CGPs), Veronica topiaria (syn. Hebe topiaria) also gave an unusual 6-O-ester of aucubin named topiarioside. The former Hebe species Veronica cupressoides and Veronica stenophylla each provided one of the two previously undescribed disaccharide esters named hebitol I and II, respectively, and the former plant also provided a CPG named cuproside, a 6-O-β-glucopyranosyl derivative of the known hebeoside. The last species, namely Veronica hulkeana (syn. Heliohebe hulkeana) only contained compounds common to other species of Veronica. The taxonomic results are discussed and it is concluded that carbohydrate esters are common in sect. Hebe. The data so far obtained indicate that the occurrences of esters of 6-O-rhamnopyranosylcatalpol are confined to the most derived species in the section.  相似文献

Supplementation with CaCl2·2H2O (50 mg l−1) or CuSO4·5H2O (10 mg l−1) improved mannitol production by Candida magnoliae by 14.5 and 18.6% (25 and 32 g/L), respectively. When used in combination, they acted synergistically: Ca2+ decreased the intracellular concentration of mannitol 30%, whereas Cu2+ increased the intracellular activity of mannitol dehydrogenase 1.6-times more than control. Ca2+ probably works by altering the permeability of cells to mannitol, whereas, Cu2+ increases the activity of an enzyme responsible for mannitol biosynthesis.  相似文献

The hydroxyl radical (OH) has detrimental biological activity due to its very high reactivity. Our experiments were designed to determine the effects of equimolar concentrations of glucose, fructose and mannitol and three phosphorylated forms of fructose (fructose-1-phosphate (F1P); fructose-6-phosphate (F6P); and fructose-1,6-bis(phosphate) (F16BP)) on OH radical production via the Fenton reaction. EPR spectroscopy using spin-trap DEPMPO was applied to detect radical production. We found that the percentage inhibition of OH radical formation decreased in the order F16BP > F1P > F6P > fructose > mannitol = glucose. As ketoses can sequester redox-active iron thus preventing the Fenton reaction, the Haber-Weiss-like system was also employed to generate OH, so that the effect of iron sequestration could be distinguished from direct OH radical scavenging. In the latter system, the rank order of OH scavenging activity was F16BP > F1P > F6P > fructose = mannitol = glucose. Our results clearly demonstrate that intracellular phosphorylated forms of fructose have more scavenging properties than fructose or glucose, leading us to conclude that the acute administration of fructose could overcome the body’s reaction to exogenous antioxidants during appropriate therapy in certain pathophysiological conditions related to oxidative stress, such as sepsis, neurodegenerative diseases, atherosclerosis, malignancy, and some complications of pregnancy.  相似文献

Oxovanadium(IV) complexes of the polyalcohols sorbitol, galactitol, and mannitol, of stoichiometry Na(2)[VO(L)(2)].H(2)O, were obtained from aqueous alkaline solutions. They were characterized by elemental analysis, infrared and UV-vis spectroscopies, thermoanalytical (thermogravimetric and differential thermal analysis) data, and magnetic susceptibility measurements. The biological activities of the complexes on the proliferation, differentiation, and glucose consumption were tested on osteoblast-like cells (MC3T3E1 osteoblastic mouse calvaria-derived cells and UMR106 rat osteosarcoma-derived cells) in culture. The three complexes exerted a biphasic effect on cell proliferation, being slight stimulating agents at low concentrations and inhibitory in the range of 25-100 microM. All the complexes inhibited cell differentiation in tumor osteoblasts. Their effects on glucose consumption were also discussed. The free ligands did not show any effect on the studied biological parameters.  相似文献

We developed a capillary column gas chromatography (CCGC) method for the measurement of urinary sucralose (S) and three other sugar probes including, sucrose, lactulose (L) and mannitol (M) for use in in vivo studies of intestinal permeability. We compared the capillary method with a packed column gas chromatography (PCGC) method. We also investigated a possible role for sucralose as a probe for the measurement of whole gut permeability. Sample preparation was rapid and simple. The above four sugars were detected precisely, without interference. We measured intestinal permeability using 5- and 24-h urine collections in 14 healthy volunteers. The metabolism of sugars was evaluated by incubating the intestinal bacteria with an iso-osmolar mixture of mannitol, lactulose and sucralose at 37 degrees C for 19 h. Sugar concentrations and the pH of the mixture were monitored. The use of the CCGC method improved the detection of sucralose as compared to PCGC. The average coefficient of variation decreased from 15% to 4%. It also increased the sensitivity of detection by 200-2000-fold. The GC assay was linear between sucralose concentrations of 0.2 and 40 g/l (r=1.000). Intestinal bacteria metabolized lactulose and acidified the media but did not metabolize sucralose or mannitol. The new method for the measurement of urinary sucralose permits the simultaneous quantitation of sucrose, mannitol and lactulose, and is rapid, simple, sensitive, accurate and reproducible. Because neither S nor M is metabolized by intestinal bacteria, and because only a tiny fraction of either sugar is absorbed, this pair of sugar probes appears to be available for absorption throughout the GI tract. Thus, the 24-h urinary concentrations of S and M, or the urinary S/M ratio following an oral dose of a sugar mixture, might be good markers for whole gut permeability.  相似文献
