To elucidate the role of the spiral limbus in glucose transport in the cochlea, we analyzed the expression and localization of GLUT1, connexin26, connexin30, and occludin in the spiral limbus of the rat cochlea. GLUT1 and occludin were detected in blood vessels. GLUT1, connexin26, connexin30, and occludin were also expressed in fibrocytes just basal to the supralimbal lining cells. Connexin26 and connexin30 were present among not only these GLUT1-positive fibrocytes but also GLUT1-negative fibrocytes. In vivo glucose imaging using 6-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-6-deoxyglucose (6-NBDG, MW 342) together with Evans Blue Albumin (EBA, MW 68,000) showed that 6-NBDG was rapidly distributed throughout the spiral limbus, whereas EBA was localized only in the vessels. Moreover, the gap junctional uncoupler heptanol inhibited the distribution of 6-NBDG. These findings suggest that gap junctions play an important role in glucose transport in the spiral limbus, i.e., that gap junctions mediate glucose transport from GLUT1-positive fibrocytes to GLUT1-negative fibrocytes in the spiral limbus.  相似文献

Four new species of hoplolaimoid nematodes (Merlinius adakensis, Pratylenchoides megalobatus, Pratylenchus pratensisobrinus, and Pratylenchus ventroprojectus) are described from Adak Island in the Aleutian chain. M. adakensis n. sp. is separated from other species by body length (0.96-1.3 mm), stylet length (32-36 μm), number of tail annules (49-68), and c'' (3.1-4.1). P. megalobatus n. sp. differs from all known Pratylenchoides spp. by having a very long esophageal gland lobe (b'' = 2.4-3.3, overlap 3-6 times the body width). P. pratensisobrinus n. sp. closely resembles P. pratensis (de Man) Filipjev, but has a longer stylet (15-17 μm), a longer tail (c = 12-15; c'' = 2.8-3.7), and more tail annules (23-37). P. ventroprojectus n. sp. is distinguished by body length (392-475 μm), three lip annules, low and flattened cephalic capsule, and presence of terminal subventral projection. Pratylenchoides variabilis Sher, Helicotylenchus amplius Anderson &Eveleigh, and H. spitsbergensis Loof are also reported from Adak and Amchitka Islands.  相似文献

A novel method of producing controlled vortices was used to reduce both concentration polarization and membrane fouling during microfiltration of Saccharomyces cerevisiae broth suspensions. The method involves flow around a curved channel at a sufficient rate so as to produce centrifugal instabilities (called Dean vortices). These vortices depolarize the build-up of suspended particles such as yeast cells at the membrane-solution interface and allow for increased membrane permeation rates. Various operating conditions under which such vortices effectively reduced cake build-up of suspended particles such as yeast cells at the membrane-solution interface and allow for increased membrane permeation rates. Various operating conditions under which such vortices effectively reduced cake build-up during microfiltration of 0 to 0.55 dry wt% yeast broth were investigated. Flux improvements of over 60% for 0.25 dry wt% yeast broth for flow with over that without Dean vortices were observed. This beneficial effect increased with increasing retentate flow rate and increasing transmembrane pressure and decreased with increasing concentration of suspended matter. Similar behavior was observed whether the cells were viable of killed. the improvement in flux in the presence over that in the absence of vortices correlated well with centrifugal force or azimuthal velocity squared. The relative cake resistances increased with reservoir yeast concentration. These values with vortices increased from 62% to 75% of that without vortices with increasing yeast concentration. The ratio of the cake thicknesses in the limiting case (at high feed concentration) was 3.25. These results suggest that self-cleaning spiral vortices could be effective in maintaining good and steady microfiltration performance with cell suspensions other than those tested. The consumption and utilization of various concentrations of the spiral blue-green algae, Spirulina platensis (Nordst.) Geitl., by the fourth-instar larvae of the silkworm, Bombyx mori L., was studied by incorporation into an artificial diet. The silkworms ingested more food on the low algal-powder diets, whereas they digested more efficiently on the high algal levels. The larvae fed on diets containing 30–40% algae had the shortest larval duration compared to those on low algal-powder diets. The freeze-dried powder of S. platensis is well utilized by B. mori larvae when added to artificial diets at levels between 15% and 30%, although the high algal content at 40% was slightly better for the efficiency of conversion of ingested diet into body substance (E.C.I.) and the cocoon production. However, analysis of the protein content in feces indicated an excess of the algal powder at 40%. Measurements of data on the food consumption and utilization in this study proposed that this spiral alga is dietetically utilizable as a protein source for rearing the silkworm in smaller quantities than on a soybean meal.
Resúmé La consommation et l'utilisation de différentes concentrations de Spirulina platensis par les chenilles de 4ème stade de Bombyx mori, ont été étudiées par incorporation dans un aliment artificiel. Les vers à soie ont ingéré plus d'aliment avec les régimes à faible concentration en algue, tandis qu'ils ont digéré plus efficacement les régimes à haute concentration d'algue. Les chenilles alimentées sur des régimes contenant 30 à 40% d'algue ont eu un développement larvaire plus bref que celles alimentées sur des régimes à faible concentration. La poudre congelée et séchée de S. platensis incorporée aux taux de 15 à 30% dans le régime artificiel est bien utilisée par les chenilles de B. mori, quoiqu'une teneur de 40% soit légèrement meilleure pour l'efficacité de conversion de l'aliment en matière vivante (E.C.I.) et la production de cocons. Cependant, l'analyse du contenu protéique dans les excréments révélait un excès de poudre d'algue à 40%. L'analyse des données au cours de cette étude, sur la consommation et l'utilisation des aliments, montre que S. platensis est utilisable diét étiquement en plus petite quantité que la farine de soja comme source de protéines pour l'élevage du ver à soie.

The neonatal heart completely regenerates after apical resection (AR), providing a desirable research model to study the mechanism of cardiac regeneration and cardiomyocyte proliferation. However, AR-induced neonatal heart regenerative phenomenon is controversial due to the variation of operative details in different laboratories. Here, we provide an optimized AR operation procedure with stable regeneration and high survival rate by achieving heart exposure, normalizing myocardium cut-offs, and reducing operation duration. We also established a whole-heart-slice approach to estimate the myocardial regeneration after the AR operation, which ensures no false-negative/positive results. The combination of the optimized AR operation and the whole-heart-slice analysis provides a stable system to study neonatal heart regeneration and cardiomyocyte proliferation in situ.  相似文献

目的:比较分析腹腔镜和开腹结肠癌根治术治疗老年局部进展期结肠癌的临床疗效和安全性及对患者免疫功能的影响。方法:根据随机数字表法,将64例老年局部进展期结肠癌患者随机分为腹腔镜组和开腹组,每组各32例,分别接受腹腔镜、开腹结肠癌根治术治疗。比较两组手术相关指标、手术前后免疫功能变化、术后近远期并发症的发生情况及预后。结果:与开腹组比较,腹腔镜组患者手术时间明显延长,而术中出血量、胃肠功能恢复时间则明显缩短(P<0.05)。两组淋巴结清扫数比较差异无统计学意义(P>0.05)。术后3个月,腹腔镜组CD4+、CD4+/CD8+比值均明显高于开腹组(P<0.05),且与术前比较差异均无统计学意义(P>0.05)。与开腹组比较,腹腔镜组患者术后切口感染的发生率明显降低(P<0.05),两组其他近期并发症如吻合口瘘、吻合口出血,远期并发症如黏连性肠梗阻、切口疝的发生率比较差异均无统计学意义(P>0.05)。腹腔镜组与开腹组术后2年的局部复发率、1年和2年生存率比较差异均无统计学意义(P>0.05)。结论:腹腔镜手术和开腹手术治疗老年局部进展期结肠癌患者的临床疗效和预后相当,但腹腔镜手术对患者的免疫功能影响更小,且安全性更高。  相似文献

目的:探讨单切口腹腔镜在结直肠手术中的应用效果及对炎症因子与应激反应的影响。方法:选择2018年1月至2019年10月我院接诊的98例结直肠癌患者,通过随机数表法分为2组,每组49例。观察组使用单切口腹腔镜结直肠癌根治术,对照组使用传统腹腔镜结直肠癌根治术,常规5孔操作法。比较两组围术期相关情况、手术前后血清炎症因子水平的变化、手术后应激反应及并发症的发生情况。结果:两组淋巴结清扫个数、中转开腹例数、术后排气时间比较差异均无统计学意义(P0.05),观察组手术时间明显长于对照组,脐切口长度、住院时间、术中出血量均显著短于或低于对照组;两组术后1 d、3 d、5 d时高敏C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、去甲肾上腺素(NE)、肾上腺素(E)、皮质醇(Cor)水平均明显高于术前(P0.05);且观察组术后1d、3d、5d时hs-CRP、TNF-α、IL-6、NE、E、Cor较对照组显著降低(P0.05);两组吻合口瘘、吻合口狭窄、吻合口出血、小肠穿孔、肠梗阻、感染的总发生率比较差异无统计学意义(P0.05)。结论:和传统5孔操作法相比,单切口腹腔镜结直肠癌根治术治疗结直肠癌患者的手术时间更长,但术后炎症因子表达更低,应激反应更小,有利于患者术后恢复。  相似文献

摘要 目的:探讨经皮穴位电刺激(TEAS)联合全身麻醉对腹腔镜结直肠癌根治术患者炎性因子、T细胞亚群和认知功能的影响。方法:选取2018年6月~2019年9月期间我院收治的行腹腔镜结直肠癌根治术患者90例,根据随机数字表法分为对照组(n=45)和研究组(n=45),对照组给予全身麻醉处理,研究组在对照组的基础上联合TEAS,比较两组患者炎性因子、T细胞亚群、认知功能及疼痛情况。结果:研究组术后1 d、术后5 d白介素-6(IL-6)、白介素-1(IL-1)和肿瘤坏死因子-α(TNF-α)低于对照组(P<0.05)。研究组术后1 d、术后5 d CD3+、CD4+/CD8+、CD4+高于对照组,CD8+则低于对照组(P<0.05)。研究组术后1 d、术后3 d、术后5 d、术后7 d简易智力状态检查表(MMSE)评分高于对照组(P<0.05)。研究组术后6 h、术后12 h、术后24 h、术后48 h视觉模拟评分法(VAS)评分低于对照组(P<0.05)。两组术后5 d、术后7 d认知功能障碍(POCD)发生率比较差异无统计学意义(P>0.05);研究组术后1 d、术后3dPOCD发生率低于对照组(P<0.05)。结论:TEAS联合全身麻醉治疗腹腔镜结直肠癌根治术患者,可降低炎性反应,减轻免疫抑制,同时还可降低对机体认知功能的损害。  相似文献

摘要 目的:探讨盐酸右美托咪定联合盐酸罗哌卡因胸椎旁神经阻滞(TPVB)对肺癌根治术患者血清炎性因子和免疫学指标的影响。方法:选取2018年4月~2020年4月期间于我院行肺癌根治术的患者400例,根据信封抽签法分为对照组和观察组,各200例,对照组给予盐酸罗哌卡因TPVB,观察组在对照组基础上联合盐酸右美托咪定,对比两组疼痛、炎性因子、免疫学指标、血流动力学及不良反应。结果:观察组术后6 h(T5)~术后48h(T8)时间点视觉模拟评分法(VAS)评分均低于对照组(P<0.05)。对照组插管后5 min(T1)~术毕(T3)时间点心率(HR)及平均动脉压(MAP)较麻醉诱导前(T0)时间点升高(P<0.013),观察组T1~T3时间点HR、MAP低于对照组(P<0.05)。两组T8时间点白介素-6(IL-6)、C反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)均较T0下降,且观察组低于对照组(P<0.05)。两组T8时间点CD4+、CD4+/CD8+均较T0时间点下降,但观察组高于对照组(P<0.05),CD8+均较T0时间点升高,但观察组低于对照组(P<0.05)。观察组不良反应总发生率低于对照组(P<0.05)。结论:盐酸右美托咪定联合盐酸罗哌卡因TPVB用于肺癌根治术患者,可稳定血流动力学,且可获得较好的镇痛效果,降低不良反应发生率,减轻术后炎性损伤及免疫抑制。  相似文献
