Abstract

The microbial cleavage of the side chain of cholesterol is a slow process due to the low solubility of the substrate in aqueous media (< 1 μM). Cell immobilization has been shown to be an efficient technology for enhancing the yield of cholesterol biotransformation. In these experiments, living cells of Mycobacterium sp. DSM 2966 and Mycobacterium sp. DSM 2967 were immobilized by passive adsorption on different types of solid carriers. As compared to the control and other solid supports, Luffa cylindrica resulted in a 3–4-fold increase of the specific side chain cleavage activity after 7 days of incubation. Luffa cylindrica had no significant negative influence on cell growth. Furthermore, it is a natural, inexpensive, non-toxic and mechanically strong material and therefore suitable for follow-up experiments.  相似文献

To determine the effectiveness of rehabilitation on improving ecosystem functions, we examined net photosynthetic rate (P N), tree species composition, soil enzyme activities, and the microclimate (air and soil temperature, relative humidity) of an area on Mt. Makiling that has been rehabilitated and protected from fire for over 12 years. After it was last burned extensively in 1991, restoration was initiated by planting Acacia mangium and Acacia auriculiformis. We selected three areas to study in 2003. Two areas were rehabilitated with A. mangium and A. auriculiformis, and one was still dominated by Imperata cylindrica and Saccharum spontaneum. P N of A. mangium and A. auriculiformis showed significantly lower values than those of I. cylindrica and S. spontaneum. The Acacia plantations had more naturally regenerated tree species than the grassland. Additionally, more tree species appeared in the A. mangium plantation than in the A. auriculiformis plantation. Ficus spetica was present in all of the study sites. Dehydrogenase and phosphatase activities were significantly higher in soil under the Acacia plantations than under grassland. Grassland showed higher air temperature, relative humidity, and soil temperature as well as a larger variation per hour in these parameters compared to the Acacia plantations. The highest air temperature, relative humidity, and soil temperature were measured in April during the dry season. From the regression analysis, soil temperature was significantly correlated with air temperature. Hence plantations, as a rehabilitation activity for grassland, promote natural regeneration and stabilize the microclimate. This stabilization of the microclimate affects establishment and growth of naturally occurring tree species.  相似文献

Coral transplantation has become a potential tool for the restoration of coral cover in degraded reef habitats. Yet, very few investigations have attempted to determine whether there is an advantage for at least two species to be used together in coral transplantation. It is hypothesized that corals would perform better in terms of survival and growth when transplanted in mixed‐ than in single‐species plots. Single‐species plots were compared with combinations of two species at several reef sites, using three separate coral species, namely, Porites cylindrica, Pavona frondifera, and Hydnophora rigida. P. cylindrica performed consistently well in terms of survival whether alone or in the presence of another species. In a stressful environment with strong wave action, P. frondifera performed better when mixed with P. cylindrica than when alone. However, this difference was not evident where wave action was weak. The influence of mixing on the growth rates of H. rigida and P. frondifera transplants could not be examined completely because of high mortality because of predation by the starfish Acanthaster planci and the gastropod Drupella sp. Interestingly, the presence of P. cylindrica appeared to minimize the impact of predation on P. frondifera transplants. The setback caused by predation stresses the importance of other factors that influence the outcome of restoration interventions. Future initiatives should take into consideration management measures when selecting sites in relation to wave action and predators, control predator outbreaks, and use coral species, e.g. P. cylindrica that are less susceptible to predation.  相似文献

The behavior of centrioles in zygotes and female gametes developing parthenogenetically in the anisogamous brown alga Cutieria cyiindrica Okamura was studied using electron and immunofluorescence microscopy. Two pairs of centrioles, detected using anti-centrin antibody, were observed in the vicinity of the male and female nuclei, respectively, just after plasmogamy. The fluorescence intensity of one of the two centrin foci became weak 6 h after plasmogamy and finally disappeared. It was impossible to determine whether the male- or female-derived centrioles disappeared in zygotes, because there was nothing to detect morphological differences between the two centrioles. However, a prominent anti-centrin staining focus was located at the condensed male nucleus in zygotes in which karyogamy had not occurred yet. As a result, it was considered that the maternally inherited centrioles had selectively disappeared during development in C. cylindrica. The paternal inheritance of centrioles in zygotes was also confirmed by electron microscopy. Considering previous observations from oogamous and isogamous species of brown algae, we concluded that the paternal inheriance of centrioles could be universal in the brown algae.  相似文献

在高渗溶液中,用0.05%溶菌酶和2—5mmol·1~(-1)EDTA 处理蓝藻柱孢鱼腥藻、多变鱼腥藻和组囊藻细胞。5—8h 后,70—90%的细胞转为对渗透压敏感的球形体(Spheroplast),又称原生质球。研究了藻的不同培养条件对球形体形成率的影响。测定了 EDTA 处理藻纽胞后外膜脂多糖的释放量。在高渗溶液中,藻细胞和经酶处理获得的球形体的光合放氧活性明显下降,固氮种类的固氮活性失去。饲养层法、固体混合法和含有0.5mg·1~(-1)BA 的液体悬滴培养的柱孢鱼腥藻的球形体,9天后出现再生藻落;在固体混合法培养中获得了组囊藻球形体的再生藻落。在第4天的悬滴培养物中,可以看到球形体发生第一次细胞分裂。再生藻细胞和酶处理物中残留细胞的抗溶菌酶特性有差异。  相似文献

Abstract The present communication defines the conditions under which thioredoxin activates glutamine synthetase from Anabaena cylindrica . Effects are obtained at pH values around neutrality, and the activation is affected by Mg2+ in the assays. The thioredoxin systems from A. cylindrica and spinach are functionally interchangeable in the activation of glutamine synthetase. The enzyme is efficiently activated by thioredoxinm and also by thioredoxinf, but at much higher concentrations. Thioredoxinm has previously been shown to activate NADPH-dependent malate dehydrogenase and isocitrate dehydrogenase from cyanobacteria. It is speculated that thioredoxinm plays a role in the differentiation of vegetative cells to heterocysts.  相似文献

Abstract: Superoxide dismutase (SOD) activity was assayed in vegetative cells, heterocysts and akinetes of Anabaena cylindrica Lemm. The iron-containing isoenzyme (Fe-SOD) was in all cases predominant over the manganese-containing isoenzyme (Mn-SOD). Differentiated cells maintained the same relative content of the two enzymes as in vegetative cells. However, heterocysts and akinetes contained only 20 and 35%, respectively, of the total SOD activity present in vegetative cells.
Both Mn-SOD and Fe-SOD activities increased in all types of cells isolated from A. cylindrica grown at high light intensity. The increase of SOD in heterocysts paralleled that of nitrogenase, suggesting a role of SOD in the protection mechanism of nitrogenase.  相似文献

Eight grassland associations with ten phases and two riverine forest associations were identified on the floodplains of Chitwan National Park in lowland Nepal. TwoThemeda arundinacea associations were primarily stable edaphic associations on mollic soils, often with aquic moisture regimes.Themeda associations were floristically similar to adjacent sal forest. Four mixed tall-grass associations primarily ofNarenga porphyrocoma, Saccharum bengalense, andSaccharum spontaneum were identified. These andThemeda arundinacea types were characterized by a very weak component of woody species. Two grass-shrub associations ofSaccharum bengalense andNarenga porphyrocoma with a strong woody species component typical of riverine forest were described.Trewia nudiflora-Bombax ceiba riverine forest and mixed riverine forest types also were identified on the basis of understory composition. Mixed tall-grass and grass-shrub associations appeared to represent early and late phases of grassland succession to riverine forest. Trajectories of succession are controlled primarily by fire and wildlife grazing.
