The effects of an aqueous plant-derived smoke extract, octanoic acid and ethylene on germination of light-sensitive Grand Rapids lettuce seeds were investigated. The smoke extract brought about a concentration dependent increase in germination and a complete inhibition of germination at high concentrations. Octanoic acid could not induce germination. Ethylene at concentrations over 5 L L–1 increased lettuce seed germination, but not to the same degree as smoke. Aqueous smoke in combination with ethylene showed a synergistic effect on germination at suboptimal smoke concentrations. At high smoke concentrations the effect of ethylene was almost completely inhibited. Octanoic acid in combination with ethylene brought about a higher level of germination than with ethylene alone, but only at the highest concentration of octanoic acid tested (1 mM). Standardized hexane and dichloromethane-partitioned smoke extracts and octanoic acid were subjected to TLC separation. The R f -fractions in the smoke lanes showing activity in the lettuce seed bioassay did not correspond to the R f -value of octanoic acid. As aqueous smoke can withstand autoclaving and can be separated by TLC and HPLC without loosing activity it is unlikely that the activity of aqueous smoke is linked to ethylene. Low molecular weight zinc(II) and copper(II) binding ligands were detected in ultrafiltered human, bovine, and goat milk by the application of the method of modified gel chromatography. Human milk contains at least three detectable low molecular weight copper binders, whereas bovine and goat milk contain at least two. All three milks show two copper binding peaks with the same elution volumes. Zinc chromatograms were less specific than copper. Zinc showed only a single detectable low molecular weight binding ligand common to all three milks. Elution volumes for both zinc(II) and copper(II) citrate and picolinate systems were measured. Elution volumes of both copper(II) and zinc(II) citrate complexes are identical to elution volumes of an intense peak observed with all three milks; it is reasonable to assume that at least part of this peak corresponds to citrate. Human milk alone has a relatively intense binding peak for copper(II) at the same elution volume as the glutamate complex. Human and goat milk have another low intensity copper(II) binding ligand peak at the same elution volume; a number of amino acid complexes have binding peaks at this position. No peak characteristic of the zinc(II) or copper(II) picolinate systems could be found with any of the milks.  相似文献

Zomepirac sodium is a new inhibitor of prostaglandin cyclooxygenase with an in vitro potency equivalent to indomethacin. Since inhibitors of prostaglandin synthesis have marked effects on renal hemodynamics, zomepirac may be expected to reduce renal blood flow (RBF) in a manner similar to indomethacin. This study compares the effects of zomepirac and indomethacin on RBF and electrolyte excretion in anesthetized Rhesus monkeys. Each experiment consisted of a control period followed by 3 or 4 drug treatment periods in which increasing doses of zomepirac (0.5 to 20 mg/kg) or indomethacin (0.5 to 10 mg/kg were given. Indomethacin (5 mg/kg) reduced RBF by 22% and the higher dose (10 mg/kg) reduced RBF by an additional 13%. Zomepirac had little effect on RBF in doses as high as 20 mg/kg. At any given dose the mean plasma concentration of zomepirac was equal to or greater than indomethacin. Peak indomethacin concentration was 48 μg/ml after the 10 mg/kg dose while the peak zomepirac, after 20 mg/kg, was 158 μg/ml. Neither drug had a significant effect on either glomerular filtration rate or excretion rate of sodium or potassium. Thus, zomepirac had only minimal effects on RBF while indomethacin decreased RBF of anesthetized monkeys in a manner qualitatively similar to its effect in other species. The minimal renal effects caused by zomepirac relative to indomethacin in this primate may indicate a therapeutic advantage for zomepirac in man.  相似文献

Global climate change is apparent within the Arctic and the south‐western deserts of North America, with record drought in the latter reflected within 640 000 km2 of the Colorado River Basin. To discern the manner by which natural and anthropogenic drivers have compressed Basin‐wide fish biodiversity, and to establish a baseline for future climate effects, the Stream Hierarchy Model (SHM) was employed to juxtapose fluvial topography against molecular diversities of 1092 Bluehead Sucker (Catostomus discobolus). MtDNA revealed three geomorphically defined evolutionarily significant units (ESUs): Bonneville Basin, upper Little Colorado River and the remaining Colorado River Basin. Microsatellite analyses (16 loci) reinforced distinctiveness of the Bonneville Basin and upper Little Colorado River, but subdivided the Colorado River Basin into seven management units (MUs). One represents a cline of three admixed gene pools comprising the mainstem and its lower‐gradient tributaries. Six others are not only distinct genetically but also demographically (i.e. migrants/generation <9.7%). Two of these (i.e. Grand Canyon and Canyon de Chelly) are defined by geomorphology, two others (i.e. Fremont‐Muddy and San Raphael rivers) are isolated by sharp declivities as they drop precipitously from the west slope into the mainstem Colorado/Green rivers, another represents an isolated impoundment (i.e. Ringdahl Reservoir), while the last corresponds to a recognized subspecies (i.e. Zuni River, NM). Historical legacies of endemic fishes (ESUs) and their evolutionary potential (MUs) are clearly represented in our data, yet their arbiter will be the unrelenting natural and anthropogenic water depletions that will precipitate yet another conservation conflict within this unique but arid region.  相似文献

Abstract

A bulk Lennard-Jones fluid was simulated using the grand canonical Monte Carlo method. Three different sampling methods were used in the transition matrix, namely the Metropolis, Barker and a third novel method. While it can be shown that the Metropolis method will give the most accurate ensemble averages in the limit of an infinitely long run, the new method termed “Modified Barker Sampling” (MBS), is shown to be superior for the runs of practical length for the particular system studied.  相似文献

2013-2018年间采用红外相机技术对西藏雅鲁藏布大峡谷国家级自然保护区的金猫(Catopuma temminckii)进行长期监测, 累计布设70个点位, 14,071个相机工作日。共收集金猫独立照片101张, 其中60张可识别色型, 占金猫独立照片总数的59.40%。共记录到4种不同色型的金猫个体, 拍摄频次比例为麻褐色型:红棕色型:黑色型:灰色型 = 46:7:4:3; 其中灰色型为我国野生金猫实体首次记录。不同色型的个体可同域分布, 而以雅鲁藏布大峡谷自然保护区为代表的藏东南地区是我国金猫色型变异最丰富的地区。基于红外相机照片对金猫的共性形态特征和不同色型的独特形态特征进行了详细的图文描述, 以期为该物种的深入研究提供详实的基础资料。活动节律的分析结果显示, 研究区内金猫以昼行性活动为主, 10:00-12:00为其最强的活动高峰。不同月份的日活动差异指数(daily-discrepancy index) α和昼行性指数(diurnal-nocturnal index) β均存在显著差异, 且活动强度具有明显的季节性变化, 不同季节的活动峰型存在显著差异。本研究为东喜马拉雅生物多样性热点区动物多样性本底的完善积累了基础资料, 也为后续的猫科动物色型多样性的形成机制研究提供了基础信息与参考。  相似文献

Zaozhuang Segment of the Beijing-Hangzhou Grand Canal is an important water conveyance line on the eastern route of the South-North Water Transfer Project in China and its water environment condition can exert a significant influence on diversion water quality safety. This paper analyzed contents of five heavy metals such as Cd, Cu, Ni, Pb and Zn as well as their chemical forms in surface sediments in this region. Methods including Threshold Effects Level-Probable Effects Level (TEL-PEL), Risk Assessment Code (RAC) and Acid Volatile Sulfide- Simultaneously Extracted Metals (AVS-SEM) were also employed to evaluate the quality of sedimentary environment in this region. With the aim to assess the eco-environmental risk revealing the information of the concentration, the chemical fractionation, the toxicity of heavy metals, a new assessment index known as Multivariable Evaluation Index (MEI) was introduced into this paper. Results indicated that contents of such five heavy metals in surface sediments were generally low. Overall, ecological risk was in the medium-low grade in addition to several locations where the level of risk was high. However, certain differences still existed among various assessment results. Comparing with other single evaluation methods, MEI may be able to reflect contamination status and ecological risks of heavy metals in sediments of a specific region more comprehensively. MEI also seemed able to overcome the limitations of other prevalent aggregated indexes to assess potential sediment pollution risk in aquatic systems.  相似文献

Cytochrome c Oxidase (CcO) reduces O2, the terminal electron acceptor, to water in the aerobic, respiratory electron transport chain. The energy released by O2 reductions is stored by removing eight protons from the high pH, N-side, of the membrane with four used for chemistry in the active site and four pumped to the low pH, P-side. The proton transfers must occur along controllable proton pathways that prevent energy dissipating movement towards the N-side. The CcO N-side has well established D- and K-channels to deliver protons to the protein interior. The P-side has a buried core of hydrogen-bonded protonatable residues designated the Proton Loading Site cluster (PLS cluster) and many protonatable residues on the P-side surface, providing no obvious unique exit. Hydrogen bond pathways were identified in Molecular Dynamics (MD) trajectories of Rb. sphaeroides CcO prepared in the PR state with the heme a3 propionate and Glu286 in different protonation states. Grand Canonical Monte Carlo sampling of water locations, polar proton positions and residue protonation states in trajectory snapshots identify a limited number of water mediated, proton paths from PLS cluster to the surface via a (P-exit) cluster of residues. Key P-exit residues include His93, Ser168, Thr100 and Asn96. The hydrogen bonds between PLS cluster and P-exit clusters are mediated by a water wire in a cavity centered near Thr100, whose hydration can be interrupted by a hydrophobic pair, Leu255B (near CuA) and Ile99. Connections between the D channel and PLS via Glu286 are controlled by a second, variably hydrated cavity.

Significance statement

Cytochrome C oxidase plays a crucial role in cellular respiration and energy generation. It reduces O2 to water and uses the released free energy to move protons across mitochondrial and bacterial cell membranes adding to the essential electrochemical gradient. Energy storage requires that protons are taken up from the high pH, N-side and released to the low pH, P-side of the membrane. We identify a potential proton exit from a buried cluster of polar residues (the proton loading site) to the P-side of CcO via paths made up of waters and conserved residues. Two water cavities connect the proton exit pathway to the surface only when hydrated. Changing the degree of hydration may control otherwise energetically favorable proton backflow from the P-side.  相似文献

【背景】怒江大峡谷怒江州段凭借独特的地理环境、丰富的动植物多样性以及极少的人为干扰,拥有很高的生物多样性调查价值,但对于该生境中的放线菌多样性研究尚未见相关报道。【目的】研究怒江大峡谷怒江州段土壤放线菌多样性情况。【方法】利用Illumina Hi Seq平台对怒江州段土壤样品放线菌免培养多样性进行高通量测序,通过相对丰度、物种聚类、主成分分析对不同样品中放线菌组成及其相似性进行了分析。同时通过9种不同类型培养基结合有效的预处理方法和非目的菌抑制剂进行放线菌的纯培养分离。【结果】高通量测序共获得Tag数目474 203条,OTU数目15 671个,它们分布于细菌域下的47个门90个纲170个目320个科561个属。通过不同样品的比较,发现BS、F-G、F-L、L样品的放线菌免培养多样性普遍优于丙中洛样点的BB、BT、BH样品。纯培养分离共获得351株放线菌,分布于放线菌纲下8个目14个科26个属。所用9种培养基中,YIM171培养基的分离效果最好。通过比较不同样品的放线菌纯培养多样性,丙中洛混合样明显优于其他样品。【结论】怒江大峡谷怒江州段土壤样品中放线菌多样性丰富,应进一步挖掘其放线菌资源潜力,为今后放线菌次生代谢产物的开发提供有效的菌株资源。  相似文献

The adsorptive and diffusive behaviour of methane and carbon dioxide in amorphous nanoporous adsorbents composed of spherosilicate building blocks, in which isolated metal sites have been distributed, is examined. The adsorbent contains cubic silicate building blocks (spherosilicate units: Si8O20), which are cross linked by SiCl2O2 bridges and decorated with either –OTiCl3 or –OSiMe3 groups of the other cube corners. The model structures were generated to correspond to experimentally synthesised materials, matching physical properties including density, surface area and accessible volume. It is shown that both methane and carbon dioxide adsorb via physisorption only in the modelled materials. Adsorption isotherms and energies at 300 K for pressures up to 100 bar were generated via molecular simulation. The maximum gravimetric capacity of CH4 is 16.9 wt%, occurring at 300 K and 97 bar. The maximum gravimetric capacity of CO2 is 50.3 wt%, occurring at 300 K and 51.6 bar. The best performing adsorbent was a low-density (high accessible volume) material with no –OTiCl3 groups. The presence of –OTiCl3 did not enhance physisorption even on a volumetric basis, and the high molecular weight of –OTiCl3 groups is a significant penalty on a gravimetric basis. Based on the pair correlation functions, the most favourable adsorption sites for both adsorbates are located in front of the faces of spherosilicate cubes. The self-diffusivity and activation energy for diffusion are also reported.  相似文献
