了解城市内河中病原菌含量对城市生态安全具有重要意义。应用定量PCR( qPCR)方法,对宁波市城区6条重要内河水体中的病原菌含量及主要水质指标进行分析。结果表明,祖关山河、苏家河、南北河、后西河、史魏家河和直落河每升水体中含有病原菌基因的拷贝数分别为:3.34×105、1.47×105、4.68×105、1.75×106、9.65×106和1.33×106;且病原菌基因的含量与水体溶解氧负相关,与总氮、氨氮、浊度、总磷、高锰酸钾指数、叶绿素a和电导率呈正相关关系,但与水体透明度、温度和酸碱度无明显关系。定量PCR分析结果可望为评价城市内河水体质量提供重要参考。  相似文献

目的测定不同生态环境中铜绿丽金龟幼虫肠道细菌产消化酶活性。方法采用平板透明圈法和分光光度计法。结果平板透明圈法测得废弃菜园和花生田中铜绿丽金龟幼虫肠道细菌产蛋白酶和淀粉酶活性差异无统计学意义(F=1.089、0.4963,P0.05),而细菌的产纤维素酶活性有显著差异,其中花生田铜绿丽金龟幼虫肠道中分离到的粘质沙雷菌产酶活性显著高于其他菌株;分光光度计测得的不同生态环境中铜绿丽金龟幼虫肠道细菌的产蛋白酶、淀粉酶和纤维素酶活性差异均有统计学意义(F=461.565、42.349、18.7673,P0.05)。从变异系数来看,分光光度计法测得的蛋白酶、淀粉酶和纤维素酶的变异程度较低,而脂肪酶测定时平板透明圈法的变异系数较低。结论平板透明圈法和分光光度计法均可用于铜绿丽金龟幼虫肠道细菌产消化酶活性测定。通过分析研究比较,分光光度计法适于测定细菌的产蛋白酶、淀粉酶和纤维素酶活性,平板透明圈法适于测定细菌的产脂肪酶活性。  相似文献

Over the past 5 years, raphidophyte blooms have been frequently observed along the South Carolina coastal zone. During the 2002, 2003, and 2004 sampling seasons, we investigated temporal fluctuations of algicidal bacteria abundance against raphidophycean flagellates (Heterosigma akashiwo, Chattonella subsalsa, and Fibrocapsa japonica) using the microplate most probable number (MPN) method in three Kiawah Island brackish stormwater detention ponds (K1, K2, and K75). Local axenic isolates of H. akashiwo, C. subsalsa, and F. japonica were obtained and their susceptibility to algicidal bacteria tested. A total of 195 algicidal bacterial strains were isolated from raphidophyte blooms in the study ponds, and 6 of them were identified at the genus level, and the taxonomic specificity of their algicidal activity was tested against local (pond) and nonlocal isolates of raphidophytes (3 species, 10 total strains). In the ponds, a consistent association was found between raphidophyte bloom development and an increase in bacteria algicidal to the bloom species. In 12 of 15 cases, bloom decline followed the increase in algicidal bacteria to maximum abundances. Although variability was found in the taxonomic specificity of the algicidal bacteria effect (i.e. the number of raphidophyte species affected by a particular bacteria strain) and raphidophyte susceptibility (i.e. the number bacteria strains affecting a particular raphidophyte species), a toxic effect was always found when strains of a raphidophyte species were exposed to algicidal bacteria isolated from a bloom caused by that same species. The results suggest that algicidal bacteria may be an important limiting factor in raphidophyte bloom sustenance and can promote bloom decline in brackish lagoonal eutrophic estuaries.  相似文献

The morphology and infraciliature of a new ciliate, Metopus yantaiensis n. sp., discovered in coastal soil of northern China, were investigated. It is distinguished from its congeners by a combination of the following features: nuclear apparatus situated in the preoral dome; 18–21 somatic ciliary rows, of which three extend onto the preoral dome (dome kineties); three to five distinctly elongated caudal cilia, and 21–29 adoral polykinetids. The 18S rRNA genes of this new species and two congeners, Metopus contortus and Metopus hasei, were sequenced and phylogenetically analyzed. The new species is more closely related to M. hasei and the clevelandellids than to other congeners; both the genus Metopus and the order Metopida are not monophyletic. In addition, the digestion‐resistant bacteria in the cytoplasm of M. yantaiensis were identified, using a 16S rRNA gene clone library, sequencing, and fluorescence in situ hybridization. The detected intracellular bacteria are affiliated with Sphingomonadales, Rhizobiales, Rickettsiales (Alphaproteobacteria), Pseudomonas (Gammaproteobacteria), Rhodocyclales (Betaproteobacteria), Clostridiales (Firmicutes), and Flavobacteriales (Bacteroidetes).  相似文献

不同氮效率水稻生育后期根表和根际土壤硝化特征   总被引:1,自引:0,他引:1  
通过田间试验研究了不同氮效率粳稻品种4007(氮高效)和Elio(氮低效)生育后期在N0(0 kgN hm-2)、N180(180 kgN hm-2)和N300(300 kgN hm-2)水平下根表、根际和土体土壤pH值、铵态氮(NH+4-N)和硝态氮(NO-3-N)含量、硝化强度和氨氧化细菌(AOB)数量.结果表明无论是齐穗期、灌浆期还是成熟期,根表土壤pH值均显著低于根际和土体土壤.土壤pH值范围在5.95至6.84之间变化.土壤NH+4-N含量随水稻生长显著下降,且随施氮量增加而显著增加.根表土壤NH+4-N有明显亏缺区,且随距水稻根表距离增加,NH+4-N含量逐渐升高.土壤NO-3-N含量随水稻生长显著增加,施氮处理均显著高于不施氮处理,但N180和N300处理差异不显著.NO-3-N含量表现为根际>土体>根表.水稻根表和根际土壤硝化强度随水稻生长显著下降,而土体土壤硝化强度随时间延长小幅增加.施氮显著提高4007水稻根表土壤在齐穗和收获期硝化强度以及Elio在齐穗期根际硝化强度,但在施氮处理N180和N300中无显著差异.在整个采样期间,土壤硝化强度均表现为根际>根表>土体.水稻根表和根际AOB数量随水稻生长而显著降低,而土体土壤AOB数量无显著变化.例如,根表土壤AOB数量在齐穗期、灌浆期和收获期分别为16.7×105、8.77×105个g-1 dry soil和8.01×105个g-1 dry soil.根表和根际土壤AOB数量无显著差异,但二者显著高于土体土壤AOB数量.就两个氮效率水稻品种而言,土壤pH值基本无差异.4007土壤NH+4-N含量均显著高于Elio.在齐穗期水稻根表、根际和土体土壤NO-3-N含量在N180水平下均表现为Elio显著高于4007.而在灌浆期和收获期,水稻根表、根际和土体土壤则表现为4007显著高于Elio.在所有采样期,两个水稻品种土体土壤硝化强度和AOB数量在3个施氮量下均无显著差异.Elio根表和根际土壤硝化强度和AOB数量在水稻灌浆期之前一直显著高于4007,而在灌浆期之后则显著低于4007,且最终产量和氮素利用率(NUE)显著低于4007,这可能是由于4007灌浆期后硝化作用强,根际产生的NO-3-N含量高,从而4007根吸收NO-3-N的量也高造成的.因此水稻灌浆期和收获期根表和根际硝化作用以及AOB与水稻高产及氮素高效利用密切相关.  相似文献

Poly-3-hydroxybutyrate (PHB) film pieces were degraded by sulfate reducing Desulfotomaculum sp. incubated under anaerobic laboratory conditions. Degradation started with adherence of the microbial cells and followed by formation of black colonies on the film surface. Scanning electron microscopic (SEM) observations revealed the presence of bacteria and formation of small holes on the film. After 60 days of incubation at 30°C, 10 % weight loss in polymer and 13 % sulfate reduction in the medium was observed. According to gel permeation chromatography (GPC) analysis, the molecular weight of the PHB decreased after 30 days and did not decrease further at a more extended incubation period. Loss of weight of PHB does not seem to be correlated with molecular weight decrease.  相似文献

The lytic transglycosylases (LTs) are bacterial enzymes that catalyze the non-hydrolytic cleavage of the peptidoglycan structures of the bacterial cell wall. They are not catalysts of glycan synthesis as might be surmised from their name. Notwithstanding the seemingly mundane reaction catalyzed by the LTs, their lytic reactions serve bacteria for a series of astonishingly diverse purposes. These purposes include cell-wall synthesis, remodeling, and degradation; for the detection of cell-wall-acting antibiotics; for the expression of the mechanism of cell-wall-acting antibiotics; for the insertion of secretion systems and flagellar assemblies into the cell wall; as a virulence mechanism during infection by certain Gram-negative bacteria; and in the sporulation and germination of Gram-positive spores. Significant advances in the mechanistic understanding of each of these processes have coincided with the successive discovery of new LTs structures. In this review, we provide a systematic perspective on what is known on the structure–function correlations for the LTs, while simultaneously identifying numerous opportunities for the future study of these enigmatic enzymes.  相似文献

叶酸普遍存在于各类食物中,但由于叶酸的不稳定性以及饮食习惯的差异性,各国叶酸缺乏现象普遍存在。叶酸是参与核酸合成及细胞分裂分化的重要物质,叶酸缺乏引起机体功能的混乱,由此引发癌症等一系列的疾病。大部分乳酸菌是叶酸缺陷型菌株,但越来越多的研究表明其很多种类都具有叶酸合成能力。本文主要概述产叶酸乳酸菌的分类,乳酸菌生物合成叶酸的机制,以及利用乳酸菌进行的叶酸基因工程研究进展。  相似文献
