脑胶质瘤在成人原发脑肿瘤中居首位,目前的治疗手段疗效较差,手术切除后复发率高,而化疗药物不能有效的穿透血脑屏障并聚集在肿瘤部位。纳米材料作为载药体为其治疗开辟了新的思路,纳米材料在保持药物稳定性,增加其血液循环时间方面有明显优势。但目前纳米材料还存在着一些亟待解决的问题,如穿透血脑屏障(BBB)、准确靶向于脑胶质瘤细胞等。本文简略论述了纳米材料载药的特性及优势,重点就目前纳米材料载药所面临的问题进行综述,总结了纳米药物穿透血脑屏障的多种策略及纳米药物靶向于脑胶质瘤的不同方式,并详细讨论了目前纳米材料载药多重靶向策略,对其未来的发展进行展望。  相似文献

恶性胶质瘤年发病率约为5/100,000。美国每年有超过14,000例的新发恶性脑胶质瘤患者。治疗主要以手术治疗为主,手术肿瘤的切除程度影响患者的预后。外科手术治疗脑肿瘤需要精确定位脑肿瘤组织在正常脑组织中的位置以便能够获得精确的组织活检和肿瘤的完全切除。量子点是稳定存在的,产生荧光的可视化半导体纳米晶体。静脉注射量子点伴随着网状内皮系统和巨噬细胞的隔离。巨噬细胞可渗入到肿瘤组织并且能够吞噬通过静脉注射的光量子来产生可视化的肿瘤标记。通过巨噬细胞介导,将光量子运输至肿瘤组织展现了一种新兴技术来标记术前肿瘤组织。由于肿瘤组织中的光量子可以被光学成像和光谱学工具来探测,因此在脑肿瘤组织活检和切除中可以为外科医生提供可视化得实时反馈。  相似文献

Summary We report the development of cytotoxic T lymphocytes specific for an allogeneic brain tumor in a rat model. DA strain cytotoxic T cell precursors stimulated by an allogeneic tumor (9L gliosarcoma) from the Fischer rat could generate a population of cytotoxic T lymphocytes that lysed the allogeneic 9L tumor but failed to lyse other targets, including Fischer concanavalin-A(ConA)-stimulated lymphoid blast targets. DA T cells depleted of reactivity to the Fischer haplotype (DA-f) retained reactivity to the 9L tumor, demonstrating that T cell precursors with specificity for normal Fischer alloantigens were not required for the generation of a response to the 9L Fischer tumor. The preferential lysis of the tumor target did not simply reflect a higher density of Fischer target antigens on the tumor than that found on normal Fischer ConA blast targets. First, the relative densities of class I antigen on the 9L tumor and normal Fischer ConA blasts were comparable. Second, cytotoxic T cells could not be generated from DA-f precursors when Fischer ConA blasts were used as stimulators. If DA-f T cells were simply responding to the higher density of Fischer antigen found on 9L tumor, it would have been expected that the ConA blasts expressing comparable levels of antigen to that found on the tumor would have generated cytotoxicity for both the 9L and ConA targets. We conclude that the cytotoxic T cells are specific for a determinant expressed only by the tumor. Such tumor-specific cytotoxic T cells could be useful in vivo for adoptive immunotherapy of brain tumors.  相似文献

目的:通过放射性核素~(99m)Tc标记BmK CT多肽制备靶向胶质瘤的显像剂,探讨~(99m)?Tc-BmK CT用于胶质瘤显像的可行性。方法:采用BmK CT多肽游离的氨基与DTPA酸酐反应得到BmK CT-DTPA,经99m Tc标记后通过柱层析分离纯化制备~(99m)?Tc-BmK CT。测定标记物在PBS溶液和血清中不同时间点放射性化学纯度,评价BmK CT-~(99m)?Tc体外稳定性。新西兰白兔耳缘静脉注射~(99m)Tc-BmK CT进行SPECT显像,观察不同时间点体内的放射性分布。皮下胶质瘤裸鼠经尾静脉注射~(99m)Tc-BmK CT,观察不同时间点肿瘤的摄取情况;注射后4 h处死裸鼠,分离肿瘤和主要器官进行离体SPECT显像,并用勾画感兴趣区法分析相对放射性计数。结果:~(99m)Tc标记BmK CT多肽标记率大于80%,经柱层析分离纯化后放射性化学纯度大于99%。标记物在PBS和血清稳定性良好,6 h内放射性化学纯度均大于95%,12 h内放射性化学纯度大于90%。正常白兔SPECT显像表明~(99m)Tc-BmK CT主要浓聚在肝脏、脾脏和肾脏,软组织持续显影微弱,甲状腺区及胃肠未见核素浓聚;显像剂主要通过泌尿系统排泄,24 h肾脏与肝脏显影接近。胶质瘤裸鼠SPECT显像表明,注射后4 h肿瘤显像清楚,ROI分析结果显示肿瘤/肌肉比4.26±0.25,标记物在肿瘤内代谢缓慢,8 h肿瘤部位仍有较高摄取。结论:本研究成功制备了~(99m)Tc标记BmK CT多肽,标记物主要被肝、脾和肾摄取,经泌尿系统排泄;~(99m)Tc-BmK CT能够在皮下胶质瘤中浓聚,注射后4 h肿瘤显影清晰,瘤内代谢缓慢,有潜力成为一种新型胶质瘤分子探针。  相似文献

Free radicals are associated with glioma tumors. Here, we report on the ability of an anticancer nitrone compound, OKN-007 [Oklahoma Nitrone 007; a disulfonyl derivative of α-phenyl-tert-butyl nitrone (PBN)] to decrease free radical levels in F98 rat gliomas using combined molecular magnetic resonance imaging (mMRI) and immunospin-trapping (IST) methodologies. Free radicals are trapped with the spin-trapping agent, 5,5-dimethyl-1-pyrroline N-oxide (DMPO), to form DMPO macromolecule radical adducts, and then further tagged by immunospin trapping by an antibody against DMPO adducts. In this study, we combined mMRI with a biotin–Gd-DTPA–albumin-based contrast agent for signal detection with the specificity of an antibody for DMPO nitrone adducts (anti-DMPO probe), to detect in vivo free radicals in OKN-007-treated rat F98 gliomas. OKN-007 was found to significantly decrease (P < 0.05) free radical levels detected with an anti-DMPO probe in treated animals compared to untreated rats. Immunoelectron microscopy was used with gold-labeled antibiotin to detect the anti-DMPO probe within the plasma membrane of F98 tumor cells from rats administered anti-DMPO in vivo. OKN-007 was also found to decrease nuclear factor erythroid 2-related factor 2, inducible nitric oxide synthase, 3-nitrotyrosine, and malondialdehyde in ex vivo F98 glioma tissues via immunohistochemistry, as well as decrease 3-nitrotyrosine and malondialdehyde adducts in vitro in F98 cells via ELISA. The results indicate that OKN-007 effectively decreases free radicals associated with glioma tumor growth. Furthermore, this method can potentially be applied toward other types of cancers for the in vivo detection of macromolecular free radicals and the assessment of antioxidants.  相似文献

The α9β1 integrin accelerates cell migration through binding of the α9 cytoplasmic domain to SSAT, which catalyzes the catabolism of higher order polyamines, spermidine and spermine, to the lower order polyamine, putrescine. SSAT levels were downregulated at both the mRNA and protein levels by shRNA-mediated simultaneous knockdown of MMP-9 and uPAR/cathepsin B. In addition, we noted a prominent reduction in the expression of SSAT with MMP-9 and uPAR/cathepsin B knockdown in the tumor regions of 5310 injected nude mice brains. Further, SSAT knockdown in glioma xenograft cells significantly reduced their migration potential. Interestingly, MMP-9, uPAR and cathepsin B overexpression in these xenograft cells significantly elevated SSAT mRNA and protein levels. The migratory potential of MMP-9/uPAR/cathepsin B-overexpressed 4910 and 5310 cells was not affected by either glybenclamide (Kir 6.x inhibitor) or tertiapin-Q (Kir 1.1 and 3.x inhibitor) but instead was significantly inhibited by either barium or Kir4.2 siRNA treatments. Co-localization of α9 integrin with Kir4.2 was observed in both 4910 and 5310 xenograft cells. However, MMP-9 and uPAR/cathepsin B knockdown in these cells prominently reduced the co-localization of α9 with Kir4.2. Taken together, our results clearly demonstrate that α9β1 integrin-mediated cell migration utilizes SSAT and the Kir4.2 potassium channel pathway, and inhibition of the migratory potential of these glioma xenograft cells by simultaneous knockdown of MMP-9 and uPAR/cathepsin B could be attributed to the reduced SSAT levels and co-localization of α9 integrin with Kir4.2 inward rectifier potassium channels.  相似文献

目的:研究脑胶质瘤中p16基因启动子区甲基化情况及其临床意义。方法:用甲基化特异性PCR技术检测42例脑胶质瘤组织和癌旁正常脑组织中p16基因启动子甲基化,并分析该基因启动子甲基化与临床病理特征之间的关系。结果:脑胶质瘤组织中p16基因异常甲基化率(38.27%)显著高于癌旁正常脑组织中p16基因的异常甲基化率(8.8%,P=0.000)。发生甲基化的肿瘤组织或者正常脑组织中p16基因mRNA和蛋白表达显著降低。此外,p16基因异常甲基化和肿瘤病理分级有相关性(P=0.007),而与患者性别、年龄及肿瘤类型等临床特征无关(P=0.669,0.869和0.944)。结论:p16基因启动子区CpG岛高甲基化与p16表达下调相关,推测p16启动子区CpG岛高甲基化是导致p16基因在脑胶质瘤中表达下调的重要因素,有望成为脑胶质瘤早期辅助诊断的分子标志物之一。  相似文献

目的:探讨放化疗联合DC-CIK治疗脑胶质瘤复发的临床疗效和安全性.方法:选择我院自2006年1月至2011年9月收治的40例脑胶质瘤复发患者,采用放化疗联合DC-CIK治疗.统计记录治疗效果、生存期、治疗并发症和生活质量评分.结果:40例患者完全缓解8例,部分缓解11例,稳定10例,恶化11例,总治疗有效率为47.5%,在消化道反应、血象异常、内分泌紊乱、感染发热、呼吸道炎症等项目上并发症的发生率均维持在低水平,3、9、15个月生存率分别为85.7%、65.7%、40.0%,KPS评分为78.5± 8.6,组间差异有统计学意义(P<0.05).结论:放疗联合DC-CIK能提高脑胶质瘤复发患者的生活质量,延长生存期.  相似文献

探讨FCFR1OP和PTEN在胶质瘤中的表达情况和临床意义。选取54例胶质瘤手术切除标本,采用SP法进行免疫组织化学染色。FGFR1OP和PTEN的阳性表达率分别为85.2%和48.1%,且随着胶质瘤的恶性程度不同表现出明显的统计学差异(P〈0.05)。FGFR1OP的高表达与PTEN的低表达可能参与胶质瘤的生长分化和进展,并提示预后不良。  相似文献

MicroRNAs have the capacity to coordinately repress multiple target genes and interfere with biological functions of the cell, such as proliferation and apoptosis. Here we report that miR-136 is downregulated in human glioma, and that the miRNA promotes apoptosis of glioma cells induced by chemotherapy. Two anti-apoptotic genes, AEG-1 and Bcl-2, are identified as targets of miR-136, and restoration of AEG-1 or Bcl-2 expression suppresses miR-136-enhanced apoptosis. Therefore, miR-136 might play a tumor-suppressive role in human glioma and thereby might represent a potential therapeutic strategy.  相似文献
