磁场对急性胃损伤的治疗效应及其机制的实验研究   总被引:4,自引:0,他引:4

目的:探讨腹部恒磁场作用,对大鼠急性胃损伤模型的治疗效应及其作用机制.方法:10只健康SD大鼠,以indomethacin胃灌注法复制急性胃损伤模型,以表面磁强度为1300-1600GS,钡铁氧体恒磁场作用大鼠腹部3小时,观察胃损伤指数及病理损伤积分,同时对血浆中内皮素(endothelin,ET)、一氧化氮(nitric oxide,NO)、谷胱甘肽过氧化物酶(GSH-Px)及超氧化物岐化酶(SOD)水平进行比较.结果:恒磁场腹部作用后,大鼠胃损伤指数及病理损伤积分较对照组均显著减轻(p均〈0.05);血浆ET-1和NO水平却无无明显改变(p均〉0.05),血GSH-Px和SOD含量较对照组均明显升高(p均〈0.05).结论:腹部1300-1600GS恒磁场作用3小时,对大鼠急性胃损伤具有明显的治疗效应,磁场的这一效应可能与其增加血浆中清除氧自由基的GSH-Px和SOD活性有关.  相似文献

谷胱甘肽过氧化物酶是机体内重要抗氧酶系之一。它的活力和含量,反映机体清除自由基的能力。自由基对细胞结构的损伤很大,随着年龄的增长,抗氧化酶活力逐渐下降,从而引起自由基及脂质过氧化产物日益增多,最终导致机体衰老和老年性疾病的发生^[1]。本试验试图探讨DNA对谷胱甘肽过氧化物酶活性影响从而探索DNA对抗自由基的作用。  相似文献

The effect of diets containing antioxidant vitamins and trace elements on chicken tissue activities of SOD, CAT, GSH-Px and of LPO levels was investigated. Chickens, 45 weeks of age were divided into six groups: control group, Cu group (13.2 mg Cu kg(-1) diet); Se group (0.07 mg Se kg(-l) diet); vitamin E group (70 mg DL-alpha-tocopherol acetate kg(-1) diet) and a constant level vitamin C, 200 mg kg(-1) diet); vitamin A group (240 mg retinol acetate kg(-1) diet) and vitamin C group (500 mg ascorbic acid kg(-1) diet). Significant variation of these antioxidant enzyme activities and LPO levels according to gender was demonstrated statistically. In the Cu group, CuZnSOD activity in the liver, erythrocyte, kidney and heart significantly increased by 75, 40, 12, 12% respectively (P<0.05). MnSOD activity in the heart, liver, kidney and brain of the vitamin C and in the heart of Cu group were found to be increased by approximately 15%, while in liver tissue of the Cu group it was reduced by 19% (P<0.05). GSH-Px activities in the Se, vitamin E and C groups were significantly increased, conversely LPO levels decreased (P<0.001). CAT activities in the liver and heart of the vitamin C group were significantly decreased (by 32%), but in kidney tissue only that of the Cu group was increased from 30.2 +/- 4.767 to 144.49 +/- 6.93 U mg(-1) P<0.001. The resistance to stress of the vitamin E and C groups, which had significantly increased activities of antioxidant enzymes and decreased lipid peroxide levels, were determined in 60% moisture medium at 45 degrees C.  相似文献

Hydroperoxide detoxification in African trypanosomes is achieved by 2-Cys-peroxiredoxin (TXNPx)- and non-selenium glutathione peroxidase (Px)-type enzymes which both obtain their reducing equivalents from the unique trypanothione/tryparedoxin system. Previous RNA interference approaches revealed that the cytosolic TXNPx and the Px-type enzymes are essential for Trypanosoma brucei. Because of partially overlapping in vitro substrate specificities and subcellular localisation the physiological function of the individual enzymes was not yet clear. As shown here, TXNPx and Px are expressed at comparable levels and in their active reduced state. Px-overexpressing parasites were less sensitive toward linoleic acid hydroperoxide but not hydrogen peroxide. Kinetic studies confirmed that Px—but not TXNPx—reduces lipophilic hydroperoxides including phospholipids with high efficiency. Most interestingly, the severe proliferation defect of Px-depleted bloodstream cells could be rescued by Trolox, but not by hydrophilic antioxidants, in the medium. This allowed us to knock-out the three Px genes individually and thus to distinguish their in vivo role. Deletion of the cytosolic Px I and II resulted in extremely fast membrane peroxidation followed by cell lysis. Cells lacking specifically the mitochondrial Px III showed a transient growth retardation and cardiolipin peroxidation but adapted within 24 h to normal proliferation.  相似文献

为探讨黄芩黄酮防治白内障的作用机理 ,采用半定量RT PCR方法比较正常组、白内障组和中药防治组大鼠晶状体中GSH Px、GR和Cu ZnSOD的mRNA水平 .白内障组GSH Px、GR和Cu ZnSOD的mRNA水平在 15d龄时显著高于正常 ,然后下降 ;在 2 7d和 31d龄 ,GR和Cu ZnSOD的mRNA水平下降至与正常无显著差异 ,GSH PxmRNA水平仍略高于正常 .中药防治组晶状体中 ,3种抗氧化酶的mRNA水平在各实验取样点无明显变化 ;其中 ,GR和Cu ZnSOD的mRNA水平一直与正常无显著差异 ,GSH PxmRNA水平略高于正常 .黄芩黄酮可能通过有效清除亚硒酸钠间接产生的活性氧来防止白内障的发生 ,并使亚硒酸钠对晶状体抗氧化酶表达的影响得以消除  相似文献

The crystal structure of reduced tryparedoxin peroxidase shows Cys47 close to Gln82 and Trp137 and helix formation of residues 87 to 97 whereas the NMR structure of the reduced C76S mutant adopts a different conformation similar to the oxidized protein. Circular dichroism (CD), fluorescence and NMR spectroscopy reveal that the fully active C76S mutant differs from the wildtype (WT) enzyme mainly in its reduced form both in secondary structure content and Trp137 environment. This implies that Cys76 plays a critical role for the reduced enzyme assuming different conformational states and that the catalytic triad may only be necessary as short-lived intermediate during catalysis.  相似文献

An enormous amount of data has been published in recent years demonstrating melatonin's defensive role against toxic free radicals. In the present study, we examined the role of melatonin as an antioxidant against the effect of continuous light exposure. Rats were divided into three groups. Control rats (group A) were kept under natural conditions whereas other group of rats (group B and C) were exposed to constant light for inhibition of melatonin secretion by the pineal gland. Group C rats also received melatonin via s.c. injection (1 mg x kg(- 1) body weight x day(- 1)). At the end of experiment, all animals were sacrificied by decapitation, serum and tissue samples were removed for determination of malondialdehyde (MDA), a product of lipid peroxidation, conjugated dienes levels and glutathione peroxidase (GSH-Px) activity levels. It was found that lipid peroxidation was increased in the rats which were exposed to constant light. Melatonin injection caused a decrease in lipid peroxidation, especially in the brain. In addition, melatonin application resulted in increased GSH-Px activity, which has an antioxidant effect. Thus, melatonin is not only a direct scavenger of toxic radicals, but also stimulates the antioxidative enzyme GSH-Px activity to detoxify hydroxyl radical produced by constant light exposure.  相似文献

In this study, the relationship between selenium (Se) and glutathione peroxidase (GSHPx) levels was investigated in 7,12-dimethylbenz(a)anthracene (7,12-DMBA)-treated mouse liver. The potential mammary carcinogen 7,12-DMBA, was injected intraperitoneally (20 mg kg(-1) day(-1)) into 10-12 month old female mice. After 21 days of application the mice were sacrificed and GSHPx and Se levels of liver homogenates were measured. Se and GSHPx levels in 7,12-DMBA-treated mice were significantly lower than those of controls (p < 0.05). The control group exhibited 0.9 +/- 0.066 U mg(-1) protein and 0.86 +/- 0.058 p.p.m. levels of GSHPx and Se respectively. The 7,12-DMBA-treated group had significantly (p < 0.05) decreased GSHPx and Se levels (0.42 +/- 0.062 U mg(-1) protein and 0.69 +/- 0.034 p.p.m. respectively). The results show a direct relationship between Se and GSHPx activity and a negative correlation between antioxidant capacity and existence of a carcinogen in metabolism.  相似文献

Recent data from several reports indicate that free radicals are involved in aetiopathogenesis of many human pathologies including neuropsychiatric disorders such as schizophrenia, bipolar disorder etc. In the present study, we aimed at determining and evaluating levels of malondialdehyde (MDA), a product of lipid peroxidation, and antioxidant enzyme superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity levels in patients diagnosed with schizophrenia (n = 25) and bipolar disorder (n = 23). The control group was composed of 20 healthy subjects. There was a significant increase in MDA levels of patients with schizophrenia and bipolar disorder compared with controls. SOD and GSH-Px activity levels were significantly higher in the schizophrenic group compared with controls. SOD activity levels in bipolar the group were significantly higher than controls whereas there were no significant changes in GSH-Px activity levels in the bipolar group and controls. Significant differences between lipid peroxidation product and antioxidant enzyme (SOD and GSH-Px) activity levels in schizophrenic and bipolar disorder patients compared with controls leads us to believe that these differences are related to the heterogenities in aetiologies of these disorders.  相似文献
