Maternally inherited bacteria of the genus Wolbachia are known to affect the physiology and reproduction of several arthropod and nematode hosts. This paper presents the effects of using tetracycline to eliminate Wolbachia on the oogenesis of its host uzifly Exorista sorbillans (Diptera: Tachinidae), a parasitoid of silkworm Bombyx mori L. Earlier we reported cytoplasmic incompatibility, nucleo‐cytoplasmic conflict, male biased sex ratio and reduction in fecundity upon curing of Wolbachia infection using an antibiotic. This paper presents the abnormal growth and proliferation of oocytes in the uninfected/cured uziflies. The anomalies include abnormal/arrested oocyte production with inhibition of normal functioning of nurse cell nuclei, malformation and agglutination in egg chambers, empty egg chambers due to arrested vitellogenesis and undeveloped ovarian tubules. These anomalies further lead to reduction in growth of the ovarian tubules, which resulted in a 20% reduction of fecundity. It is hypothesized that normal oocyte production in uzifly requires the infection of Wolbachia bacteria. Further, in the uzifly, infection is at transition stage from a facultative to an obligatory mode of symbiosis. In light of this, this paper discusses the relationship between Wolbachia and oocyte production.  相似文献

Exorista (=Tachina) larvarum (L.) (Diptera, Tachinidae), a polyphagous parasitoid that attacksLymantria dispar L. andHyphantria cunea (Drury), was rearedin vitro from egg to adult on four tissue culture media-based diets (TMM-FH, SCHNEIDER'S, EX-CELL 400, and SF-900). The kind of tissue culture media in the diets did not influence the adult yield (34 to 55%) and puparium weight (26–27 mg). Adult yield and the puparium weight ofE. larvarum developed on TNM-FH and SCHNEIDER'S-based diets containing different amounts ofGalleria mellonella pupal extract (PE) (0, 1.25, 2.5 and 5%), were lower on diets without PE. In diets without PE development times from oviposition to adult emergence, were shorter on TNM-FH (19 days) than on SCHNEIDER'S-based diet (25–26 days). The adults that developed on artificial diets were able to parasitize the factitious hostG. mellonella and produce viable progeny. The results demonstrate thatE. larvarum is the most promising parasitoid ever studied forin vitro mass production.  相似文献

为从分子水平探索不同地区伞裙追寄蝇种群间的内在联系,本文利用ISSR分子标记技术对8个不同地区伞裙追寄蝇自然种群进行遗传多样性、种群间分化程度以及聚类分析等研究。结果表明:筛选出11对多态性稳定的ISSR引物,对8个地区伞裙追寄蝇群体的80个个体进行PCR扩增,共获得166个重复性好且清晰可辨的ISSR条带,平均每条引物扩增出15.0909个片段,且均为多态性条带,多态信息含量(PIC)为0.8441-0.8653;香农信息指数(I)为0.1240-0.3455;Nei's遗传多样性指数(H)为0.0841-0.2285;基因分化率(Gst)为28.78%,基因流(Nm)的均值为1.5702,即遗传变异主要存在于个体之间,不同种群间基因交流处于中等水平;8个地区伞裙追寄蝇种群被划分为4个类群,种群间的遗传分化与地理距离呈正相关关系。  相似文献

Exorista larvarum (L.) (Diptera: Tachinidae), a larval parasitoid of Lepidoptera, can be reared from egg to fecund adult on artificial media composed of crude components. The standard in vitro culture is performed in 24‐well plastic rearing plates. Exorista larvarum eggs, removed from superparasitized larvae of Galleria mellonella (L.) (Lepidoptera: Pyralidae), are individually placed in the wells, each containing a cotton swab soaked in liquid medium. The plates are then sealed until parasitoid puparium formation. To avoid contamination by microorganisms, the artificial medium is routinely supplemented with 0.01% solution of gentamicin. Experiments were carried out to assess whether this broad‐spectrum antibiotic may be replaced with hydrolate of Monarda fistulosa L. (Lamiaceae), which was selected due to its high in vitro activity against pathogenic microorganisms for humans and plants. The hydrolate was either supplemented to the artificial medium (0.5% wt/wt) (first experiment) or placed in an empty well (200 μl) of the rearing plate, to be supplied as saturated air due to evaporation (second experiment). In both experiments, a standard medium with gentamicin and an antimicrobial‐free medium were maintained as positive and negative controls, respectively. In the first experiment, in the hydrolate‐supplemented medium fewer E. larvarum completed egg‐to‐adult development than in the standard medium, but significantly more parasitoids developed from egg to adult compared to the antimicrobial‐free medium. No significant difference was found between the numbers of eggs laid by the females obtained from the standard medium vs. those from the hydrolate‐supplemented medium. In the second experiment, the hydrolate‐saturated air significantly decreased E. larvarum egg hatching, puparium formation, and female fecundity compared to the standard medium. In perspective, M. fistulosa hydrolate supplemented to the artificial media for E. larvarum may be considered as a promising candidate to replace the gentamicin solution, as suggested also by the microbiological analyses of the media, performed at various growth stages of the parasitoid in a separate trial. Conversely, the hydrolate‐saturated air treatment was deemed unsuitable.  相似文献

为探索伞裙追寄蝇蛹滞育调控的分子机制,本文对伞裙追寄蝇Exorista civilis滞育蛹和非滞育蛹进行转录组测序以及代谢组检测,鉴定其关键的滞育关联基因(diapause-associated genes, DEGs)和滞育关联代谢物(diapasuse-associated metabolites, DEMs)。本研究基于高通量测序以及液质联用技术,通过筛选,在转录组中获得了差异表达基因7 513个,在代谢组中,获得差异代谢物501个,其中氨基酸占比最多。将所有差异表达基因与差异表达代谢物同时向KEGG映射,获得两者共同的pathway信息,明确差异表达基因与差异代谢物共同参与的主要生化途径和信号转导途径。本研究通过转录组和代谢组的联合分析,在正、负离子模式下,差异表达基因与差异代谢物共同富集到70条通路。在负离子模式下,滞育关联因子主要参与氨基酸代谢和神经系统;在正离子模式下,滞育关联因子主要参与消化系统和信号转导途径。本研究重点分析了柠檬酸循环、cAMP信号通路、氨酰-tRNA的生物合成,将为进一步深入研究伞裙追寄蝇滞育调控的分子机制奠定理论基础。  相似文献

Laboratory tests were conducted in Italy to evaluate the acceptance and suitability of the alien butterfly Cacyreus marshalli Butler as host for three indigenous parasitoids, Trichogramma brassicae (Bezdenko), Exorista larvarum (L.) and Brachymeria tibialis (Walker). Only E. larvarum and B. tibialis showed potential to adapt to C. marshalli. Their contribution to biological control appeared to be especially related to host mortality due to incomplete parasitoid development.  相似文献

Exorista sorbillans is a tachinid endoparasitoid of silkworm, Bombyx mori, and is globally known as uzi. It causes economic injury to the cocoon crop in silkworm cultivating areas of India, except those above 400 m above mean sea level (AMSL) in the foothills of the Himalayas (Darjeeling). It is reported that the sericulture tract of south India became infected with this pest only since 1980 through an accidental transportation of cocoons from West Bengal. To ascertain whether the genome of this parasitoid is differentiating into discrete gene pools in contrasting geo-climatic conditions, molecular profiling of four populations (Es Annatapur, EsRamanagaram, Es Channapatna and EsKodathi from south India and EsMurshidabad from Murshidabad, West Bengal was undertaken with 13 ISSR, 3 RAPD and six non-random primers designed from various repeat sequences of B. mori . MANOVA indicated significance for the Roy’s largest root estimate (55.4; F =18.47; p = 0.002) for the variability contributed by the replication. Further, hierarchical clustering done on the basis of Euclidean distance matrix and Nei’s unbiased Phylip clustering put EsMurshidabad at the maximum distance from those of south India and 29 markers could also be identified which significantly differentiateEsMurshidabad from others. However, Nei’s statistics for gene diversity in sub-populations reveal considerably high gene-flow (3.44 and 2.51) among the populations around Bangalore. The gene-flow between EsMurshidabad and other population is lowest but cannot be ignored. The comparison of endosymbiont specific 16SrRNA and fts Z gene (partial) sequences through clustalW (gcgMSF) revealed a closer relationship of EsMurshidabad withEsAnnatapurand Es Ramanagaram and is not congruent with the relationships discussed above. The significance of this maiden study with a tachinid fly-pest is discussed in the context of understanding the diversification of Uzi fly-pest and also establishing this pest as a relevant biological material for studying microevolution in future.  相似文献

The tachinid Exorista larvarum (L.), a polyphagous gregarious larval endoparasitoid of Lepidoptera, was reared from egg to fecund adult on media containing commercial meat homogenates for babies as the main ingredient. Four media, each containing a diverse homogenate supplemented with extract of Galleria mellonella L. pupae, were tested first. Despite the difference in nutrient content, the kind of homogenate did not significantly affect the adult yields (30.2 to 40.7%) or puparial weights. Two other diets free of host materials (I and II) were then tested. Both were based on Gerber veal homogenate combined with different amounts of yeast extract and chicken egg yolk and were supplemented with wheat germ (I) or saccharose (II). Adult yields (28.7 to 32.7%) and puparial weights did not differ significantly between the two diets. Fly longevity and fecundity of the females obtained on diet I were comparable to those of the females emerged from puparia formed in G. mellonella larvae. Male and female puparial weights were, however, higher and development times longer on the diet than in the host.  相似文献
