用常规电镜方法观察了锦橙[Citrussinensis (L.) Osb.]汁囊从原始细胞到发育为一个具柄的成熟汁囊的过程中,汁囊构成细胞超微结构的变化。锦橙汁囊原始细胞及发育为球状体时的构成细胞以及柱状结构顶端的细胞都是一种典型的分生组织细胞。在细胞质中有包括线粒体、质体、内质网、核糖体等丰富的细胞器,但没有观察到高尔基体。这些分生细胞分裂一段时期后就停止活动,逐渐分化为适应贮藏功能的液泡化薄壁细胞。分生细胞开始分化时,在细胞中出现许多小液泡和高尔基体。这些小液泡逐渐地融合,同时细胞质变少,最后形成一个有中央大液泡的薄壁细胞,在紧贴细胞膜的薄薄的一层细胞质中有线粒体、质体、高尔基体以及含有许多脂滴的杂色体。但成熟果实中汁囊的薄壁细胞中几乎没有任何细胞器。  相似文献

We investigated the effects of antifreeze protein (AFP) III supplementation on the cryopreservation of rabbit sperm cells and embryos. Ejaculated semen was collected from male Japanese white (JW) rabbits and divided into four AFP-supplemented groups (0.1 μg/ml, 1 μg/ml, 10 μg/ml, 100 μg/ml) and one control group with no AFP-supplementation. The semen samples were treated with egg-yolk HEPES extender containing 6% acetamide before the sperm was cooled from room temperature to 5 °C, then packed into sperm straws. The straws were frozen in steam of liquid nitrogen (LN2) and then preserved in the LN2. The motility of the sperm after thawing in 37 °C water was analyzed. The percentage of rapidly motile sperm in the 1 μg/ml AFP group was significantly higher than in the control group. Morulae were collected from female JW rabbits and divided into three AFP-supplemented groups (100 ng/ml, 500 ng/ml, 1000 ng/ml) and one control group. The morulae, immersed in an embryo-freezing solution (M199-HEPES containing 20% ethylene glycol, 20% dimethylsulfoxide, 10% fetal bovine serum and 0.25 M sucrose), were packed into open pulled embryo straws and vitrified in LN2. The frozen embryos were thawed in the embryo-freezing solution, and the rates of embryo survival and development to blastocyte stage were analyzed after incubation for 72 h. The development rate of the embryos in the 500 ng/ml AFP group was significantly higher than in the control group, but that in the 1000 ng/ml AFP group was significantly lower. In conclusion, the appropriate dose of AFP III increased the number of rapidly motile sperm and embryo survival following freezing and thawing. The results suggest that supplementation with AFP III can increase the efficiency of cryopreservation of rabbit sperm cells and embryos.  相似文献

The effects of in ovo peptide YY (PYY) or epidermal growth factor (EGF) administration on chick growth, yolk absorption and yolk stalk function in posthatch (0–5 days) meat-type or broiler chicks were determined. At Day 18 of incubation, treated eggs were injected into the air cell with 100 μl of either PYY (Trial 1) or EGF (Trial 2) at a dosage of 600 μg/kg egg weight. Saline-treated control eggs were injected similarly with 0.9% saline. At hatch, 200 μl of 51Cr-labeled microspheres were injected into chick yolk sacs. Epidermal growth factor increased ileal wet weight adjusted for body weight as well as ileal serosal dry matter. Body weight, feed consumption and excreta weight per bird, and relative weights of the yolk sac, intestine and liver were significantly affected by age of the chick in both trials. Relative radioactivity of the yolk sac, yolk stalk, blood, liver, and kidneys were affected by bird age in Trial 2; however, there were no significant effects due to PYY or EGF treatments on relative radioactivity of the tissues and organs examined. These data suggest that PYY and EGF had no effect on yolk absorption or yolk stalk function through 5 days in the posthatch chick.  相似文献

目的观察血管内皮生长因子受体3(vascular endothelial growth factor receptor-3, VEGFR-3)蛋白在大鼠第15d胚胎和第21d胚胎皮肤淋巴管和血管内皮的表达水平,探讨VEGFR-3在胚胎淋巴管和血管发生发育中的生物功能.方法应用免疫组织化学技术(SABC法)对大鼠第15d和第21d胚胎共61例皮肤样本进行抗鼠VEGFR-3免疫组织化学染色,观察VEGFR-3在皮肤淋巴管和血管内皮细胞的表达情况.结果大鼠胚胎皮肤淋巴管和血管内皮细胞均阳性表达VEGFR-3蛋白,VEGFR-3蛋白在大鼠胚胎第15d、胚胎第21d皮肤淋巴管内皮细胞的阳性表达率分别是38.71%(12/31)和73.33%(22/30),第21d胚胎皮肤淋巴管VEGFR-3蛋白的表达水平明显高于第15d胚胎皮肤淋巴管VEGFR-3蛋白的表达水平(P<0.01).VEGFR-3蛋白在大鼠胚胎第15d、胚胎第21d皮肤血管内皮细胞的阳性表达率分别是48.39%(15/31)和46.67%(14/30),第15d和第 21d胚胎皮肤血管VEGFR-3蛋白的表达水平之间无明显差异(P>0.05).结论随着大鼠胚胎的发育,VEGFR-3蛋白在皮肤淋巴管的表达水平表现为明显上升的趋势,提示VEGFR-3在淋巴管发生发育中的作用逐渐增强.  相似文献

Embryo rescue technique was used successfully to produce interspecific hybrids by crossing peach (P. persica) as a female parent with apricot (P. armeniaca) and plum (P. salicica). In those crosses that had ‘Yuhualu’ or ‘Zhonghuashoutao’ as female parents, hybrid embryos aborted from the 7th or 8th week after pollination mainly due to post-pollination incompatibility. An embryo rescue protocol was established to rescue such embryos and recover hybrid plants. Modified half-strength MS medium containing 4 mg l−1 6-BA and 0.5 mg l−1 IBA produced up to 90% germination in the embryos. Modified MS medium with 1.0 mg l−1 6-BA and 1.0 mg l−1 IBA gave the highest bud induction and multiplication whereas modified MS medium containing 0.5 mg l−1 IAA and 0.2 mg l−1 NAA gave the best rooting percentage. All the hybrids obtained using this embryo rescue technique were verified using simple sequence repeat (SSR) markers. A series of pollen treatments were carried out to partially overcome pre-pollination incompatibility, and it was found accidentally that pollen treatment with electrostatic field not only improved pollen germination but also increased the multiplication coefficient of embryo-induced shoots.  相似文献

利用透射电镜和扫描电镜技术研究了中华绒螯蟹(Eriocheir sinensis)胚胎附着系统的结构及形成机制。中华绒螯蟹受精卵附着在雌性腹肢内肢的携卵刚毛上。该附着系统由三个连续部分组成:卵膜、卵柄和被膜,后者覆盖在携卵刚毛的绒毛上。研究结果显示:中华绒螯蟹的成熟胚胎由三层明显的卵膜组成,即E1、E2和E3层,但胚胎附着系统的卵柄及被膜仅为外层(E1)。卵巢中成熟卵的卵膜仅由E1层组成,E1分为两个亚层(E1a′、E1b′)。胚胎附着系统的形成与雌蟹的行为、腹肢粘液腺分泌的粘液、卵膜的超微结构及各层的变化有关。受精卵刚从生殖孔中排出时,卵膜(E1a′、E1b′)并不能直接粘附在携卵绒毛上。产卵后不久,雌蟹腹肢粘液腺分泌粘液的量增多,E1a′、E1b′的结构发生变化,表现为边界模糊,卵膜出现很强的粘性。在产卵后约60min E1层又明显分为两个亚层(E1a、E1b),同时排卵后雌蟹腹部的携卵绒毛不断地运动,这种运动促使携卵绒毛外的被膜形成。随着E1层亚结构的变化,E2层也开始形成,当E1新的两个亚层出现时,部分区域的E1层与E2层发生分离,卵柄开始形成,并牢固地附着在携卵绒毛上。被膜、卵柄与卵膜最外层的结构相同,均由E1层构成[动物学报51(5):852-861,2005]。  相似文献

Somites are embryonic precursors of the ribs, vertebrae and certain dermis tissue. Somite formation is a periodic process regulated by a molecular clock which drives cyclic expression of a number of clock genes in the presomitic mesoderm. To date the mechanism regulating the period of clock gene oscillations is unknown. Here we show that chick homologues of the Wnt pathway genes that oscillate in mouse do not cycle across the chick presomitic mesoderm. Strikingly we find that modifying Wnt signalling changes the period of Notch driven oscillations in both mouse and chick but these oscillations continue. We propose that the Wnt pathway is a conserved mechanism that is involved in regulating the period of cyclic gene oscillations in the presomitic mesoderm.  相似文献
