Availability, uptake and turnover of glycerol in hypersaline environments   总被引:4,自引:0,他引:4  
Abstract A sensitive assay for glycerol and other polyols was developed, based on periodate oxidation to formaldehyde, followed by a colorimetric assay with 3-methyl-2-benzothiazolone hydrazone. Apparent glycerol concentrations thus measured in saltern crystallizer ponds were around 20–36 μM, while in the Dead Sea, during a Dunaliella bloom, values were up to 27 μM. However, these values probably overestimate the glycerol concentrations present, as shown by labeled glycerol uptake experiments. Values of [K + Sn] (natural concentration + affinity constant) in saltern ponds were as low as 0.76–1.4 μM, with Vmax values of 193–303 nmol 1−1h−1, and turnover times between 2.6–7.2 h at 35°C. Similar measurements in the Dead Sea were: [K + Sn] 0.07–1.41 μM, Vmax values 160–426 nmol 1−1h−1, and turnover times in the range of 0.45–3.3 h.  相似文献

A new species of halophilic anoxygenic purple bacteria of the genus Rhodospirillum is described. The new organism, isolated from water/sediment of the Dead Sea, was vibrio-shaped and an obligate halophile. Growth was best at 12% NaCl, with only weak growth occurring at 6% or 21% NaCl. Growth occurred at Mg2+ concentrations up to 1 M but optimal growth was obtained at 0.05–0.1 M Mg2+. Bromide was well tolerated as an alternative anion to chloride. The new organism is an obligate phototroph, growing photoheterotrophically in media containing yeast extract and acetate or a few other organic compounds. Growth of the Dead Sea Rhodospirillum species under optimal culture conditions was slow (minimum td 20 h). Cells contained bacteriochlorophyll a and carotenoids of the spirilloxanthin series and mass cultures were pink in color. Absorption spectra revealed the presence of a B875 (light-harvesting I) but no B800/B850 (light-harvesting II) photopigment complex. The new organism shares a number of properties with the previously described halophilic phototrophic bacterium Rhodospirillum salinarum and was shown to be related to this phototroph by 16S rRNA sequencing. However, because of its salinity requirements, photosynthetic properties, and isolation from the Dead Sea, the new phototroph is proposed as a new species of the genus Rhodospirillum, R. sodomense.  相似文献

Age-related delays in bone repair remains an important clinical issue that can prolong pain and suffering. It is now well established that inflammation increases with aging and that this exacerbated inflammatory response can influence skeletal regeneration. Recently, simple dietary supplementation with beneficial probiotic bacteria has been shown to influence fracture repair in young mice. However, the contribution of the gut microbiota to age-related impairments in fracture healing remains unknown. Here, we sought to determine whether supplementation with a single beneficial probiotic species, Bifidobacterium longum (B. longum), would promote fracture repair in aged (18-month-old) female mice. We found that B. longum supplementation accelerated bony callus formation which improved mechanical properties of the fractured limb. We attribute these pro-regenerative effects of B. longum to preservation of intestinal barrier, dampened systemic inflammation, and maintenance of the microbiota community structure. Moreover, B. longum attenuated many of the fracture-induced systemic pathologies. Our study provides evidence that targeting the gut microbiota using simple dietary approaches can improve fracture healing outcomes and minimize systemic pathologies in the context of aging.  相似文献

The proximal half of a hominid femur was recovered from deep within a paleokarst feature at the Berg Aukas mine, northern Namibia. The femur is fully mineralized, but it is not possible to place it in geochrono logical context. It has a very large head, an exceptionally thick diaphyseal cortex, and a very low collodiaphyseal angle, which serve to differentiate it from Holocene homologues. The femur is not attributable to Australopithecus, Paranthropus, or early Homo (i.e., H. habilis sensu lato). Homo erectus femora have a relatively longer and AP flatter neck, and a shaft that exhibits less pilaster than the Berg Aukas specimen. Berg Aukas also differs from early modern femora in several features, including diaphyseal cortical thickness and the degree of subtrochanteric AP flattening. The massive diaphyseal cortex of Berg Aukas finds its closest similarity within archaic H. sapiens (e.g., Castel di Guido) and H. erectus (e.g., KNM-ER 736) samples. It has more cortical bone at midshaft than any other specimen, although relative cortical thickness and the asymmetry of its cross-sectional disposition at this level are comparable with those of other Pleistocene fem ora. The closest morphological comparisons with Berg Aukas are in archaic (i.e., Middle Pleistocene) H. sapiens and Neandertal samples. © 1995 Wiley-Liss, Inc.  相似文献

Bacteriorhodopsin in a bloom of halobacteria in the Dead Sea   总被引:1,自引:0,他引:1  
A dense bloom of red halobacteria developed in the Dead Sea in the summer 1980, bacterial densities of up to 1.9 x107 cells ml-1 were observed. The population consisted of two types: pleomorphic, cup-shaped cells and rod-shaped cells. A high content of bacteriorhodopsin was found in the bloom (up to 0.4 nmol per mg protein). Wood-pastures are fragile ecosystems because they were formed by, and depend on specific, low-intensity multifunctional management. Although their ecological and cultural significance is high, wood-pastures are rapidly deteriorating all over Europe, mainly due to changing land use. We still lack a basic understanding of the ecological value of wood-pastures, and in which features they differ from other landscape elements. In this paper we investigated the ecological value of wood-pastures for passerine birds by (i) comparing bird assemblages of wood-pastures with those of closed forests and open pastures and (ii) exploring the relationships between variables describing wood-pastures and species traits of the bird assemblages. Our study region (Southern Transylvania, Romania) provides a unique opportunity to understand the importance of a traditional cultural and ecological environment for many different organisms. Wood-pastures had a higher overall number of bird species, and a higher spatial turnover in bird community composition than closed forests and open pastures. We found significant associations between bird species traits and habitat structural elements in wood-pastures such as large trees, oak- and pear trees and shrubs. Our findings suggest that traditional wood-pastures in Southern Transylvania have distinct and rich passerine bird communities. This richness is inextricably linked to the multifunctional, low-intensity land use traditionally applied in the wood-pastures that promotes high niche diversity. For effective conservation of the biodiversity of wood-pastures, a detailed understanding is needed of how different management regimes may influence the key structural elements of wood-pastures relevant for biodiversity and these should be protected.  相似文献
