目的:评价针刺联合红外线照射治疗腰椎间盘突出症的临床疗效。方法:选择2010 年9 月~2012 年1 月我院收治的90 例 腰椎间盘突出症患者为研究对象,并将其随机分为对照组和治疗组,每组45 例。对照组患者给予药物治疗,而治疗组患者给予针 刺联合红外线照射治疗,治疗后评价和比较两组患者的临床疗效及腰腿痛的改善情况。结果:针刺联合红外线照射治疗腰椎间盘 突出症显效率为62.2%,药物组显效率为26.7%,两组有显著性差异(P<0.05)。治疗后,治疗组患者腰腿痛疼痛评分明显低于对照 组,差异有统计学意义(P<0.01)。结论:以针刺联合红外线照射治疗腰椎间盘突出症的临床疗效肯定,值得临床推广。  相似文献

乳腺癌是一种女性最常见的恶性肿瘤,近年来我国乳腺癌的发病率呈逐年上升趋势。研究证实,新辅助化疗不仅可使不能手术的晚期乳腺癌患者获得手术机会,而且也增加了部分患者的保乳概率,但是约20%的乳腺癌患者不能从新辅助化疗中获益,并影响后续治疗效果。因此,制定合理的、个体化的新辅助化疗方案对于提高乳腺癌患者的生存质量和改善其预后尤为重要。目前,研究已发现的NCT相关的预测因子主要包块肿瘤类型、分子分型、ER、PR、Ki67等,而HER2、TOP2A、P53、PRAP单独作为预测因子的还存在争议。本文主要对乳腺癌新辅助化疗的现状、各种预测因子的作用及其不足之处进行了综述。  相似文献

Cholesterol-dependent cytolysins (CDCs) are a large family of bacterial toxins that exhibit a dependence on the presence of membrane cholesterol in forming large pores in cell membranes. Significant changes in the three-dimensional structure of these toxins are necessary to convert the soluble monomeric protein into a membrane pore. We have determined the crystal structure of the archetypical member of the CDC family, streptolysin O (SLO), a virulence factor from Streptococcus pyogenes. The overall fold is similar to previously reported CDC structures, although the C-terminal domain is in a different orientation with respect to the rest of the molecule. Surprisingly, a signature stretch of CDC sequence called the undecapeptide motif, a key region involved in membrane recognition, adopts a very different structure in SLO to that of the well-characterized CDC perfringolysin O (PFO), although the sequences in this region are identical. An analysis reveals that, in PFO, there are complementary interactions between the motif and the rest of domain 4 that are lost in SLO. Molecular dynamics simulations suggest that the loss of a salt bridge in SLO and a cation–pi interaction are determining factors in the extended conformation of the motif, which in turn appears to result in a greater flexibility of the neighboring L1 loop that houses a cholesterol-sensing motif. These differences may explain the differing abilities of SLO and PFO to efficiently penetrate target cell membranes in the first step of toxin insertion into the membrane.  相似文献

Fucose metabolism pathways are present in many bacterial species and typically contain the central fucose-processing enzymes fucose isomerase (FcsI), fuculose kinase (FcsK), and fuculose-1-phosphate aldolase (FcsA). Fucose initially undergoes isomerization by FcsI producing fuculose, which is then phosphorylated by FcsK. FcsA cleaves the fuculose-1-phosphate product into lactaldehyde and dihydroxyacetone phosphate, which can be incorporated into central metabolism allowing the bacterium to use fucose as an energy source. Streptococcus pneumoniae has fucose-processing operons containing homologs of FcsI, FcsK, and FcsA; however, this bacterium appears unable to utilize fucose as an energy source. To investigate this contradiction, we performed biochemical and structural studies of the S. pneumoniae fucose-processing enzymes SpFcsI, SpFcsK, and SpFcsA. These enzymes are demonstrated to act in a sequential manner to ultimately produce dihydroxyacetone phosphate and have structural features entirely consistent with their observed biochemical activities. Analogous to the regulation of the Escherichia coli fucose utilization operon, fuculose-1-phosphate appears to act as an inducing molecule for activation of the S. pneumoniae fucose operon. Despite our evidence that S. pneumoniae appears to have the appropriate regulatory and biochemical machinery for fucose metabolism, we confirmed the inability of the S. pneumoniae TIGR4 strain to grow on fucose or on the H-disaccharide, which is the probable substrate of the transporter for the pathway. On the basis of these observations, we postulate that the S. pneumoniae fucose-processing pathway has a non-metabolic role in the interaction of this bacterium with its human host.  相似文献

Symmetry is an important feature of protein tertiary and quaternary structures that has been associated with protein folding, function, evolution, and stability. Its emergence and ensuing prevalence has been attributed to gene duplications, fusion events, and subsequent evolutionary drift in sequence. This process maintains structural similarity and is further supported by this study. To further investigate the question of how internal symmetry evolved, how symmetry and function are related, and the overall frequency of internal symmetry, we developed an algorithm, CE-Symm, to detect pseudo-symmetry within the tertiary structure of protein chains. Using a large manually curated benchmark of 1007 protein domains, we show that CE-Symm performs significantly better than previous approaches. We use CE-Symm to build a census of symmetry among domain superfamilies in SCOP and note that 18% of all superfamilies are pseudo-symmetric. Our results indicate that more domains are pseudo-symmetric than previously estimated. We establish a number of recurring types of symmetry–function relationships and describe several characteristic cases in detail. With the use of the Enzyme Commission classification, symmetry was found to be enriched in some enzyme classes but depleted in others. 硒对培养人胚肝细胞Ⅲ型前胶原,羟脯氨酸合成的影响   总被引:7,自引:0,他引:7  
原代培养人胚肝细胞经1.156×10 ̄(-7)mol/L硒预处理4h,加入20mmol/L四氟化碳作用20h,观察硒对其Ⅲ型前胶原(PCⅢ)和羟脯氨酸(Hyp)生成的影响。结果培养液中PCⅢ水平、细胞内Hyp含量及细胞内外丙二醛(MDA)水平均降低,与未加硒对照组比较差别有显著性(P<0.01)。而硒谷腕甘肽过氧化物酶(Se-GSH-PX)活性则较对照组显著增高(P<0.001),且PCⅢ水平与Se-GSH-P_X/MDA比值呈负相关(r=-0.9156,P<0.01)。提示硒可提高Se-GSH-P_X/MDA比值,抑制脂质过氧化激发的肝细胞胶原合成。  相似文献

The development of novel leishmanicidal agents that are capable of being replaced by the available therapeutic options has become a priority. In the present study, the synthesis and leishmanicidal activity of a series of 5-(nitroheteroaryl-2-yl)-1,3,4-thiadiazole derivatives are described. All compounds appeared to be potent anti-leishmanial agents against both promastigote and amastigote forms of Leishmania major (L. major). Amongst the synthesized compounds, 2-([1,4′-bipiperidin]-1′-yl)-5-(5-nitrofuran-2-yl)-1,3,4-thiadiazole (IIa) and 1-(5-(1-methyl-5-nitro-1H-imidazole-2-yl)-1,3,4-thiadiazol-2-yl)-4-(piperidine-1-yl) piperidine (IIc) are the most effective. Infection index was statistically declined in the presence of all compounds. The analysis of redox-related factors revealed that exposure of L. major cells to IIa and IIc led to an increase in reactive oxygen species (ROS). Furthermore, two compounds were able to increase ROS and NO levels in infected macrophages in a dose-independent manner. In addition, we showed that these compounds induced cell death in promastigotes. Altogether, our results indicated the anti-leishmanial potential of IIa and IIc is mediated by apoptosis through an imbalance in the redox system resulting in the elevation of ROS. This new class of compound seems to hold great promise for the development of new and useful anti-leishmanial agents.  相似文献

阿尔茨海默病(Alzheimer's disease,AD)是一种慢性进行性神经变性疾病,早期临床表现为近期记忆力下降,后来逐渐发展为生活不能自理最终死亡。目前,有许多种假说阐述其发病机制,如β-淀粉样蛋白、载脂蛋白E、一些金属元素、炎症因子等,但都不全面,也有人认为其可能与朊蛋白有关。部分药物被用于治疗AD,如美金刚、胆碱酯酶抑制剂等,但疗效均不显著。一些新的治疗理念和方法也逐渐被发现,如脑源性神经生长因子(brain-derived neurotrophic factor,BDNF)以及免疫疗法等,但其远期疗效还需进一步证实,且一些副作用还并未被人们所知晓。本文主要对阿尔茨海默病的发病机制及药物治疗的研究进展进行了综述。  相似文献

Analyses of knowledge gaps can highlight imbalances in research, encouraging greater proportionality in the distribution of research efforts. In this research we used generalized linear mixed models (GLMM) with the aim to determine if research efforts for the period 2005–2015 for terrestrial vertebrates of Amphibia, Aves, Mammalia and Reptilia in the South Pacific region were correlated with conservation status (critically endangered (CR), endangered (EN), vulnerable (VU), least concern (LC) and near threatened (NT)) or population trends (increasing, stable, decreasing and unknown) through the International Union for Conservation of Nature (IUCN) database. Our results showed that research distribution was uneven across different classes. Out of 633623 investigated papers, the average number of publications per species was 43.7, 306.7, 717.6 and 115.3 for Amphibia (284 species), Aves (1306 species), Mammalia (243 species) and Reptilia (400 species), respectively. Consistently, the lower publication effort on Amphibia compared to other taxonomic classes was revealed as significant by GLMM analysis. There was no significant differences in research effort among levels of conservation status. However, we found significantly different publication efforts among population trends of all examined species in that species with “unknown” population trends gained significantly lower researchers’ attention compared to species with “decreasing” trend. Results also indicated that, although it was not significant, the highest attention is given to species with “increasing” population trend over all taxonomic classes. Using the Information Theoretic approach we also generated a set of competing models to identify most important factors influencing research efforts, revealing that the highest ranked model included taxonomic class and population trend.  相似文献
