Spectroscopic and in silico study of binding mechanism of cynidine‐3‐O‐glucoside with human serum albumin and glycated human serum albumin 下载免费PDF全文
The drug–serum albumin interaction plays a dominant role in drug efficacy and disposition. The glycation of serum albumin that occurs during diabetes may affect its drug‐binding properties in vivo. In order to evaluate the interactivity characteristics of cyanidin‐3‐O‐glucoside (C3G) with human serum albumin (HSA) and glycated human serum albumin (gHSA), this study was undertaken using multiple spectroscopic techniques and molecular modeling analysis. Time‐resolved fluorescence and the thermodynamic parameters indicated that the quenching mechanism was static quenching, and hydrogen bonding and Van der Waals force were the main forces. The protein fluorescence could be quenched by C3G, whereas the polarity of the fluorophore was not obviously changed. C3G significantly altered the secondary structure of the proteins. Furthermore, the interaction force that existed in the HSA–C3G system was greater than that in the gHSA–C3G system. Fluorescence excitation emission matrix spectra, red edge excitation shift, Fourier transform infrared spectroscopy and circular dichroism spectra provided further evidence that glycation could inhibit the binding between C3G and proteins. In addition, molecular modeling analysis supported the experimental results. The results provided more details for the application of C3G in the treatment of diabetes. 相似文献

Removal of feral cats from island environments is a useful mechanism by which their ecological impact on endangered species can be reduced or ended. Nevertheless, because cats are anthropogenic in their origins, social perceptions of management practices play a large role in their implementation. Four-hundred questionnaires were delivered (386 were returned) with 100 going to each of the following: local residents; environmental workers; tourists; and, hunters. Questions explored respondents’ knowledge about island biodiversity and invasive species as well as attitudes towards cat population management methods. Habitat destruction and introduction of invasive species were considered the main threats for the conservation of island biodiversity. Most respondents considered cats to have a negative impact on biodiversity and sterilization campaigns were considered most appropriate for cat population control. Several free sterilization campaigns have been conducted in La Palma Island Biosphere Reserve in order to reduce free-ranging cats and were well received by local people. This research, which combined concepts of management, ecology and social sciences, provides valuable insights which may to be applicable on several other islands where cats and people are present and in conflict with conservation priorities. 相似文献

Relative abundance and within-lake distributions of three fishes, northern redbelly dace (Phoxinus eos), finescale dace (Phoxinus neogaeus), and central mudminnow (Umbra limi), were examined using minnow traps in Tuesday Lake, a small bog lake in the Upper Peninsula, Michigan. For these species, catches in minnow traps placed at the perimeter of the lake were 21 to 52 times higher than catches in midlake traps. Variance: mean ratios of perimeter trap catches indicated that both dace species were highly aggregated while the distribution of mudminnows was less aggregated or random. Over an 11 day period during which all fish caught were removed from the lake, catch per unit effort (CPUE) of both dace species declined in response to fish removal. In contrast, CPUE for mudminnows was low initially, increased to an asymptote and then declined only in the last 5 days of the fish removal. The patterns of CPUE for mudminnows indicated that mudminnow trapability and/or activity was reduced in the presence of high densities of dace. The low abundance of dace in traps with many mudminnows suggested mudminnows avoided traps already containing dace. Throughout the removal period, CPUE provided an accurate index of dace abundance, whereas this was true for mudmnnows only after dace populations had been reduced drastically. Therefore, in any use of minnow traps to estimate populations, both spatial distributions and relative species abundance of small fishes must be taken into account. 相似文献

Headwater species and peripheral populations that occupy habitat at the edge of a species range may hold an increased conservation value to managers due to their potential to maximize intraspecies diversity and species' adaptive capabilities in the context of rapid environmental change. The southern Appalachian Mountains are the southern extent of the geographic range of native Salvelinus fontinalis and naturalized Oncorhynchus mykiss and Salmo trutta in eastern North America. We predicted distributions of these peripheral, headwater wild trout populations at a fine scale to serve as a planning and management tool for resource managers to maximize resistance and resilience of these populations in the face of anthropogenic stressors. We developed correlative logistic regression models to predict occurrence of brook trout, rainbow trout, and brown trout for every interconfluence stream reach in the study area. A stream network was generated to capture a more consistent representation of headwater streams. Each of the final models had four significant metrics in common: stream order, fragmentation, precipitation, and land cover. Strahler stream order was found to be the most influential variable in two of the three final models and the second most influential variable in the other model. Greater than 70% presence accuracy was achieved for all three models. The underrepresentation of headwater streams in commonly used hydrography datasets is an important consideration that warrants close examination when forecasting headwater species distributions and range estimates. Additionally, it appears that a relative watershed position metric (e.g., stream order) is an important surrogate variable (even when elevation is included) for biotic interactions across the landscape in areas where headwater species distributions are influenced by topographical gradients. 相似文献

Synopsis Since 1952, a sport fishery for the blue shark,Prionace glauca, has existed off the south coast of Cornwall in England. Annual catches from this fishery have ranged from < 200 to>6000 sharks. The fishery was based on a previously unexploited stock in the 1950s. The abundance of the species in the English Channel declined in the early 1960s and again in the mid-1970s. The declining abundance was investigated in relation to sea surface temperature (SST), prey abundance, and fishing pressure. Short-term fluctuations in SST were found to be responsible for changes in the distribution of the population, but not for changes in abundance. The abundance of prey species in the Channel was observed to be inversely related to the abundance of blue sharks. The reduced abundance of blue sharks lowered the level of effective predation on the prey populations, allowing their abundance to increase. It was concluded that the nature of the fishing practice off the Cornish coast was responsible for a significant part of the decrease in shark abundance. By killing large numbers of sub-adult females, the reproductive capacity of the population was lowered. Continued fishing pressure prevented the population from recovering; and as of 1987, the abundance of the species in the Channel is still declining. Over recent decades wild rabbit populations have undergone a sharp decline in Spain with consequent negative impact both on
a game-based economy, and on the conservation of threatened species that depend on rabbits. We investigated the relationships
between rabbit population change and habitat and game management in central-southern Spain. To determine recent rabbit population
changes we revisited 60 localities during summer 2002 to repeat surveys previously carried out in 1993. Each survey consisted
of 4-km walked transects to record indices of rabbit abundance. The percentage of vegetation cover and of different soil types
were also estimated during these transects. In the same areas, the type and intensity of game management practices were obtained
by interviewing hunting managers, hunters or gamekeepers. Rabbit populations were stable or increasing only in 26.6% of the
studied populations, more commonly in areas with soft soils where warren building is easier for rabbits and where rabbits
were an important game species and managed to increase their numbers. Although we could not establish causality, habitat management
and predator removal were the main management practices related to rabbit population change. Rabbit scarcity in Spain constitutes
a serious problem for conservation, so hunters, researchers and policy makers need to reach a consensus to establish a long-term
program to monitor rabbit population trends and share results obtained, especially when intense manage programs are being

Objective: Imprint cytology provides a rapid preliminary diagnosis shortly after the completion of breast biopsy. This study aims to assess the validity of imprint cytology for the pre-operative diagnosis of non-palpable mammographic solid lesions excised by vacuum-assisted breast biopsy (VABB).
Methods: Seventy-two women with non-palpable Breast Imaging Reporting and Data System 3 and 4 mammographic solid lesions without microcalcifications underwent VABB on the stereotactic Fischer's table with 11-G Mammotome vacuum probes. Imprint samples were examined (Diff-Quick stain, modified Papanicolaou stain and May-Grünwald–Giemsa). The cores were dipped into a CytoRich Red Collection fluid for a few seconds in order to obtain samples with the use of the specimen wash. After the completion of cytological procedures, the core was prepared for routine pathological study. The pathologist was blind to the preliminary cytological results. The cytological and pathological diagnoses were comparatively evaluated.
Results: The sensitivity of the cytological imprints for cancer was 90%. The specificity of the method for cancer diagnosis was 100%. Two precursor lesions were present in the material: one case of atypical ductal hyperplasia, which was successfully detected, and one case of lobular neoplasia, which escaped detection. The cytological imprints were inadequate in four out of 72 cases (5.6%), but none of them were included within the malignant subgroup.
Conclusions: Imprint cytology seems to be an important adjunctive tool in the management of patients with non-palpable mammographic solid lesions. Its very satisfactory sensitivity and optimal specificity could establish its use in general clinical practice. 相似文献

Understanding the evolutionary dynamics of inbreeding and inbreeding depression requires unbiased estimation of inbreeding depression across diverse mating systems. However, studies estimating inbreeding depression often measure inbreeding with error, for example, based on pedigree data derived from observed parental behavior that ignore paternity error stemming from multiple mating. Such paternity error causes error in estimated coefficients of inbreeding (f) and reproductive success and could bias estimates of inbreeding depression. We used complete “apparent” pedigree data compiled from observed parental behavior and analogous “actual” pedigree data comprising genetic parentage to quantify effects of paternity error stemming from extra‐pair reproduction on estimates of f, reproductive success, and inbreeding depression in free‐living song sparrows (Melospiza melodia). Paternity error caused widespread error in estimates of f and male reproductive success, causing inbreeding depression in male and female annual and lifetime reproductive success and juvenile male survival to be substantially underestimated. Conversely, inbreeding depression in adult male survival tended to be overestimated when paternity error was ignored. Pedigree error stemming from extra‐pair reproduction therefore caused substantial and divergent bias in estimates of inbreeding depression that could bias tests of evolutionary theories regarding inbreeding and inbreeding depression and their links to variation in mating system. 相似文献

Conservation management of endangered African wild dogs (AWD; Lycaon pictus) can benefit greatly from development of sperm freezing and artificial insemination. Previous freezing attempts yielded nearly 0% motile sperm within 2 h of thawing. In this study, two canine freezing protocols were tested: Protocol 1: a one-step dilution in TRIS-20% egg yolk containing 8% glycerol; and Protocol 2: a two-step dilution in TRIS-20% egg yolk containing a final extender concentration of 5% glycerol and 0.5% Equex STM, coupled with a TRIS-citrate-fructose thawing solution. Semen was collected by electroejaculation from n = 24 AWDs, of which eight ejaculates of sufficient quality (four good quality with initial sperm motility of 75.0 ± 4.4% and four poor quality; showing rapid decrease in sperm motility to 3.3 ± 3.3% prior to freezing) were frozen. For good quality samples, motility and sperm motility index persisted for up to 8 h for Protocol 2, and was higher between 2 and 6 h after thawing with a decrease from 4 h of incubation. Motility dropped to nearly 0% after 2 h incubation for Protocol 1. Viability was higher for Protocol 2 throughout the 8 h of incubation, with a decrease after 6 h, compared to 4 h for Protocol 1. Acrosome integrity was higher for Protocol 2 throughout post-thaw incubation, with a decrease after 2 h for both protocols. Protocols did not differ in normal sperm morphology or DNA integrity. Poor quality samples yielded similar results, except for acrosome integrity, which declined for Protocol 2. In conclusion, a two-step dilution in TRIS-egg yolk-glycerol extender containing Equex STM yields significantly improved post-thaw quality and longevity of AWD spermatozoa, making it suitable for sperm banking and artificial insemination initiatives. 相似文献
