The inwardly-rectifying potassium channel subunit Kir5.1 selectively co-assembles with members of the Kir4.0 subfamily to form novel pH-sensitive heteromeric channels with unique single channel properties. In this study, we have cloned orthologs of Kir4.1 and Kir5.1 from the genome of the amphibian, Xenopus tropicalis (Xt). Heteromeric XtKir4.1/XtKir5.1 channels exhibit similar macroscopic current properties to rat Kir4.1/Kir5.1 with a faster time-dependent rate of activation. However, single channel analysis of heteromeric XtKir4.1/XtKir5.1 channels reveals that they have markedly different long-lived, multi-level subconductance states. Furthermore, we demonstrate that the XtKir5.1 subunit is responsible for these prominent subconductance levels. These results are consistent with a model in which the slow transitions between sublevel states represent the movement of individual subunits. These novel channels now provide an excellent model system to determine the structural basis of subconductance levels and contribution of heteromeric pore architecture to this process.  相似文献

The convergence of complementary and alternative medicine (CAM) and evidence-based medicine (EBM) is a prominent feature of healthcare in western countries, but it is currently undertheorised, and its implications have been insufficiently considered. Two models of convergence are described – the totally integrated evidence-based model (TI) and the multicultural-pluralistic model (MP). Both models are being incorporated into general medical practice. Against the background of the reasons for the increasing utilisation of CAM by the public and by general practitioners, TI-convergence is supported and MP-convergence is rejected. MP-convergence is epistemologically and clinically incoherent, and it cannot be regulated. It is also inconsistent with developments in the legal determination of the standard of care for both diagnosis/treatment and disclosure. These claims concerning MP-convergence are justified by the fact that science is not a member of the group of perspectives or world-views which postmodernism treats as equally valid, and this is especially important for healthcare.  相似文献

Agave tequilana Weber (Rigidae, Agavaceae), blue agave, is a native Mexican plant that has been associated with tequila since the 17th century. The tequila industry has matured over time and now has a geographical indication (Denominación de Origen; DOT). The tequila industry has grown substantially in the last 15 years (19.82% annual increase between 1995 and 2008), resulting in an increase in agave production and associated residue (leaves) and bagasse that can be used for second‐generation biofuels. At a time when the biofuel industry is undergoing unprecedented changes, with diversified demand and predictions of increased competitiveness, this paper presents a review of agave landraces that have been affected by tequila production but may be beneficial for a biofuel industry. Conventional botanical studies have revealed domestication syndromes in races related to blue agave (‘azul listado’, ‘sigüín’ and ‘pata de mula’) specifically for production of fructans in the plant core as would be expected in mezcal agaves (including those used for tequila). Some others, such as the ‘moraleño’ and ‘bermejo’ cultivars (Sisalanae) show domestication syndrome only in the fibers, while others, such as ‘chato,’A. americana L. subtilis (Americanae) show domestication syndrome in fructans and fibers and ‘zopilote,’A. rhodacantha (Rigidae) a relatively low domestication syndrome. No specimens of the cultivars named ‘mano larga’, ‘mano anchaque’ and ‘cucharo’ were found in the Tequila Region of Origin (Western Mexico). The genetic resources from landraces ignored by the tequila industry may be valuable for both ethanol production and conservation.  相似文献

Previous research has identified METCAM/MUC18, an integral membrane cell adhesion molecule (CAM) in the Ig-like gene super-family, as a promoter or a suppressor in the development of human breast cancer by MCF7, MDA-MB-231, and MDA-MB-468. To resolve these conflicting results we have investigated the role of this CAM in the progression of the three aforementioned cell lines plus one additional human breast cancer cell line, SK-BR-3. We transfected the SK-BR-3 cells with human METCAM/MUC18 cDNA to obtain G418-resistant clones, which expressed different levels of the protein and which were used to test the effect of human METCAM/MUC18 expression on in vitro motility, invasiveness, anchorage-independent colony formation in soft agar, disorganized growth in a 3D basement membrane culture assay, and in vivo tumorigenesis in athymic nude mice. Enforced METCAM/MUC18 expression increased in vitro motility, invasiveness, and anchorage-independent colony formation of SK-BR-3 cells and favored disorganized growth of the cells in 3D basement membrane culture. Enforced expression also increased tumorigenicity and final tumor weights of SK-BR-3 clones/cells after subcutaneous injection of the cells under the left third nipple of female athymic nude mice. To understand the mechanisms, we also determined the expression of several downstream key effectors in the tumors. Tumor cells from METCAM/MUC18 expressing clones exhibited elevated expression of an anti-apoptotic and survival index (Bcl2), an aerobic glycolysis index (LDH-A), and pro-angiogenesis indexes (VEGF and VAGFR2). We concluded that human METCAM/MUC18 promotes the development of breast cancer cells by increasing an anti-apoptosis and survival pathway and augmenting aerobic glycolysis and angiogenesis.  相似文献

Summary Sedum wrightii is one of only a few species in the Crassulaceae for which there is evidence for a high degree of variability in the ratio of daytime to nighttime CO2 assimilation. There are both environmental and genetic components to this variability. S. wrightii grows over a wide altitudinal gradient. The purpose of this study was to compare low, intermediate, and high altitude populations with respect to the degree of CAM expression and the capability to tolerate limited water availability. We utilized clonallyreplicated genotypes of plants from each population in common environment greenhouse experiments. Genetic differences among the populations were found in long-term water use efficiency, in 24 hour CO2 exchange patterns, in biomass 13C values, in carbon allocation, and in water status and ultimately survival during prolonged drought. The differences among the populations appear to be closely related to differences in the native habitats. The low altitude, desert plants had the greatest ability to grow and survive under conditions of limited water availability and appear to have the greatest shift to nighttime CO2 uptake during periods without water, while the high altitude plants had the poorest performance under these conditions and appear to shut down net carbon uptake when severely water limited.  相似文献

The studies described in the paper were conducted with tissue slices of Crassulacean acid metabolism (CAM) plants floating in isotonic buffer. In a first series of experiments, temperature effects on the efflux of [14C]malate and14CO2 were studied. An increase of temperature increased the efflux from the tissue in a non-linear manner. The efflux was markedly influenced also by the temperatures applied during the pretreatment. The rates of label export in response to the temperature and the relative contributions of14CO2 and [14C]malate to the label export were different in the two studied CAM plants (Kalanchoë daigremontiana, Sempervivum montanum). In further experiments, temperature response of the labelling patterns produced by14CO2 fixation and light and darkness were studied. In tissue which had accumulated malate (acidified state) an increase of temperature decreased the rates of dark CO2 fixation whilst the rates of CO2 fixation in light remained largely unaffected. An increase of temperature shifted the labelling patterns from a C4-type (malate being the mainly labelled compound) into a C3-type (label in carbohydrates). No such shift in the labelling patterns could be observed in the tissue which had depleted the previously stored malate (deacidified state). The results indicate that in the acidified tissue the increase of temperature increases the efflux of malate from the vacuole by changing the properties of the tonoplast. Guanine nucleotide exchange factors (GEFs) are essential for small G proteins to activate their downstream signaling pathways, which are involved in morphogenesis, cell adhesion, and migration. Mutants of Gef26, a PDZ-GEF (PDZ domain-containing guanine nucleotide exchange factor) in Drosophila, exhibit strong defects in wings, eyes, and the reproductive and nervous systems. However, the precise roles of Gef26 in development remain unclear. In the present study, we analyzed the role of Gef26 in synaptic development and function. We identified significant decreases in bouton number and branch length at larval neuromuscular junctions (NMJs) in Gef26 mutants, and these defects were fully rescued by restoring Gef26 expression, indicating that Gef26 plays an important role in NMJ morphogenesis. In addition to the observed defects in NMJ morphology, electrophysiological analyses revealed functional defects at NMJs, and locomotor deficiency appeared in Gef26 mutant larvae. Furthermore, Gef26 regulated NMJ morphogenesis by regulating the level of synaptic Fasciclin II (FasII), a well-studied cell adhesion molecule that functions in NMJ development and remodeling. Finally, our data demonstrate that Gef26-specific small G protein Rap1 worked downstream of Gef26 to regulate the level of FasII at NMJs, possibly through a βPS integrin-mediated signaling pathway. Taken together, our findings define a novel role of Gef26 in regulating NMJ development and function.  相似文献

In many cases, an increase in the surface density of cell adhesion molecules (CAM) in the distal parts of a growing neurite is favorable for the neurite elongation. This increase is attained by exocytotic insertion of CAM-containing vesicles into the growth cones with subsequent redistribution of CAM along the cell surface due to lateral diffusion and endocytosis. Using a mathematical model describing these processes, we quantitatively describe conditions providing two qualitatively different profiles in a branching neurite: (i) the CAM surface density increases along both daughter branches, which would be in favor of further outgrowth of both branches, i.e., successful branching, or (ii) the CAM surface density increases along one daughter branch and decreases along another branch, which could lead to the retraction of the latter. The geometric factors and mechanisms underlying the intracellular CAM transport to the daughter growth cones were proved to determine the profile of CAM surface density. A similarity in the diameters of daughter branches, their short lengths, a high value of the lateral transfer constant, and partitioning of CAM transport at the branching point proportionally to the surface areas of daughter branches are in favor of an increase in the CAM surface density along both daughter branches. Asymmetric branching can lead to a decrease in the CAM surface density along the thinner or thicker daughter branch, if CAM trafficking was equally partitioned or was proportional to the branch cross-sectional areas, respectively. The proposed model helps to understand possible relationships between the intracellular CAM trafficking, CAM surface distribution, and geometry of branching of the neurites.  相似文献
