The ability of PVM, PVY and PVX viruses and their progeny to distribute themselves and accumulate in primary infected potato plants by mono‐ and mixed infections is analysed. It is shown, that the transport and accumulation of virus in inoculated potato plants depends on variety resistance, combination of viruses and sequence of their application.  相似文献

Latterly, the upsurge in use of antifungal drugs has brought about the emergence of several drug-resistance strains, making it skeptical to continue relying on current therapeutic regime. In the necessity of resistance-free antifungal agent, flavonoids presented possibilities of replacing existing drugs, displaying antifungal activity against pathogenic fungi. Among them, quercetin, one of the most representative flavonoids, exhibited antifungal activity against Candida albicans. To inspect the further understanding regarding quercetin, the antifungal mode of action of quercetin was investigated. In the initial step, the apoptosis was monitored after quercetin treatment. Moreover, intracellular levels of Mg2+ was assessed and was determined that Mg2+ increase occurred under the influence of quercetin. In addition, several features of mitochondrial dysfunction were monitored. Mitochondrial dysfunction triggers decrease in mitochondrial redox levels and leads to disruption in mitochondrial antioxidant system. Increased intracellular ROS and decreased intracellular redox levels were also displayed, indicating the occurrence of overall disruption in antioxidant systems. Sequentially, DNA fragmentation was observed and this DNA damage in turn induces apoptosis. In analyses, hexaamminecobalt(III) chloride (Cohex) was applied to inhibit Mg2+ transport between cytosol and mitochondria. Cohex attenuated the effects induced by quercetin, which demonstrates that the presence of Mg2+ is essential in quercetin-induced apoptosis.  相似文献

Along-shore currents can propagate harmful algal blooms (HABs) over long distances in many coastal areas of the ocean. Harmful dinoflagellate blooms on the west coast of Iberia frequently occur when the Iberian poleward current (IPC) establishes on the continental slope. This has led to the suggestion that HABs could be transported northward by the IPC. To examine this possibility, the microplankton composition along the west coast of Iberia was studied in May 1993 coinciding with the presence of the IPC. The microplankton of the IPC was almost exclusively composed of small flagellates, with the notable absence of the harmful species usually associated with coastal waters. The primary influence of the IPC was to confine coastal microplankton populations to the shelf, where a downwelling convergence prevented their export from the coastal environment. Microplankton assemblages on the shelf revealed a north–south gradient related to different stages of succession. Earlier stages of succession in which diatoms were prominent were found on the northern shelf, whereas dinoflagellates were more abundant in the south. The toxic species Gymnodinium catenatum, which was only present in the southern shelf, did not show a northward transport associated with the IPC. It is suggested that the northward spreading of HABs along the west coast of Iberia must be related to the interaction between the IPC, which accumulates coastal populations on the shelf, and the latitudinal progress of microplankton succession that determines species composition. Thus, during the course of the season, HABs are likely to be observed in the south prior to their development in the north.  相似文献

This study investigates ciguatoxin dynamics in mullet after controlled feeding of Gambierdiscus polynesiensis cells as a model to characterize the absorption, distribution, retention and accumulation of ciguatoxins into the second trophic level of southwestern Pacific coral reef ecosystems. Mullet (Mugil cephalus) were fed once every other day over a period of 16 days for nine toxic feedings, and ciguatoxin activity was assessed over time in blood and seven tissues using the Neuro2a assay. Within 3 h of feeding on G. polynesiensis cells, ciguatoxins attained maximal blood concentrations, indicating rapid absorption of toxins into the systemic circulation. The time course for distribution of the estimated total tissue burden of ciguatoxin closely followed the time course for blood toxin levels, indicating a rapid distribution of the ciguatoxins throughout the fish body. The large majority (95%) of the ciguatoxin ingested dose was eliminated from the examined fish tissues 24 h after a single toxic meal, indicating little retention potential for ciguatoxin. We found no evidence for ciguatoxin accumulation after nine repeated feedings spaced two days apart, indicating that mullet did not accumulate ciguatoxin. These results provide the first experimental evidence supporting the central tenet of Randall's food chain hypothesis that ciguatoxins enter the food chain by transfer from unicellular algae to herbivorous and detritus-feeding fish. We propose that a time-dependent transformation of oxopene ciguatoxins may be necessary for the concentration of ciguatoxin through higher trophic levels.  相似文献

  • The Cadmium (Cd)‐polluted soils are is an increasing concern worldwide. Phytoextraction of Cd pollutants by high biomass plants, such as sweet sorghum, is considered an environmentally‐friendly, cost‐effective and sustainable strategy for remediating this problem. Nitrogen (N) is a macronutrient essential for plant growth, development and stress resistance. Nevertheless, how nitrate, as an important form of N, affects Cd uptake, translocation and accumulation in sweet sorghum is still unclear.
  • In the present study, a series of nitrate levels (N1, 0.5 mm ; N2, 2 mm ; N3, 4 mm ; N4, 8 mm and N5, 16 mm ) with or without added 5 μm CdCl2 treatment in sweet sorghum was investigated hydroponically.
  • The results indicate that Cd accumulation in the aboveground parts of sweet sorghum was enhanced by optimum nitrate supply, resulting from both increased dry weight and Cd concentration. Although root‐to‐shoot Cd translocation was not enhanced by increased nitrate, some Cd was transferred from cell walls to vacuoles in leaves. Intriguingly, expression levels of Cd uptake and transport genes, SbNramp1, SbNramp5 and SbHMA3, were not closely related to increased Cd as affected by nitrate supply. The expression of SbNRT1.1B in relation to nitrate transport showed an inverted ‘U’ shape with increasing nitrate levels under Cd stress, which was in agreement with trends in Cd concentration changes in aboveground tissues.
  • Based on the aforementioned results, nitrate might regulate Cd uptake and accumulation through expression of SbNRT1.1B rather than SbNramp1, SbNramp5 or SbHMA3, the well‐documented genes related to Cd uptake and transport in sweet sorghum.

根据室内饲养的3头长江江豚(Neophocaena phocaenoides asiaeorientalis)食物鲫(Carassius auratus)中锌(Zn)、铜 (Cu)、铅(Pb)、镉(Cd)、砷(As)的浓度值和饲养记录,推算出了饲养条件下江豚这些微量元素每日及每周估计摄入 量的范围。必需元素的需求方面与世界卫生组织/联合国粮农组织(WHO/FAO)提出的人暂定每周耐受摄人量 (PTWI)相类似,可是毒性较强的元素Cd、As、Pb的摄入量大大高于人体的PTWI。对湖北天鹅洲故道收集到的一头 江豚的10种组织、器官的研究发现,摄入后的Zn、Cu、Pb、Cd、As显示出有组织、器官选择性累积的倾向。这可能主 要与鲸类组织、器官对相应的元素有特殊的要求相关。本研究的结果还显示出有必要建立饲养鲸类动物食物安全 管理规范以控制食源污染。更深入地研究各组织、器官中微量元素与生命机能的关系,应该是鲸类动物微量元素 的营养化学和生态毒理学发展的重要方向之一。  相似文献

When compiling an inventory of hyperdiverse taxa, it is impossible to record the total number of species during fieldwork. To ensure the accuracy of species-richness data it is necessary to assess the reliability of inventories. Accumulation curves are an easy method for doing this and are extensively described in the literature. In this study, we compare the relative fit of various models of species-accumulation functions for six local butterfly inventories, evaluating them by a consideration of the values of the fit, coefficient of determination and sum-of-squares, and the residual patterns and Akaike’s Information Criterion. In general, complex functions, such as the Weibull or Chapman-Richards, performed better than simpler and more widely used models (e.g., the Clench and negative exponential models). The performance of models varied among sampling plots, indicating the influence of factors such as land use and community structure. Thus, although the application of more complex models should replace the use of simple ones, further research into the factors affecting model fit of accumulation functions is necessary.  相似文献

One-step conversion of aniline, phenol and some of their monochlorinated derivatives into the corresponding catechols by resting pre-adapted cells of the Rhodococcus mutant strain AM 144 (defective in synthesis of catechol 1,2-dioxygenase) was shown to depend on the availability of an additional metabolizable carbon substrate, e.g. glucose or acetate. A stoichiometric relation existed between the amount of the latter compounds added and the amount of aniline (or phenol, respectively) converted into catechol suggesting that the primary function of the cosubstrates was to provide reducing power to the oxygenative transformation reaction. The observed cosubstrate-dependence generally parallels that seen in previous studies on turnover of different monochloroaromatic non-growth substrates by aromatics-utilizing Rhodococcus wildtype-strains. Cell cultures of strain AM 144 growing at the expense of acetate also proved able to convert aniline into catechol. Typically, growth of the cells was retarded during the phase of aniline transformation as compared to the respective control cultures. Based on the results of these model experiments, it was concluded that (i) in natural microbial communities cometabolically active bacteria would hardly enrich under cometabolic conditions over fast-growing non-cometabolizing bacteria if the latter organisms will tolerate the particular non-growth substrate, and (ii) cometabolizing bacteria would have a selective advantage only if the non-growth substrate to be transformed is a toxic one or if it can serve as a potential nutrient source (e.g., of nitrogen or sulfur).Abbreviations MCA monochloroaniline - MCP monochlorophenol - MCC monochlorocatechol - TLC thin-layer chromatography - MS mass spectrometry - GLC gas-liquid chromatography - UV ultraviolet (range of the spectrum)  相似文献

应用石蜡切片法、荧光显微镜和紫外分光光度法,对不同年生巴戟天根组织结构的变化进行了观察、对蒽醌类化合物在根中的分布场所及其积累动态进行了研究。结果表明:巴戟天根的结构类似一般多年生草本植物,薄壁细胞是巴戟天根中蒽醌类化合物的分布储存场所,蒽醌类化合物含量随着根生长年限的增加而增加。由以上研究总结出巴戟天以四年或四年以上采收为好,并以根皮厚、木心细者为上品。  相似文献

Rapid and reliable estimation of population size is needed for the efficient monitoring of animal populations of conservation concern. Unfortunately, technical advances in this area have not been paralleled in uptake in conservation, which may be due to difficulties in implementation or the lack of general guidelines for application. Here we tested five different methods used to estimate population size [capture–mark–recapture (CMR), finite-mixture models, model averaging of finite-mixture models, accumulation curve methods (ACM), and the line transect method (LT)] using extensive capture–recapture data of the giant day gecko (Gekkonidae, Phelsuma madagascariensis grandis, Gray 1870) at the Masoala rainforest exhibit, Zurich Zoo. When the complete data were analyzed [30 sessions (and 27 sessions for the LT)], all methods except the LT produced similar estimates of population size. The simple ACM gave a small coefficient of variation (CV), but did not cover the most likely value of population size at moderate sampling effort. Nevertheless, the ACM was the only method that showed a reasonable convergence when subsets of data were used. CMR and Pledger models included the reference value in their confidence intervals (CI) after 25 and 30 sessions, respectively. Although model averaging did slightly improve the estimate, the CV was still high for the full dataset. Our method of using subsets of data to test the robustness of estimates is simple to apply and could be adopted more widely in such analyzes to evaluate sensitivity to method of evaluation. In conclusion, simple accumulation methods showed similar efficiency to more complex statistical models, and are likely to be sufficiently precise for most conservation monitoring purposes.
