采用PCR-SSCP方法检测猪胰岛素样生长因子2 (insulin-like growth factor 2,IGF2)基因外显子3多态性,分析其对初生重、断奶重、6月龄重和背膘厚的遗传效应.根据猪IGF2基因的DNA序列(AY044828)设计引物,结果在其扩增片段上检测到多态性,对纯合子进行测序,发现IGF2-ex3-A36T和IGF2-ex3-G109A两个多态性位点,并且这2个多态性位点完全连锁,检测到3种基因型(A36A/G109G,A36T/G109A 和 T36T/A109A).统计结果表明,基因型在各品种中分布不一致,长白猪和大白猪与莱芜猪、大薄莲猪、沂蒙黑猪和里岔黑猪比较基因型分布差异极显著(P<0.01);其它猪种间基因型分布差异均不显著(P>0.05).固定效应模型分析结果表明,初生重和背膘厚基因型间差异显著(P<0.05),而断奶重和6月龄重基因型间差异不显著(P>0.05).最小二乘分析结果表明,A36A/G109G基因型个体同A36T/G109A和 T36T/A109A基因型个体比较初生重和背膘厚的差异显著(P<0.05),3种基因型初生重的大小排列顺序为A36A/G109G < A36T/G109A < T36T/A109A,背膘厚的大小排列顺序为A36A/G109G > A36T/G109A >T36T/A109A.因此,推测IGF2基因对个体的初生重和胴体瘦肉率存在一定的影响,将IGF2基因应用于猪育种过程中的标记辅助选择,将可以改善猪肉品质,加快猪的育种进程.  相似文献

In Australia, in the past, pasture legumes were rotated mainly with cereals, but increasingly these rotations now involve pasture legumes with a wider range of crops, including legumes. This increasing frequency of the leguminous host in the rotation system may be associated with increased root rots in legumes in the current pasture-crop rotations. The primary aim of this study was to see whether the pathogenicity on pasture legumes of strains of Rhizoctonia solani sourced from lupins and cereals (common crops in rotation with pastures) is associated with increased incidence of root rots in pasture legumes in the disease conducive sandy soils of the Mediterranean regions of southern Australia. The second aim was to determine sources of resistance among newly introduced pasture legumes to R. solani strains originating from rotational crops as this would reduce the impact of disease in the pasture phase. Fifteen pasture legume genotypes were assessed for their resistance/susceptibility to five different zymogram groups (ZG) of the root rot pathogen R. solani under glasshouse conditions. Of the R. solani groups tested, ZG1–5 and ZG1–4 (both known to be pathogenic on cereals and legumes) overall, caused the most severe root disease across the genotypes tested, significantly more than ZG6 (known to be pathogenic on legumes), in turn significantly >ZG4 (known to be pathogenic on legumes) which in turn was >ZG11 (known to be pathogenic on legumes including tropical species). Overall, Ornithopus sativus Brot. cvs Cadiz and Margurita, Trifolium michelianum Savi. cvs Paradana and Frontier and T. purpureum Loisel. cv. Electro showed a significant level of resistance to root rot caused by R. solani ZG11 (root disease scores ≤1.2 on a 1–3 scale where 3 = maximum disease severity) while O. sativus cvs Cadiz and Erica showed a significant level of resistance to root rot caused by R. solani ZG4 (scores ≤1.2). O. compressus L. cvs Charano and Frontier, O. sativus cv. Erica, and T. purpureum cv. Electro showed some useful resistance to root rot caused by R. solani ZG6 (scores ≤1.8). This is the first time that cvs Cadiz, Electro, Frontier, Margurita and Paradana have been recognised for their levels of resistance to root rot caused by R. solani ZG11; and similarly for cvs Cadiz and Erica against ZG4; and for cvs Charano, Erica, and Electro against ZG6. These genotypes with resistance may also serve as useful sources of resistance in pasture legume breeding programs and also could potentially be exploited directly into areas where other rotation crops are affected by these R. solani strains. None of the tested genotypes showed useful resistance to R. solani ZG1–4 (scores ≥2.0) or ZG1–5 (scores ≥2.5). This study demonstrates the relative potential of the various R. solani ZG strains, and particularly ZG1–4, ZG1–5, ZG4 and ZG6 to attack legume pastures and pose a significant threat to non-pasture crop species susceptible to these strains grown in rotation with these pasture legumes. Significantly, the cross-pathogenicity of these strains could result in the continuous build-up of inoculum of these strains that may seriously affect the productivity eventually of legumes in all rotations. In particular, when choosing pasture legumes as rotation crops, caution needs to be exercised so that the cultivars deployed are those with the best resistance to the R. solani ZGs most likely to be prevalent at the location.  相似文献

Methods are described for rearing large quantities of Ditylenchus dipsaci on alfalfa tissues. Nematodes and alfalfa seed were disinfected and nematodes were reared in quantities sufficient to provide a continuous supply of inoculum for our alfalfa-breeding program. Nematodes reproduced best in darkness at 20-25 C. Cultures reached maximum numbers in 3-6 wk.  相似文献

Genetic variability in 10 natural Tunisian populations of Medicago laciniata were analysed using 19 quantitative traits and 12 polymorphic microsatellite loci. A large degree of genetic variability within-populations and among-populations was detected for both quantitative characters and molecular markers. High genetic differentiation among populations for quantitative traits was seen, with Q ST = 0.47, and F ST = 0.47 for microsatellite markers. Several quantitative traits displayed no statistical difference in the levels of Q ST and F ST . Further, significant correlations between quantitative traits and eco-geographical factors suggest that divergence in the traits among populations may track environmental differences. There was no significant correlation between genetic variability at quantitative traits and microsatellite markers within populations. The site-of-origin of eco-geographical factors explain between 18.13% and 23.40% of genetic variance among populations at quantitative traits and microsatellite markers, respectively. The environmental factors that most influence variation in measured traits among populations are assimilated phosphorus (P205) and mean annual rainfall, followed by climate and soil texture, altitude and organic matter. Significant associations between eco-geographical factors and gene diversity, H e , were established in five-microsatellite loci suggesting that these simple sequence repeats (SSRs) are not necessarily biologically neutral.  相似文献

Seeds were collected from plants of Medicago sativa var. local inoculated with Glomus macrocarpum and G. fasciculatum separately in pot experiments. These seeds were sown in garden soil and the percentage germination, general health and yield of subsequent plants (the F1 generation) were studied. The percentage germination was highest in seeds of G. macrocarpum-inoculated parents followed by those inoculated with G. fasciculatum; seeds of uninoculated parent plants showed the lowest germination. Vegetative yield of the progeny decreased in the order of plants inoculated with G. fasciculatum, with G. macrocarpum, and uninoculated. On the other hand, reproductive yield was highest for plants whose parents were inoculated with G. macrocarpum, followed by G. fascicullatum, and lowest for seeds of uninoculated parent plants.  相似文献

Summary An optimum 10-day exposure of petioles of alfalfa [Medicago sativa ssp.falcata (L.) Arcangeli] to 2,4-dichlorophenoxyacetic acid or 2,4,5-trichlorophenoxyacetic acid results in the semisynchronous production of somatic embryos starting about 4 days after transfer to a non-auxin-containing medium. The timing of cell division induction in the petiole explants was found to vary depending on the petiole tissue type. Cells adjacent to the vascular bundles divide first at about 48 h after exposure to auxin, closely followed by those of the inner parenchyma, whereas most of the cells of the subepidermal and epidermal layers start to divide later, between 72 and 120 h. Two different sources of callus were also evident. Cells adjacent to the vascular bundles and the inner parenchyma cells were the primary source of callus when a short, 2-day (non-embryo-producing) exposure to auxin was used. In contrast, the subepidermal and epidermal cells were the primary source of callus tissue when a longer, 10-day (embryo producing) exposure was used. It is concluded that the source of somatic embryos is primarily the daughter cells of the subepidermal or epidermal tissue or both.  相似文献

Growth and nutrient utilization of alfalfa (Medicago sativa L. cv. Arc) and common bean (Phaseolus vulgaris L. cv. Carioca) were studied in an acid soil adjusted to eight levels of soil acidity by lime addition. Application of lime significantly (P<0.05) increased shoot and root growth for both species. However, common bean was far less sensitive to soil acidity than alfalfa. Maximum alfalfa growth was obtained at a soil pH of 5.8 and maximum bean growth was achieved at pH 5.0. Root and shoot growth of both legumes was positively correlated (P<0.01) with soil pH, exchangeable Ca and exchangeable Mg and negatively correlated (P<0.01) with soil exchangeable Al. Common bean had a lower internal P requirement for maximum growth and was more efficient than alfalfa in taking up Ca and Mg. These characteristics would contribute to the favorable growth of common bean in acid-infertile soils.  相似文献

This study explores methods to use information gathered from genomics technology to understand evolutionary relationships in the hyperdiverse legume group Neo-Astragalus. These species inhabit deserts and mountains of North and South America, and even though the monophyly of the group is well established, relationships within it are still poorly understood. Plastid genes, commonly used to infer phylogenies in plants, are usually not useful for closely related taxa because of low levels of genetic variation. TheMedicago truncatula genome project provided a suite of candidate nuclear loci with high levels of variation that might prove suitable for low-level phylogenetics. This paper reports the development of methods for screening a large number of these nuclear loci, and detailed analysis of four of them. Four different patterns of phylogenetic diversification occur in the loci sampled from these genomes ofAstragalus species. One locus (CNGC4) was single copy and could be directly used in phylogenetic analyses. Two loci (ARG10 and FENR) showed patterns strongly suggestive of duplication events in some taxa, and one locus (tRALS) has apparently undergone a cryptic duplication, making it very difficult to diagnose. Potential methods for using the information provided by these loci are discussed.  相似文献

通过室内生长箱内盆栽实验,比较了苜蓿和沙打旺苗期的根冠生长和水分利用对5种土壤水分环境变化的响应和差异.结果表明,充分供水下苜蓿和沙打旺苗期生物量和蒸腾效率均最高,苜蓿均显著高于沙打旺.土壤水分减少后苜蓿苗期生物量和蒸腾效率下降幅度均大于沙打旺.从低水到阶段低水处理后土壤水分逐渐降低和降低后再复水到低水处理,苜蓿和沙打旺的生物量分别较持续低水处理显著减少47.8%和27.9%.旱后复水后苜蓿根冠比和单位根量耗水量较显著增加,蒸腾效率显著下降; 沙打旺根冠比显著下降,单位根量耗水量和蒸腾效率变化不显著.  相似文献

以陕西省神木县六道沟小流域为研究区域,探讨了黄土高原北部紫花苜蓿人工草地退化过程中植物多样性的变化.结果表明,在黄土高原北部森林草原气候区,紫花苜蓿人工草地的退化演替过程可以划分为3个阶段:紫花苜蓿草地阶段(1~6年)、人工草地向天然草地演替的过渡阶段(6~10年)与长芒草次生天然草地阶段(10年以上).在1~30年的演替过程中,调查样方内共出现高等植物32种,分属于13科28属,其中90%以上出现在前6年.植被演替过程中累积出现的植物科属种数的动态变化可以用对数函数进行较好地描述.在群落演替过程中,物种丰富度指数、多样性指数和Pielou均匀度指数的变化趋势基本一致,在演替前期(第1个阶段)增加较快,在第2个阶段(过渡阶段)达到最高,然后有所降低并渐趋稳定.种植紫花苜蓿可显著地加快植被的自然演替进程,这与人工草地水分消耗强烈加速了土壤的旱化过程密切相关.发展人工草地是黄土高原北部加速天然植被恢复与发展畜牧业、增加农民收入的有机结合点,是西部生态环境建设中生态效益与经济效益兼顾的良好范式.  相似文献
