The quality of milk products is threatened by the formation of biofilms of thermophilicstreptococci on the internal surfaces of plate heat exchangers used in milk processing. Althoughattachment to stainless steel surfaces is one of the first stages in the development of a biofilm, themechanisms involved in attachment have not been reported. The cell surface properties of 12strains of thermophilic streptococci were examined to determine their importance in attachment tostainless steel surfaces. Hydrophobicity, extracellular polysaccharide production and cell surfacecharge varied between the different strains but could not be related to numbers attaching. Treatingthe cells with sodium metaperiodate, lysozyme or trichloroacetic acid to disrupt cell surfacepolysaccharide had no effect on attachment. Treatment with trypsin or sodium dodecyl sulphate toremove cell surface proteins resulted in a 100-fold reduction in the number of bacteria attaching.This result suggests that the surface proteins of the thermophilic streptococci are important intheir attachment to stainless steel.  相似文献

A differential screening study using high-resolution (HR)-hydrophilic interaction chromatography (HILIC)-electrospray ionization (ESI)–quadrupole time-of-flight mass spectrometry (Q-TOF MS) was conducted to identify saxitoxin (STX) analogues in the marine dinoflagellate toxic sub-clone Alexandrium tamarense Axat-2 and the non-toxic sub-clone UAT-014-009 derived from the same Japanese isolate. One unknown compound was identified only in the toxic sub-clone and was found to have the molecular formula C9H16N6O2. This structure differed from that of decarbamoyl STX (dcSTX; C9H16N6O3) by the loss of a single oxygen. A 12-deoxy-dcSTX standard (a mixture of 12α- and β-deoxy-dcSTX) was chemically prepared from dcSTX by reduction with sodium borohydride. The unknown compound in the toxic strain of A. tamarense was identified as 12β-deoxy-dcSTX by comparison of its HR-HILIC-LC–MS retention time and HR–MS/MS spectrum with those of the chemically prepared standard, and the identification was confirmed by high-sensitivity HPLC analysis with post-column fluorescent derivatization. Moreover, two Japanese isolates of A. catenella showing toxin profiles different from that of A. tamarense were also found to contain 12β-deoxy-dcSTX. Previously, 12β-deoxy-dcSTX was isolated from the freshwater cyanobacterium Lyngbya wollei, which produces a unique set of STX analogues. This study is the first evidence of the presence of 12β-deoxy-dcSTX in marine dinoflagellates.  相似文献

Azaspiracids (AZA) are a group of food poisoning phycotoxins that are known to accumulate in shellfish. They are produced by some species of the planktonic dinophycean taxon Amphidomataceae. Azaspiracids have been first discovered in Ireland but are now reported in shellfish from numerous global sites thus showing a wide distribution. In shellfish samples collected in 2009 near Huelva (Spain), AZA was also found along the Andalusian Atlantic coast for the first time. Analysis using LC–MS/MS revealed the presence of two different AZA analogues in different bivalve shellfish species (Chamelea gallina, Cerastoderma edule, Donax trunculus, and Solen vagina). In a number of samples, AZA levels exceeded the EU regulatory level of 160 μg AZA-1 eq. kg−1 (reaching maximum levels of >500 μg AZA-1 eq. kg−1 in Chamelea gallina and >250 μg AZA-1 eq. kg−1 in Donax trunculus) causing closures of some local shellfish production areas. One dinophyte strain established from the local plankton during the AZA contamination period and determined as Amphidoma languida was in fact toxigenic, and its AZA profile disclosed it as the causative species: it contained AZA-2 as the main compound and the new compound AZA-43 initially detected in the shellfish. AZA-43 had the same mass as AZA-3, but produced different collision induced dissociation (CID) spectra. High resolution mass spectrometric measurements indicated that there is an unsaturation in the H, I ring system of AZA-43 distinguishing it from the classical AZA such as AZA-1, -2, and -3. Furthermore, the Spanish strain was different from the previously reported AZA profile of the species that consist of AZA-38 and AZ-39. In molecular phylogenetics, the Andalusian strain formed a monophyletic group together with other strains of Am. languida, but ITS sequences data revealed surprisingly high intragenomic variability. The first Andalusian case of AZA contamination of shellfish above the EU regulatory limit reported here clearly revealed the risk of azaspiracid poisoning (AZP) for this area and also for the Atlantic coast of Iberia and North Africa. The present study underlines the need for continuous monitoring of AZA and the organisms producing such toxins.  相似文献

Summary This study was undertaken to investigate thermal adaptations in muscle contractile properties in closely-related lizards with different preferred body temperatures (PBT). The species examined all belong to theSphenomorphus group of Australian skinks (Scincidae: Lygosominae). Preferred body temperatures are conservative at the generic level as follows:Ctenotus, 35°C;Sphenomorphus, 30°C;Eremiascincus, 25°C. Contractile properties of the fast glycolytic portion of the iliofibularis muscle were measured. Translational adaptations are evident in several isometric factors, including tetanic tension (Po), twitch tension (Pt), twitch time to peak tension (TPT), and twitch half-relaxation time (1/2 RT). Capacity adaptations are not evident in rates of tetanic tension development (dPo/dt) or in maximal velocities of isotonic shortening (V max). Rotational adaptations are not evident in any contractile properties. Thermal limits on upper response temperatures are about 5°C warmer inCtenotus than in the more cryophilic species, indicative of resistance adaptation in muscle performance. Methanogenesis by a Syntrophomonas wolfei/ Methanospirillum hungatei coculture was inhibited in presence of ethylene and the hydrogenation catalyst Pd-BaSO4. However, butyrate oxidation by S. wolfei continued and ethylene was reduced to ethane. Per mol of butyrate oxidized, 2.4 mol acetate was produced and 0.8 mol ethylene was reduced. Acetylene, propylene and butene were less effective as H2 acceptors than ethylene, and addition of bromoethanesulfonic acid was necessary to inhibit methanogenesis in the presence of the two longer-chain olefins. Other hydrogenation catalysts were less effective in the order Pd-charcoal < PE-asbestos < Pd-PEI beads < Pt-Al2O3, Pd-CaCO3. Optimal ethylene hydrogenation was achieved with still incubation in presence of 7.2 mg Pd-BaSO4 and 0.7 g sand per ml medium. The higher catabolic rate of S. wolfei in presence of the methanogen indicated that the biological H2 removal mechanism was more efficient than the catalytic olefin reduction.Abbreviations BES bromoethane sulfonic acid - VFA volatile fatty acid  相似文献

Histochemical studies on localisation of lipids, lipase and phosphomonoeste-rases in the crown skin of male baya during non-breeding and breeding seasons were carried out. The results indicated a high turnover rate for lipid synthesis and its utilisation, increased acid and alkaline phosphatase activities in the crown skin of breeding male baya as compared to that in non-breeding season. It is surmised that crown skin of male baya becomes metabolically hyperactive during breeding season aiding processes such as cell growth and proliferation, keratinization and production of coloured feathers related to breeding.  相似文献

  • 1 New Zealand was colonized by the German wasp, Vespula germanica (F.), in the 1940s and it subsequently became established throughout the country. The common wasp, V.vulgaris (L), colonized in the late 1970s and is still spreading.
  • 2 The common wasp has replaced the German wasp in some habitats in New Zealand. Samples from a nationwide postal survey indicate that the common wasp is now the more abundant species in honeydew beech forests (Nothofagus spp.), and to a lesser extent in other native forests. The German wasp is still the more abundant wasp in rural areas (excluding forest). The two species are at present co-dominant in urban areas, although this may be a transient phase.
  • 3 In honeydew beech forest the two species show different foraging patterns that provide the potential for local coexistence. Although both species are generalist feeders, the German wasp is more commonly found foraging for protein amongst the forest litter, whereas the common wasp forages more on shrubs and tree saplings. Despite this difference, the common wasp can still replace the German wasp in honeydew beech forest within a few years of invasion.
  • 4 In honeydew beech forests in which the German wasp is the more abundant species it dominates honeydew trunks (sugar resource), whereas the common wasp dominates honeydew trunks in areas where it is the more abundant species. The change from German to common wasp domination of honeydew trunks is more rapid than the change in dominance in other microhabitats. Aggressive interactions may be taking place on this high quality, potentially defensible sugar resource.
