The Escherichia coli secA gene, whose translation is responsive to the proficiency of protein export within the cell, is the second gene in a three-gene operon and is flanked by gene X and mutT. By using gene fusion and oligonucleotide-directed mutagenesis techniques, we have localized this translationally regulated site to a region at the end of gene X and the beginning of secA. This region has been shown to bind SecA protein in vitro. These studies open the way for a direct investigation of the mechanism of secA regulation and its coupling to the protein secretion capability of the cell.  相似文献

Relaxin     
1. Relaxin is a hormone of reproduction that appears to affect parturition, uterine accommodation, and sperm motility to varying degrees in many species. 2. All relaxins have the same two chain, disulfide-linked insulin-like structure and two arginine residues in the midregion of the B chain. 3. The active relaxin molecule is produced by excision of a connecting peptide from the prohormone. 4. The biosynthetic pathways of insulin and relaxin are alike, but the relaxin prohormone is about twice as large as the corresponding proinsulin. 5. The primary structures of relaxins from apparently closely related species differ significantly in their amino acid compositions and do not fit into the traditional scheme of molecular evolution.  相似文献

The Gambierdiscus genus is a group of benthic dinoflagellates commonly associated with ciguatera fish poisoning (CFP), which is generally found in tropical or sub-tropical regions around the world. Morphologically similar species within the genus can vary in toxicity; however, species identifications are difficult or sometimes impossible using light microscopy. DNA sequencing of ribosomal RNA genes (rDNA) is thus often used to identify and describe Gambierdiscus species and ribotypes, but the expense and time can be prohibitive for routine culture screening and/or large-scale monitoring programs. This study describes a restriction fragment length polymorphism (RFLP) typing method based on analysis of the large subunit rDNA that can successfully identify at least nine of the described Gambierdiscus species and two Fukuyoa species. The software programs DNAMAN 6.0 and Restriction Enzyme Picker were used to identify a set of restriction enzymes (SpeI, HpyCH4IV, and TaqαI) capable of distinguishing most of the known Gambierdiscus species for which DNA sequences were available. This assay was tested using in silico analysis and cultured isolates, and species identifications of isolates assigned by RFLP typing were confirmed by DNA sequencing. To verify the assay and assess intra-specific heterogeneity in RFLP patterns, identifications of 63 Gambierdiscus isolates comprising ten Gambierdiscus species, one ribotype, and two Fukuyoa species were confirmed using RFLP typing, and this method was subsequently employed in the routine identification of isolates collected from the Caribbean Sea. The RFLP assay presented here reduces the time and cost associated with morphological identification via scanning electron microscopy and/or DNA sequencing, and provides a phylogenetically sensitive method for routine Gambierdiscus species assignment.  相似文献

Histone deacetylases (HDACs) constitute a super-family of enzymes grouped into four major classes (Class I–IV) that deacetylate histone tails leading to chromatin condensation and gene repression. Whether stroke-induced oligodendrogenesis is related to the expression of individual HDACs in the oligodendrocyte lineage has not been investigated. We found that 2 days after stroke, oligodendrocyte progenitor cells (OPCs) and mature oligodendrocytes (OLGs) were substantially reduced in the peri-infarct corpus callosum, whereas at 7 days after stroke, a robust increase in OPCs and OLGs was observed. Ischemic brains isolated from rats sacrificed 7 days after stroke were used to test levels of individual members of Class I (1 and 2) and Class II (4 and 5) HDACs in white matter oligodendrocytes during stroke-induced oligodendrogenesis. Double immunohistochemistry analysis revealed that stroke substantially increased the number of NG2+ OPCs with nuclear HDAC1 and HDAC2 immunoreactivity and cytoplasmic HDAC4 which were associated with augmentation of proliferating OPCs, as determined by BrdU and Ki67 double reactive cells after stroke. A decrease in HDAC1 and an increase in HDAC2 immunoreactivity were detected in mature adenomatous polyposis coli (APC) positive OLGs, which paralleled an increase in newly generated BrdU positive OLGs in the peri-infarct corpus callosum. Concurrently, stroke substantially decreased the acetylation levels of histones H3 and H4 in both OPCs and OLGs. Taken together, these findings demonstrate that stroke induces distinct profiles of Class I and Class II HDACs in white matter OPCs and OLGs, suggesting that the individual members of Class I and II HDACs play divergent roles in the regulation of OPC proliferation and differentiation during brain repair after stroke.  相似文献

The common flesh color of commercially grown watermelon is red due to the accumulation of lycopene. However, natural variation in carotenoid composition that exists among heirloom and exotic accessions results in a wide spectrum of flesh colors. We previously identified a unique orange flesh watermelon accession (NY0016) that accumulates mainly β-carotene and no lycopene. We hypothesized this unique accession could serve as a viable source for increasing provitamin A content in watermelon. Here we characterize the mode of inheritance and genetic architecture of this trait. Analysis of testcrosses of NY0016 with yellow and red fruited lines indicated a codominant mode of action as F1 fruits exhibited a combination of carotenoid profiles from both parents. We combined visual color phenotyping with genotyping-by-sequencing of an F2:3 population from a cross of NY0016 by a yellow fruited line, to map a major locus on chromosome 1, associated with β-carotene accumulation in watermelon fruit. The QTL interval is approximately 20 cM on the genetic map and 2.4 Mb on the watermelon genome. Trait-linked marker was developed and used for validation of the QTL effect in segregating populations across different genetic backgrounds. This study is a step toward identification of a major gene involved in carotenoid biosynthesis and accumulation in watermelon. The codominant inheritance of β-carotene provides opportunities to develop, through marker-assisted breeding, β-carotene-enriched red watermelon hybrids.  相似文献

Functional and anatomical small animal imaging represents one of the most promising tool in preclinical research capable of in vivo quantitative analysis of induced abnormalities in animal models and evaluation of metabolic, functional, and morphological consequences of genomic alterations in order to establish new therapeutic and diagnostic drugs. Among all the methods, SPECT imaging has recently benefited from numerous technological developments leading to submillimeter spatial resolutions with increasing detection sensitivity. Moreover, availability and sensitivity of single photon radiotracers are serious advantages of the method, which should play an important role in pre clinical imaging. The aim of this article is to present and discuss these small animal SPECT imaging advances.  相似文献

Summary A child with cri-du-chat syndrome, 46,XY,5p-, was born to a mother who had two normal children and two abortuses. The mother was shown to carry a balanced translocation. Giemsa and fluorescent banding demonstrate the exact location of the translocated segment. The deleted short arm of chromosome number 5 was shown to be attached to the long arm of chromosome number 11.

Bei der Mutter wurde eine balancierte Translokation nachgewiesen; Chiemsa- und Fluorescens-Bandenmuster zeigten die genaue Lokalisation des translozierten Segmentes: Der deletierte kurze Arm des Chromosoms Nr.5 war an den langen Arm vom Chromosom Nr. 11 angeheftet.

The temporal and spatial abundance of postlarval Penaeus setiferuswas studied from plankton samples in oceanic and estuarine watersnear the North Edisto Inlet, South Carolina (USA), during threecruises in 1993 and 1994 (May 1993, August/September 1993 andJune 1994). Each covered a full cycle of neap-spring tides.On each cruise, bongo nets were towed at three stations acrossthe inlet throat on nightly flood tides. During the first twocruises, tows were made at surface and near-bottom depths inthe inlet, while sampling was confined to surface depths duringthe last cruise. Plankton tows were also made outside the inletalong transects extending in a cross-shelf direction and alongan arcuate transect around the inlet mouth. Stations along thearcuate transect were intensively sampled over a full tidalcycle during the last cruise. Extensive oceanographic and meteorologicalobservations were obtained from moored instrument arrays andshipboard sampling in order to relate fluctuations in tidal,lunar and wind phenomena to temporal changes in postlarval density.Densities of postlarvae were greater in the inlet throat thanat stations offshore. A significant interaction of postlarvaldensity among inlet stations and depth was noted in May. Forother cruises, no significant differences in density were notedamong stations across the inlet, but postlarvae were concentratedat the surface. The lack of a consistent horizontal salinitygradient and obvious pattern in water masses across the inletmay explain why postlarval densities did not consistently differlaterally in the inlet. Greater densities generally occurredduring the first quarter moon, although a clear relationshipof larval density to the spring-neap cycle was not observed.Highest mean densities of ingressing postlarvae in surface floodtide collections from the inlet were generally associated withdownwelling onshore winds which generate onshore flow near thesurface. The similarity between the time series curves of postlarvaldensity and the tidal component of currents just offshore ofthe inlet suggests that tidal transport may facilitate movementinto the estuary. Based on increased postlarval density at thesurface during early strong flood tides and a reduced densityat depth in the inlet, we hypothesize that postlarval P setiferusare utilizing selective tidal stream transport.  相似文献
