MiR-30b is involved in methylglyoxal-induced epithelial-mesenchymal transition of peritoneal mesothelial cells in rats

Epithelial-mesenchymal transition (EMT) of peritoneal mesothelial cells (PMC) is a major contributor to the pathogenesis of peritoneal fibrosis. EMT is at least in part caused by repeated exposure to glucose degradation products (GDPs), such as methylglyoxal (MGO). MiRNA contributes greatly to the EMT of PMCs. In this study, we tried to profile whether differences exist between the peritoneal membrane (PM) miRNA expression seen in control rats and that seen in rats injected intraperitoneally with MGO. We assessed whether miR-30b has a possible role in MGO-induced EMT of PMCs in rats. Comparative miRNA expression array and real-time PCR analyses were conducted for the control group at the start of the experiment and for the MGO group after 1 and 2 weeks. During the second week, the MGO rats were treated with: a chemically modified antisense RNA oligonucleotide (ASO) complementary to the mature miR-30b (ASO group); an miR-30b mismatch control sequence (MIS group); or a citrate buffer (EMT group). Bioinformatic analyses indicated that the 3′ untranslated region (3′-UTR) of bone morphogenetic protein 7 (BMP7) mRNA did contain a putative binding site for miR-30b. We also tried to investigate whether miR-30b targeted BMP7 in vitro by transfection. Of the upregulated miRNAs, miR-30b expression demonstrated the greatest increase. The administration of miR-30b ASO for two weeks significantly reduced α-SMA excretion and upregulated E-cadherin and BMP-7 expression. Our in vitro study showed that miR-30b directly targeted and inhibited BMP7 by binding to its 3’-UTR. Our results revealed that miR-30b is involved in MGO-induced EMT of PMCs in rats.     

Uterine Artery Suture: A Preventive Approach for Pernicious Placenta Previa

Pernicious placenta previa is an obstetric complication that results in a crisis situation for many patients. Most existing options deal with the problems after delivery leading to unfavorable outcomes. To circumvent this situation, a few preventive simple surgical options have been discussed in this paper. The highlight of the approach is the application of ascending uterine artery suture that drastically reduced the amount of bleeding as compared to that observed in regular surgeries. Combined with the placenta accrete location, wedge resection and ‘patch’ homeostasis of the serous membrane, the surgical modifications adopted effectively reduced the need for hysterectomy thus preserving the fertility of the patients for the future. Since fetuses were safely removed within 1–2 min, the Apgar scores of the neonates were also largely unaffected.     

森林可燃物计划烧除的相关分析

森林火灾带来了严重后果，但许多人对黑色防火感到一些困惑，计划烧除是“防火”、“用火”还是“放火”．随着数据分析技术的进步，运用数据库、多元统计分析等方法可以分析森林火灾调查和统计的数据，探索森林防火的最佳措施．针对黑色防火的深入理解，对试验数据进行了相关分析．该相关分析是利用了云南松林里进行计划烧除后得到的各种数据，然后通过SPSS软件来分析数据．试验结果可以表明：计划烧除时火强度、火蔓延速度与火焰高度属于正相关；火强度与森林可燃物的生物量也属于正相关，通过小强度的计划烧除能有效的减少森林可燃物的生物量，从而提高森林的自防能力．     

Expression, purification, and in vitro refolding of soluble tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a new member of the TNF superfamily. Here, a recombinant form of the extracellular domain of the TRAIL (sTRAIL) was expressed in Escherichia coli BL21(DE3) under the control of a T7 promoter. The resulting insoluble bodies were separated from cellular debris by centrifugation and solubilized with 8 M urea. A rapid and simple on-column refolding procedure was developed. It was applied and then the refolded sTRAIL was purified by anion-exchange chromatography. The purified final product was >98% pure by SDS-PAGE stained with Coomassie brilliant blue R-250. Mass spectroscopic analysis indicated the protein to be 19.2 kDa, which equalled the theoretically expected mass. N-terminal sequencing of refolding sTRAIL showed the sequence which corresponded to the designed protein. The renatured protein displayed its immunoreactivity with the antibodies to TRAIL protein by Western blotting. The purified sTRAIL had a strong cytotoxic activity against human cervical cancer HeLa cells with ED50 about 1.5 mg/L. Circular dichroism and fluorescence spectrum analysis showed that the refolded sTRAIL had a structure similar to that of native protein with beta-sheet secondary structure. This efficient procedure of sTRAIL renaturation may be useful for the mass production of this therapeutically important protein.     

浙江丽水生态示范区植物的分布特点

1999年底被国务院批准设立为全国第4个生态示范区的丽水市（包括莲都区，缙云县，云和县，松阳县，遂昌县，庆元县，青田县，龙泉市和景宁畲族自治县），由于地形复杂，原生，次生林等保存较好，生态环境保持良好，植物资源极为丰富。笔者根据多年来对该地区的资源调查和标本采集统计，已知有维管束植物206科，94属，2881种，其中蕨类植物的种类和数量在中国东半部仅次于台湾和海南，尤其是鳞毛蕨属和复叶耳蕨属可以认为是在我国的一个分布中心，种子植物的种类和数量与邻近周边地相比也占有明显优势，区内珍衡濒危植物众多，区系起源古老，不乏古老的种，属及残遗种，且区系成分复杂，来源于多种地理成分。     

黄心夜合不同季节鲜叶挥发性成分研究

利用气相色谱-质谱联用（GC-MS）技术，对黄心夜合（Michelia martinii Levl.）春、夏、秋、冬4季不同鲜叶挥发油成分及其含量进行了分析研究。结果显示，去掉峰值0.49以下的色谱峰从4个季节黄心夜合鲜叶样品中共鉴定出65种挥发油成分，其中，春、夏、秋、冬4个季节鲜叶分别检测出26种、29种、31种和32种成分，4个季节鲜叶样品共有成分为三环烯、α-蒎烯、莰烯、香桧烯、β-蒎烯等14种。黄心夜合4个季节鲜叶及干燥花蕾中的共有挥发性成分有α-蒎烯、莰烯、β-蒎烯、芳樟醇4种。黄心夜合鲜叶挥发油成分中的α-蒎烯、β-蒎烯、β-月桂烯、β-芳樟醇、芳樟醇、月桂烯含量高，且春季鲜叶挥发油得率最高，最适合挥发油提取。鲜叶具有保健功能，适于四季园林养生配置。本研究黄心夜合4个季节鲜叶挥发油成分差异及季节性变化，可为黄心夜合挥发油提取的季节选择及养生保健群落的营建提供科学依据和指导。     

Effects of fertilization on the relations of diameter at breast height between different-aged ramets of Phyllostachys edulis population

克隆植物不同年龄分株间可以通过生理整合作用相互传递资源, 实现资源共享。施肥可以改变林地资源状况, 进而可能影响分株间的整合作用及不同年龄分株间的生长关系。该文采用标准主轴(SMA)估计法和多元回归分析, 对不同施肥管理下大型克隆植物毛竹(Phyllostachys edulis)种群不同年龄分株(1年生分株、3年生分株和5年生分株)间的胸径(DBH)关系进行了研究, 探讨毛竹不同年龄分株DBH大小关系对施肥的响应。结果表明: 连续5年施肥有效地增加了毛竹1年生分株的DBH, 而施肥1年处理下, 1年生分株DBH与3年生分株和5年生分株平均DBH差异不显著(p > 0.05)。1年生分株与3年生分株和5年生分株DBH之间的SMA斜率为0.88-1.10, 均与1.00不存在显著差异(p > 0.05), 表现为等速生长关系。施肥1年和连续施肥5年没有改变不同年龄分株DBH之间的SMA斜率, 但连续5年施肥导致SMA的y轴截距显著增大(p < 0.001), 即在3年生分株和5年生分株DBH相同的情况下, 连续5年施肥使1年生分株具有更大的DBH增长量。采用多元线性逐步回归分析发现, 毛竹1年生分株的DBH大小在很大程度上受3年生分株DBH大小的影响, 而连续5年施肥可增强5年生分株对1年生分株DBH大小的影响。
