Previous studies upon zinc-iron interactions gave conflicting results that could come from differences in protocol design or in trace element status of subjects. The present work assessed the influence of zinc : iron ratio and iron deficiency upon zinc absorption. The digestive absorption of zinc sulphate (100 mol Zn/l) in presence of iron gluconate was studied in perfused jejunal loops (n = 6/group) of normal rats (range 0–1000 mol Fe/l) and iron deficient rats (200–750 mol Fe/l). In normal rats no significant iron inhibition on zinc absorption occurred at Fe:Zn ratio below 2:1. At higher ratios zinc uptake and net absorption decreased significantly (p<0.05). Between 2:1 and 5:1 a dose dependent inhibition of zinc absorption occurred and reached a plateau beyond this ratio. In iron deficient animals no changes in zinc uptake, mucosal retention and absorption compared to normal animals occurred at ratio 2:1. At higher ratios differences were observed at every zinc absorption step except for mucosal retention at 7.5:1 ratio.

Iron-zinc interactions depend on their ratio and on previous trace elements status of subjects. Due to the wide and unknown variations that were likely to occur between the subjects of previous human and experimental studies, these results could explain some of the discrepancies between their results.  相似文献

Tracer kinetic techniques based on zinc stable isotopes have a vital role in advancing knowledge of human zinc physiology and homeostasis. These techniques have demonstrated the complexity of zinc metabolism, and have been critical to estimating the size and interrelationships of those pools of zinc that exchange rapidly with zinc in plasma and which are likely to be especially important for zinc dependent biology. This paper presents findings from recent research linking a steady state compartmental model with non-steady state post-prandial sampling from the intestine, utilizing a combination of intestinal intubation/perfusion and stable isotope tracer kinetic techniques. The gastrointestinal tract has a central role in maintaining whole body zinc homeostasis. While the fractional absorption of zinc from a meal depends on the quantity of exogenous zinc and on such dietary factors as phytic acid, the fractional absorption does not appear to be dependent on the size of the rapidly exchanging pool of the host. In contrast, the quantity of endogenous zinc excreted via the intestine is positively correlated with both the amount of absorbed zinc and the zinc `status' of the host, and thus this process has an equally critical role in maintaining zinc homeostasis. The observed alterations in zinc metabolism in some disease states can be understood in the context of known homeostatic processes. In other conditions, however, such alterations as inflammation-associated hyperzincuria and zinc redistribution, the links between homeostatic perturbation and cellular biology are yet to be explained. Thus the challenge remains for research at the whole body level to carefully characterize zinc distribution and exchange under diverse circumstances, while research at the cellular level must elucidate the regulatory processes and the factors to which they respond.  相似文献

用蛭石与石英砂作为混合培养介质研究了低磷胁迫对水稻(Oryza sativa L.)苗期侧根发生发育的影响及其与磷吸收的相关关系。结果表明:低磷对水稻的侧根发生发育具有明显的诱导作用及基因型差异。相关性分析表明:单位侧根长度的增加与单位根表面积的增大极显相关,而单位侧根数量的增多与单位根表面积的增大无显的相关性。表明单位根表面积的增加主要来自于单位侧根的伸长。侧根参数与磷含量的相关性分析表明:低磷条件下,侧根总长度和侧根数量都与植株磷含量存在显的正相关,根系总表面积与磷含量存在极显的正相关。表明在低磷条件下,侧根的发生发育对水稻的磷吸收具有重要的作用。根系和地上部的可溶性糖含量分析表明;低磷胁迫改变了同化物在地上部和根系的分配。生物量测定表明:低磷胁迫显增大了植株的根冠比。  相似文献

An assay for cadmium in whole blood and urine using deuterium background-correction electrothermal atomic absorption spectroscopy (D(2)-ETAAS) was developed. Cadmium (in a 1- to 2-ml sample) was bound to 15 mg anion-exchange resin, interfering ions were removed in a 2-ml Bio-Spin column, and cadmium was extracted into 100 microl 1M nitric acid for analysis. Cadmium in the sample extract was concentrated 7-fold for blood and 10-fold for urine over the starting material. These steps produced cadmium atomic absorption traces with high signal to background ratios and allowed analysis against aqueous standards. At approximately 0.1 ng Cd/ml, mean intra- and interassay coefficients of variation were 11-12%. Cadmium recovery for 0.1 to 0.6 ng added cadmium was 107+/-4% for blood and 94+/-4% for urine (mean+/-SE, n=3). The mean detection limit (mean + 3 x SD of blank) was 0.008 ng/ml for blood and 0.003 ng/ml for urine. Samples from "unexposed" animals including humans ranged from 0.051+/-0.000 to 0.229+/-0.035 ng/ml. Values were approximately 10-fold lower than those obtained by the method of Stoeppler and Brandt using Zeeman background-correction ETAAS. This new high-sensitivity, low-volume assay will be useful for epidemiological studies, even those involving children, and will provide a means to help determine the contribution of cadmium to disease incidence in the general population.  相似文献

This article reports on the levels of cadmium, lead, and mercury in 21 benign brain tumors and 23 malignant brain tumors. All measurements were performed by atomic absorption spectrometry following digestion by nitric acid. Average concentrations of cadmium, lead, and mercury in brain tumors were 2.02 (ND-72.78), 0.625 (ND-83.13), and 0.118 (ND-1.96) μg/g wet weight, respectively. This article describes a study in which four trace elements (Se, Mn, Cu, and Fe) were analyzed in the blood serum of the patients with colorectal cancer from the Moravian region of the Czech Republic. Atomic absorption spectrometry with graphite furnace atomization was used for analysis of selenium and manganese and with flame atomization for analysis of copper and iron. The observed serum concentrations in adenocarcinoma colorectal patients of selenium were significantly lower (41.8 ± 11.6 μg/L) and those of manganese (16.3 ± 4.5 μg/L) and iron (2.89 ± 1.23 mg/L) were significantly higher as compared to the age-matched control group. Copper serum content (0.95 ± 0.28 mg/L) did not significantly differ as compared to healthy population.  相似文献

To further examine the interrelationships between manganese and iron absorption, the mucosal uptake, initial rate of loss, wholebody retention, and tissue distribution of an orally administered 54Mn radiotracer were compared between normal and β2-microglobulin knockout [β2m(-/-)] mice. These mutant mice are commonly used as a model for the study of human hemochromatosis, a hereditary ironoverload disease. Initial uptake of 54Mn by the intestinal mucosa, the liver, and the brain was not different between the two strains. The mutant mice had much higher concentrations of nonheme and total iron in the liver, but hepatic manganese, copper, magnesium, and zinc concentrations were similar between the two strains. In summary, the mucosal uptake and whole-body retention of manganese and tissue manganese concentrations were not altered in β2m(-/-) mice; this suggests that normal homeostasis of manganese is not affected by the altered HFE protein-β2m complex in these mice. The light-induced difference absorption spectra associated to the photo-accumulation of reduced pheophytin a were studied in the isolated D1–D2–Cyt b559 complex in the presence of variable methyl viologen concentrations and different illumination conditions under anaerobiosis. Depending on the methyl viologen/reaction centre ratio, the relative intensities of the spectral bands at 681.5±0.5, 667.0±0.5 and 542.5±0.5 nm were modified. The reduced pheophytin a located at the D1-branch of the complex absorbs at 681.7±0.5 nm, and at least two additional pigment species contribute to the Qy band of the difference absorption spectra with maxima at 667.0±0.5 and 680.5±0.5 nm. We propose the additional species correspond to a peripheral chlorophyll a and the pheophytin a located at the D2-branch of the complex, respectively. The blue absorbing chlorophyll at 667 nm is susceptible to chemical redox changes with a midpoint reduction potential of +470 mV. The Qx absorption bands of both pheophytins localised at the D2- and D1-branch of the D1–D2–Cyt b559 complex were at 540.7±0.5 and 542.9±0.5, respectively. The results indicated that the two pheophytin molecules can be photoreduced in the D1–D2–Cyt b559 complex in certain experimental conditions. Treatment of last-instar larvae of multi-resistant cotton leafworm Spodoptera littoralis with four dibenzoylhydrazines, methoxyfenozide (RH-2485), tebufenozide (RH-5992), halofenozide (RH-0345), and RH-5849, resulted in premature molting leading to death. Methoxyfenozide was the most toxic followed by tebufenozide, halofenozide, and RH-5849. To explain differences in toxicity, especially between multi-resistant and laboratory strains, absorption in the body tissues and oxidative metabolism were tested with 14C-labeled ecdysone agonist and a Lineweaver-Burk assay, respectively. Then to address different compound potencies in multi-resistant strains, the potency of the four ecdysone agonists was measured based on their ability to mimic the natural insect molting hormone, 20-hydroxyecdysone (20E) by inducing evagination in isolated imaginal wing discs. Using monoclonal antibody 9B9, the presence of ecdysteroid receptors in imaginal discs in vitro was confirmed. In parallel, Scatchard plot analysis with whole imaginal wing discs cultured with different concentrations of 3H-labeled ponasterone A indicated no significant difference in affinity and in number of target sites for binding between multi-resistant and susceptible laboratory strains. The four compounds tested caused the effect as agonists of 20E in vitro, and typically the order of their toxicities (LC50s) corresponded with that for evagination-induction with whole imaginal discs.  相似文献

Pre-epithelial ``unstirred' layers (abbr. pL) are generally regarded as undesirable diffusion barriers that impede access to absorptive cells of exogenous hexoses, amino acids or other experimental probes added to fluids bathing the mucosal surface. In the present paper it is suggested that the pL may have a functional role. Diffusion plus convection of saccharides and oligopeptides from lumen to brush border, combined with absorption to blood of their hydrolytic products, confers rectifying properties to the pL. The proposed model, based on experimental data from segmental jejunal perfusions in normal human subjects, indicates that the functional pathway for diffusion plus convection through the pL of hexoses and amino acids bound in the form of oligomers is only 10 ± 2 μm or little more than the anatomical thickness of the glycocalyx and mucus layers. In contrast, the pathlength from brush border to lumenal perfusion fluid for diffusion minus convection of monomers generated by membrane bound hydrolases is 50–150 μm. According to this model the pL offers little resistance to the passage of saccharides or oligopeptides from lumen to brush border but at the same time it provides a protective blanket that diminishes diffusional losses to lumenal chyme of hexoses and amino acids generated in the brush border. The model provides a theoretical explanation for the ``kinetic advantage' of transporting hexoses or amino acids through the pL in the form of oligomers and it predicts the proximal-distal concentrations of free glucose or fructose found experimentally in the outflows from jejunal segments perfused with sucrose or maltose.
