Somatic embryogenesis in white spruce (Picea glauca): genetic control in somatic embryos exposed to storage,maturation treatments,germination, and cryopreservation

Genetic controls for growth of embryogenic cultures, storage, maturation treatments, germination and cryopreservation in white spruce somatic embryogenesis (SE) were examined. These SE processes were under genetic control but less strongly so than the initiation phase. For all the SE characters examined, variance due to clones within families was significant and often the largest genetic component of variance. This was further partitioned using an additive-dominance-epistasis model. A relatively-large proportion of the total genetic variance was due to epistatic variance in the maturation and germination of somatic embryos. Embryogenic lines were cryopreserved easily without a distinct genetic influence being noticed.     

Isolation and function of a low molecular weight protein of mung bean embryonic axes

A low molecular weight protein from dry mung bean (Vigna radiata) embryonic axes has been purified to near homogeneity by chromatography on DEAE-cellulose and hydroxylapatite. It shows a molecular weight of about 12,000 in sodium dodecyl sulfate-polyacrylamide gels and a sedimentation coefficient of about 2 S in sucrose gradients. This protein occurs in greater amounts in dry axes than in dry cotyledons, and it dramatically disappears during early germination of the seed. Affinity chromatography tests do not indicate it as a trypsin inhibitor or as a glycoprotein. It is a water-soluble cytoplasmic protein exhibiting an amino acid composition characteristic of storage proteins with a high content of glutamic acid/glutamine. Germination and establishment of Sahelian rangeland species

Summary In the context of a study on the dynamics of Sahelian rangelands, seed characteristics of about 30 species have been described, with particular attention to germination characteristics. Data on both form and weight of the seeds are presented. Germination was studied under controlled conditions in a climate room. Optimum conditions for germination were established, and the species were characterized by induction time, slope of germination vs. time curve and final germination percentage. The results obtained under climate room conditions appear to be correlated satisfactorily with the behaviour of the different species in the field.     

The effects of thermal scarification and seed storage on germination of four heathland species

The germination responses of 4 shrubs, Erica arborea, E. australis and Daboecia cantabrica (Ericaceae) and Genistella tridentata (Papilloneaceae) in relation to variable light and temperature regimes was determined for freshly harvested and 1 year-old seeds. Temperature treatments consisted of 5 levels of heat from 60 °C to 140 °C and two levels of exposure time, 2 and 4 min. We also examined the effects of stratification on germination of Erica arborea and E. australis. The seeds of Erica arborea were the first to begin germinating and the freshly collected ones reached the highest level of germination with light. The seed viability of E. arborea was affected by storage, decreasing germinative capacity, while mean germination in Erica australis ssp. aragonensis was lowest. However, the stored E. australis seeds germinated better than the fresh ones. Germination response to ageing varies in Daboecia cantabrica depending on heat pretreatment. A significantly positive effect of high temperature and exposure time on germination was shown in Daboecia cantabrica and Genistella tridentata. Cold pre-treatment of seeds did not enhance germination.
