Deleterious mutation accumulation plays a central role in evolutionary genetics, conservation biology, human health, and evolutionary medicine (e.g., methods of viral attenuation for live vaccines). It is therefore important to understand whether and how quickly populations with accumulated deleterious mutational loads can recover fitness through adaptive evolution. We used laboratory experimental evolution with four long-term mutation-accumulation (MA) lines of Caenorhabditis elegans nematodes to study the dynamics of such fitness evolution. We previously showed that when homozygous mutant populations are evolved in large population sizes, they can rapidly achieve wild-type fitness through the accumulation of new beneficial or compensatory epistatic mutations. Here, we expand this approach to demonstrate that when replicate lineages are initiated from the same mutant genotype, phenotypic evolution is only sometimes repeatable. MA genotypes that recovered ancestral fitness in the previous experiment did not always do so here. Further, the pattern of adaptive evolution in independently evolved replicates was contingent upon the MA genotype and varied among fitness-related traits. Our findings suggest that new beneficial mutations can drive rapid fitness evolution, but that the adaptive process is rendered somewhat unpredictable by its susceptibility to chance events and sensitivity to the evolutionary history of the starting population.  相似文献

毛乌素沙地沙漠化逆转过程土壤颗粒固碳效应   总被引:3,自引:0,他引:3  
为揭示毛乌素沙地沙漠化逆转过程中土壤颗粒的固碳效应,选择陕北榆林治沙区从流沙地、半固定沙地到林龄为20~55年生的灌木和20~50年生的乔木固沙林地,采用物理分组法分析了土壤砂粒、粉粒、黏粒结合碳的演变特征和累积速率.结果表明: 对比流沙地,土壤总有机碳及各颗粒碳含量在两种固沙林地均呈显著增加趋势,并以表层0~5 cm土壤碳含量增幅最高.从流沙地到55年生灌木和50年生乔木固沙林地,0~5 cm土层砂粒碳密度增速均为0.05 Mg·hm-2·a-1,粉粒碳密度增速分别为0.05和0.08 Mg·hm-2·a-1,而黏粒碳密度增速分别为0.02和0.03 Mg·hm-2·a-1.0~20 cm土层,两种林地各颗粒碳密度增速平均为0~5 cm土层的2.1倍.按此增速到50~55年生的固沙林地时,两种林地0~20 cm土层的砂粒碳、粉粒碳和黏粒碳密度分别比流沙地平均提高6.7、18.1、4.4倍,并且颗粒碳对总有机碳的累积贡献率平均为粉粒碳(39.7%)≈砂粒碳(34.6%)>黏粒碳(25.6%).综上,毛乌素沙地沙漠化逆转过程土壤颗粒均表现出显著的固碳效应,且以砂粒和粉粒为主要固碳组分.  相似文献

科尔沁沙地沙漠化土地恢复面临的挑战   总被引:8,自引:0,他引:8  
对科尔沁沙地近50年来沙漠化土地面积、水资源可利用性、土地利用与草地植物生产力的变化进行了分析.结果发现:科尔沁沙地的土地沙漠化在经历了20世纪50—80年代中期的快速发展之后,80年代后期发生了逆转.在过去的50年中,西辽河径流量持续减小,1999年在西辽河中游的内蒙古自治区通辽市段断流;近20年来,位于科尔沁沙地腹地的西湖水位下降近10 m, 2001年干涸.科尔沁沙地草场的地上生物量由1937年的520 g·m-2下降到2005年的197 g·m-2.产生这种结果的主要原因是土地利用方式的变化,即基于灌溉的沙漠化治理规模的扩大和灌溉农田面积的增加,导致用水量增加.这也是科尔沁沙地实现土地沙漠化持续逆转面临的严峻挑战.  相似文献

In this paper, we report the effects of laser microirradiation of prophase nucleoli and mitotic chromosomes in cells of female rat kangaroo kidney epithelial cell line PTK1. When the laser power delivered to sample surface was 90–190 mW, irradiation of one of the two nucleoli in the prophase cell did not inhibit the mitotic progress, but resulted in the loss of the irradiated nucleolus in daughter cells. When the laser power was increased to 360–420 mW, either irradiation of the nucleolus or chromosome in midprophase caused a blockage of mitosis at terminal midprophase. The irradiated cells returned morphologically to early prophase. No mitotic reversion occurred in the case of irradiation of chromosomes at late prophase, prometaphase, metaphase, and anaphase. Irradiation of the cytoplasm in prophase cells caused a 50–70 min mitotic delay at prophase. However, the irradiated cells underwent successive mitotic divisions. The mechanism of laser-induced mitotic prophase reversion is discussed.  相似文献

Bacteriophage lambdahyp mutants have been isolated as survivors of Escherichia coli K-12 bacteria lysogenic for lambda Nam7am53cI857. The hyp mutants are characterized by (i) their localization in the y region very close to the imm lambda/imm434 boundary, (ii) polarity on O gene expression, (iii) immediate recovery of lambda immunity at 30 degrees C after prolonged growth of lambda Nam7am53cI857 hyp lysogens at 42 degrees C even in the presence of an active cro gene product, (iv) ability of phage lambda v2v3vs326 but not lambda v1v2v3 to propagate on lambda cI+hyp lysogens, (v) inability to express lambda exonuclease activity after prophage induction, and (vi) inviability at any temperature of phage carrying the hyp mutation. All these properties are referred to collectively as the Hyp phenotype. We show that the Hyp phenotype is due to cII-independent constitutive cI-gene-product synthesis originating in the y region, which results in the synthesis of anti-cro RNA species, and constitutive levels of cro gene product present even in lambda cI+hyp lysogens. A model is presented which is consistent with all the experimental observations.  相似文献

Reversion mutagenesis of three single P elements located in the cytogenetic interval 1E-2A at the tip of the X chromosome of Drosophila melanogaster was used to recover new deletions in this chromosomal region. The deletions obtained include small aberrations within region 2A and larger lesions extending from 2A into 1E and 1B. All three screens also yielded terminal deficiencies. The new deficiencies, together with previously characterized rearrangements, were analyzed for their complementation behaviour with the maternal effect locus fs(1)Nasrat and lethal loci in the region. These analyses provide an overall genetic map of the interval 1E-2A. In addition, the smaller deletions were physically mapped within cloned genomic DNA of the 2A region.  相似文献

SaMADS D gene of Sinapis alba was isolated by screening a cDNA library from young inflorescences with a mixture of MADS-box genes of Antirrhinum majus (DEF, GLO, SQUA) as probe. Amino acid sequence comparison showed a high degree of similarity between the SaMADS D and AGL9, DEFH200, TM5, FBP2 and DEFH 72 gene products. Analysis of the SaMADS D gene expression by in situ hybridization reveals a novel expression pattern for a MADS-box gene and suggests a dual function for this gene: first, as a determinant in inflorescence meristem identity since it starts to be expressed directly beneath the inflorescence meristem at the time of initiation of the first floral meristem, is no longer expressed in the inflorescence meristem forced to revert to production of leafy appendages, and is expressed again when the reverted meristem resumes floral meristem initiation, and, second, as an interactor with genes specifying floral organ identity since it is expressed in the floral meristem from the stage of sepal protrusion.  相似文献

An assessment was made of the response of 10 blackcurrant genotypes, differing in resistance to the agent of blackcurrant reversion disease (BRD) and to its gall mite vector, in field trials in Scotland and Finland over 5 yr. At each location, the 10 genotypes were planted in plots containing infector plants with high inoculum levels of the two organisms. In Scotland, the infector plants contained large numbers of gall mites (Cecidophyopsis ribis) and were infected with the European (E) form of BRD; in Finland, infector plants contained a different species of gall mite (C. spicata) and the severe Russian form (R) of BRD. As expected, at both sites, almost all plants of cvs Ben Alder, Ben Lomond, Ben Tirran, Ojebyn and an SCRI selection F4/1/66, which are susceptible to gall mite and BRD, became infested with each of these organisms. However, in Scotland but not in Finland, 58% of cv. Ojebyn plants were affected by BRD. The cv. Foxendown, which contains gene Ce that confers apparent immunity to C. ribis, was free from galls and failed to develop distinctive BRD symptoms at both sites. The cvs Rus and Neosypajuscajaija, which contain gene P which is reported to confer resistance to C. ribis, were infested more slowly by mites than the mite-susceptible genotypes and showed a smaller number of galls per plant. Also, they were infected with BRD more slowly than some mite-susceptible genotypes although by the end of the experiment, most plants were affected by BRD. All plants of cvs Golubka and Ben Gairn, which are resistant to the agent of BRD, remained free from distinct BRD symptoms at both sites despite the fact that plants contained galls. These data indicate the superiority of gene Ce over gene P for resistance to gall mites with the added benefit that the virtual immunity to gall mites provided by gene Ce provides a high level of protection against infection with BRD. The relative merits of these different forms of resistance to gall mites and BRD in blackcurrant are discussed in relation to present control methods for these two organisms and in the light of recent findings of different species of eriophyid mites on Ribes species.  相似文献
