The kinetic mechanism and evaluation of several potential inhibitors of purified arginine kinase from the cockroach (Periplanta americana) were investigated. This monomeric phosphagen kinase is important in maintaining ATP levels during the rapid energy demands of muscle required for contraction and motility. Analysis reveals the following dissociation constants (mM) for the binary complex: E.Arg P-->E+Arg P, K=1.0; E.Arg-->E+Arg, K=0.45; E.MgATP-->E+MgATP, K=0.17; E.MgADP-->E+MgADP, K=0.12; and the ternary complex: Arg P.E.MgADP-->E.MgADP+Arg P, K=0.94; Arg.E.MgATP-->E.MgATP+Arg, K=0.49; MgATP.Enz.Arg-->E.Arg+MgATP, K=0.14; MgADP.E.Arg P-->E.Arg P+MgADP, K=0.09. For a particular substrate, the ratio of the dissociation constants for the binary to ternary complex is close to one, indicating little, if any, cooperativity in substrate binding for the rapid equilibrium, random addition mechanism. The time course of the arginine kinase reaction exhibits a pronounced curvature, which, as described for enzyme from other sources, is attributed to formation of an inhibitory catalytic dead-end complex, MgADP.E.Arg. The curvature is accentuated by the addition of monovalent anions, including borate, thiocyanate, and, most notably, nitrite and nitrate. This effect is attributed to stabilization of the dead-end complex through formation of a transition state analog. However, the substantial decrease in initial velocity (92%) caused by nitrate is due to an additional inhibitory effect, further characterized as non-competitive inhibition (Ki=8.0 mM) with the substrate L-arginine. On the other hand, borate inhibition of the initial velocity is only 30% with significant subsequent curvature, suggesting that this anion functions as an inhibitor mainly by formation of a transition state analog. However, some component of the borate inhibition appears to be mediated by an apparent partial competitive inhibition with L-arginine. D-arginine is not a substrate for arginine kinase from the cockroach, but is an effective competitive inhibitor with a Ki=0.31 mM. L-Canavanine is a weak substrate for arginine kinase (Km=6.7 mM) with a Vmax for the pure enzyme that is approximately one-third that of L-arginine. However, initial velocity experiments of substrate mixtures suggest that competition between L-canavanine and L-arginine may not be a simple summation effect and may involve a structural modification. Sensitivity of arginine kinase activity to D-arginine as well as nitrate and borate anions, coupled with the fact that L-arginine is an essential amino acid for the cockroach, suggest that arginine kinase could be a useful chemotherapeutic target for the control of cockroach proliferation. 相似文献

The South Island of New Zealand offers unique opportunities to study insect evolution due to long-term physical isolation, recent alpine habitats and high levels of biotic endemism. Using DNA sequence data from cytochrome oxidase subunit 1, we investigated the phylogeographical pattern among 10 endemic cockroach species within the genus Celatoblatta Johns (Blattidae). We tested the hypothesis that an ancestral cockroach species underwent rapid speciation in response to major climatic differentiation induced by mountain building. Results suggest that speciation was a twofold process, with an interspecific radiation of Pliocene/Pleistocene age followed by intraspecific diversification during the mid Pleistocene. Average genetic distance (maximum likelihood GTR + I + Gamma) was 9.17%, with a maximum of 14.5%. Data revealed eight deep well-supported branches, each with terminal clades. Six clades were differentiated according to morphological species, while the seventh was composed of three sympatric species. We consider the latter to be a phylogenetic species, possibly as a result of hybridization within a defined geographical area. This finding seriously challenges species distinctions for these three cockroach species. Correlation between genetic distances and a Climate Similarity Index (CSI) was negative, suggesting that species found in similar habitats are also genetically closely related. A Mantel test on within-clade genetic distances vs. linear geographical distance was positive, suggesting allopatric isolation for those haplotypes. Summary The mechanism of long-distance flight in insects was investigated by comparing lipid mobilization and transport in gregarious- and solitary-phase locusts and in the American cockroach. Unlike the gregarious-phase locust, both the American cockroach and the solitary locust were unable to form low-density lipophorin (loaded with increased amount of diacylglycerol) even when injected with adipokinetic hormone (AKH). The cockroach fat body responded to AKH. However, not only does the American cockroach lack apolipophorin-III (apoLp-III) in the haemolymph, but the fat body contains only an extremely small amount of diacylglycerol and a relatively large triacylglycerol pool. By contrast, the solitary-phase locust had apoLp-III in the haemolymph, but the fat body was only one-seventh or less in weight of the fat body of the gregarious locust. Furthermore, the fat body of the solitary locust contains a very small amount of triacylglycerol (1/20 or less of that of the gregarious locust) with only a trace of diacylglycerol. Glucose aversion(Glu) is a naturally occurring behavioral mutant of the German cockroach,Blattella germanica. Earlier work suggested thatGlu is a semidominant autosomal trait. Further study was undertaken to place the mutant gene in the linkage map of the species and expand the information on its genetic basis. Linkage was investigated in test crosses with morphologic markers for 7 of the 11 autosomal linkage groups. Assays for amounts of glucose ingested per individual confirmed the expected low consumption ofGlu heterozygotes(Glu/+) and that continuous selection pressure on the parental strain had resulted in near- and probably complete homozygosity forGlu. Glu genotypes were identified by means of a discriminating ingestion that separated heterozygotes from wild type(Glu+). Variations in expression indicate that genetic factors influence food consumption inB. germanica. Positive results were obtained in linkage tests ofGlu with ruby eye(ru), indicating that theGlu locus lies in linkage group VIII (chromosome 9), a group notable for mutants that apparently result in the expression of latent, highly primitive development pathways. 相似文献

The relationship between the particle size of an inert silica dust, its up-take from different surfaces and the grooming behaviour
of males, gravid females, and fifth and sixth instar nymphs of the German cockroachBlattella germanica (Dictyoptera: Blattellidae) (L.) was investigated. The normal grooming behaviour of gravid females, nymphs and males differed
according to sex and age. The gravid females and nymphs exhibited greater grooming activity than the males, especially of
the antennae and the legs. Gravid females, nymphs, and adult males exhibited increased grooming activity after exposure to
dust in the size range 0.5–63 μm, but there was no significant difference in grooming behaviour from the control when cockroaches
were exposed to dust sizes greater than 70 μm. Antennal grooming by males was greater than leg grooming when exposed to all
dust sizes, except size particles ranging 4.5–7.5 μm. A dust pick-up experiment indicated that the average amount of dust
transferred toB. germanica is affected by particle size, the porosity of the treated surface, and the sex and age of the cockroaches. Gravid females
picked up greater amounts of dust than fifth and sixth instar nymphs, which in turn picked up more dust than males. Silica
dust particles (0.5–7.5 μm) were picked up more effectively than larger particle sizes, by all three categories, males, females
and fifth and sixth instar nymphs of cockroach on all three test surfaces plastic, ceramic and unpainted plywood. Plywood

Nymphal development and adult female reproduction were examined in the German cockroach, Blattella germanica, using a defined artificial diet in which the type of protein was varied. Milk proteins, including casein, supported development poorly compared to meat and plant proteins. Soybean protein supported development better than all other highly purified proteins including vitamin-free casein which is commonly used in artificial diets. Last instar females fed the soybean-based diet eclosed earlier at higher eclosion weights, developed their oocytes at a faster rate and experienced higher fecundity than females fed a vitamin-free casein-based diet. Last instar females exhibited different dose-response patterns on diets containing soybean isolate or vitamin-free casein. However, at all concentrations soybean protein was superior to casein in supporting development. The results of a food utilization study during the last instar revealed that consumption rates varied between females fed the soybean and casein based diets. However, approximate digestibility, efficiency of conversion of digested food and the efficiency of conversion of ingested food did not vary significantly between the two dietary treatments. Differential development of females fed the two diets was attributed to differences in stage-specific consumption rates and the poorer quality of casein as a source of protein for development in this species. 相似文献

Summary Currently, short-term culture of insect corpora allata is most often performed in TC199. We now show that L-15B, a medium
widely used in arthropod tissue culture, is superior to TC199 for both short- and long-term culture of cockroach corpora allata.
In 3-h and 48-h incubations, juvenile hormone biosynthesis by corpora allata from Diploptera punctata was significantly higher in L-15B than in TC199. In addition, in both media, corpora allata activity was significantly improved
by flotation of glands at the medium surface. Characteristics of L-15B responsible for its superiority were examined by comparison
of gland activities in several TC199 formulations that had been modified in different ways to be more similar to L-15B. Adjusting
the osmotic pressure of TC199 (288 mOsm/l) to near that of L-15B (362 mOsm/l) and D. punctata hemolymph (360 mOsm/l) significantly improved gland activity during the second 12 h of a 36-h incubation. Increasing the
concentrations of amino acids, sugars, and organic acids in TC199 to the same levels as in L-15B significantly improved gland
activity during both the second and third 12-h intervals of a 36-h incubation. These results suggest that L-15B is superior
to TC199 because L-15B is isoosmotic with D. punctata hemolymph and because L-15B, like cockroach hemolymph, contains a high level of organic constituents. It is therefore more
appropriate to use L-15B than TC199 for short-term in vitro assays of juvenile hormone biosynthesis and for extended corpora allata culture. 相似文献

We have observed changes with time in the fine structure of corpora allata (CA) during a known cycle of increasing and decreasing juvenile hormone (JH) synthesis in late embryos of Diploptera punctata. A previous report showed that rates of JH release were relatively low in 28-day-old embryos, but CA activity subsequently rose linearly to a peak on about day 42, and thereafter steadily declined to a low level on day 64 just before birth (Holbrook et al., 1997). We now show that, regardless of rate of JH synthesis, CA cells are large and replete with organelles which nevertheless exhibit variable morphology in embryos of different age. Highly active CA cells on day 40 contain abundant ring-form mitochondria, whereas CA cells of low activity on days 28 and 64 contain mitochondria that are rod-shaped or globular. Mitochondrial cristae were scarce and indistinct on day 28 but numerous and well developed on day 64. Endoplasmic reticula (ER) are rare on day 28 and appear in increasing numbers when CA activity rises. On day 40, ER are abundant and often exhibit a whorl-like appearance which is not observed on day 28. After day 44, when biosynthetic activity is declining, whorls of ER gradually decrease in number and are ultimately replaced by vesicular smooth ER on day 64. Neurosecretions are found only after day 38, by which time rates of JH synthesis have increased substantially from those of day 28. Except for membranous autophagic vacuoles, which are frequently found when ER whorls disintegrate as rates of JH synthesis decline toward birth, most autophagic vesicles such as multivesicular vesicles and dense bodies occur only sporadically among CA cells at all examined ages. We conclude that synchronous autophagy of exhausted organelles, which results in atrophy of CA cells and long-term arrest of JH synthesis in adult females of D. punctata, does not occur in embryos. The slow cyclic change in rate of JH synthesis in embryonic CA is most likely due to asynchronous autophagic activity and to alterations in certain unique features of intracellular organelles. 相似文献

Insecticide resistant and susceptible field-collected strains of the German cockroach were surveyed for the occurrence of
chromosome polymorphisms. Analyses were on meiotic cells of males that hatched from oothecae with unusual numbers (>5) of
incompletely developed eggs (aberrant oothecae). Three males were reciprocal translocation heterozygotes. A spontaneous translocation
was noted in one cell of a fourth male. The males were from insecticide resistant strains. Partially developed eggs in oothecae
from outcrosses of two of the translocation herozygotes ceased development in early stages and were characterized by gross
morphologic abnormalities. Chromosome abnormalities other than translocations occurred in a low percentage of males from aberrant
oothecae in both resistant and susceptible strains. The only general differences between susceptible and resistant strains
were that, in pooled data, the frequencies of unhatched oothecae and of aberrant oothecae were higher in the resistant than

Allatostatins are a family of neuropeptides first isolated from the cockroach, Diploptera punctata, that inhibit juvenile hormone production in that species (but do not do so in earwigs), and inhibit hindgut muscle contractions in some insects, including the earwig, Euborellia annulipes. We examined whether material from earwig brains is similar to cockroach allatostatins biochemically, immunologically and physiologically. Brain extracts from adult female earwigs were separated by high performance liquid chromatography (HPLC), followed by radioimmunoassay using antibodies to cockroach allatostatin (Dip-AST). Fractions that co-eluted with cockroach allatostatins were immunoreactive, and at least two peaks of immunoreactivity were detected. Material from each peak at 10 nM Dip-AST equivalents inhibited juvenile hormone biosynthesis in vitro by corpora allata of 2-day virgin D. punctata cockroaches; 1 nM was less effective, and non-immunoreactive fractions failed to inhibit juvenile hormone biosynthesis. Both crude and Sep-Pak (Waters) purified extracts of brains of earwigs containing 1 nM Dip-AST equivalents failed to suppress hindgut contractions in vitro of 2-day earwigs and of brooding female earwigs. In contrast, 1 nM cockroach allostatin 1 (Dip-AST 7) reversibly inhibited hindgut contractions in vitro. These results suggested the presence of another brain factor, such as proctolin, that counteracts the inhibitory effects of Dip-AST. In support of this hypothesis, proctolin stimulated hindgut contractions in vitro at 1 nM; the effects of equal concentrations of allatostatin and proctolin varied with the stage of the female. Furthermore, HPLC-separated fractions that co-eluted with cockroach allatostatin and were immunoreactive with antibodies to Dip-AST suppressed hindgut contractions in vitro of 2-day female earwigs. Finally, crude brain extracts of earwigs suppressed earwig juvenile hormone biosynthesis in vitro in glands of low, but not in glands of high, activity. Thus, earwig brain extract after HPLC separation has Dip-AST-like material that inhibits cockroach corpora allata and suppresses earwig hindgut contractions. Sep-Pak-extracted earwig brain material, however, does not inhibit earwig gut contraction. Although synthetic Dip-AST 7 does not inhibit juvenile hormone synthesis by earwig corpora allata, there is heat-stable material in earwig brain extract that does have this action. 相似文献
