褪黑激素对绒山羊皮肤中毛囊周期相关miRNAs表达模式的影响

褪黑激素(Melatonin, MT)和miRNAs在毛囊的生长发育过程中发挥重要的作用, 但MT对绒山羊皮肤毛囊miRNAs表达模式的影响尚未见报道。为探索MT从miRNAs层次影响山羊绒生长的机制, 文章在内蒙古绒山羊中实施了褪黑激素埋植试验：5只青年母羊作为实验组埋植褪黑激素, 另外5只青年母羊作为对照。利用荧光定量PCR检测褪黑激素埋植前后毛囊周期相关miRNAs的表达变化。结果表明, 埋植MT明显改变了6个绒毛相关miRNAs的表达规律：在一个绒毛周期内, 除let-7a外, miR-203、miR-205、miR-96、miR-183和miR-199a的表达量均发生3次跃迁;埋植MT改变了miRNAs之间的共表达模式。对照组各miRNA之间相关系数范围为0.87~0.99(P<0.01)。与对照组相比, 埋植组中let-7a与miR-96、miR-199a、miR-205, miR-203与miR-96、miR-199a, miR-96与miR-183, miR-183与miR-199a之间的相关系数被明显消弱;MT通过下调6月份埋植组各miRNA的表达量提早诱发二次生绒。     

Synthesis and structure-immunosuppressive activity relationships of bakuchiol and its derivatives

A series of derivatives of bakuchiol were synthesized and tested in vitro for their cytotoxicity, and inhibition of T cell proliferation and B cell proliferation. The data obtained provided preliminary structure-activity relationships of the compounds as immunosuppressive activity.     

Hymenobacter tibetensis sp. nov., a UV-resistant bacterium isolated from Qinghai–Tibet plateau

A brick-red-pigmented strain (XTM003^T) isolated from the Qinghai–Tibet plateau was investigated using a polyphasic taxonomy approach. Phylogenetic analyses based on 16S rRNA gene sequence indicated that the organism belonged to the genus Hymenobacter. The predominant menaquinone was MK7. The major fatty acids included iso-15:0, 16:1w5c and summed feature 3 (C_{16: 1}ω7c and/or C_{16: 1}ω6c). The G+C content of the DNA was 55.8%. In addition, DNA–DNA hybridization studies demonstrated that strain XTM003^T had a relatedness value of 50.7% with the phylogenetically most closely related species Hymenobacter norwichensis DSM 15439^T. Based on the results of phenotypic characteristics and DNA–DNA hybridization studies, strain XTM003^T is considered to represent a novel species, for which the name Hymenobacter tibetensis sp. nov. is proposed. The type strain is XTM003^T (=CCTCC AB 207089^T=NRRL B-51271^T).     

Density-dependent effects on the dieback of exotic species Spartina anglica in coastal China

The exotic species Spartina anglica C.E. Hubbard has been experiencing dieback in coastal China during the past decade, while it is invading other places in the world. Thus, an explanation for the dieback of the populations in coastal China would be helpful for controlling invasions of S. anglica in other places of the world. Both field and greenhouse experiments were conducted to determine the possible role of density in the dieback of the populations in China. The density effects were tested by maintaining six density levels in field experiments and three density levels in greenhouse experiments over 7 months. The results showed that growth, asexual reproduction and biomass accumulation in medium density were significantly higher than those in other density treatments for field experiments. The growth and asexual reproduction in high-density patches were lowest among the treatments in the field. Data showed the same pattern for the greenhouse experiments. The results implied that density regulation had a great effect on most of the studied parameters. Because the species is surviving at high density along the Chinese coast, and has a much lower growth rate due to strong intraspecific competition, and also because high-density populations may decline into low density populations due to resource exhaustion, high plant density might be one of the important factors in the dieback of S. anglica.     

Design,synthesis and biological evaluation of novel dual inhibitors of acetylcholinesterase and β-secretase

To explore novel effective drugs for the treatment of Alzheimer’s disease (AD), a series of dual inhibitors of acetylcholineterase (AChE) and β-secretase (BACE-1) were designed based on the multi-target-directed ligands strategy. Among them, inhibitor 28 exhibited good dual potency in enzyme inhibitory potency assay (BACE-1: IC₅₀ = 0.567 μM; AChE: IC₅₀ = 1.83 μM), and also showed excellent inhibitory effects on Aβ production of APP transfected HEK293 cells (IC₅₀ = 98.7 nM) and mild protective effect against hydrogen peroxide (H₂O₂)-induced PC12 cell injury. Encouragingly, intracerebroventricular injection of 28 into amyloid precursor protein (APP) transgenic mice caused a 29% reduction of Aβ_1–40 production. Therefore, 28 was demonstrated as a good lead compound for the further study and more importantly, the strategy of AChE and BACE-1 dual inhibitors might be a promising direction for developing novel drugs for AD patients.     

斑马鱼干扰素基因工程菌发酵条件及稳定性研究

采用液体传代的方法,研究表达斑马鱼干扰素(IFN)工程菌株pRSET-B-zIFN的遗传稳定性,并对其发酵工艺进行了优化.结果表明,构建的工程菌株遗传稳定性良好,在没有选择压力的情况下,传代100次质粒稳定性仍然保持100%,菌落形态及生长情况与原代菌无明显区别,SDS-PAGE结果显示zIFN表达量无明显差异.发酵条件研究表明,工程菌在LB培养基中,接种量为1%,诱导剂终浓度为0.1 mmol/L,zIFN的表达量最高.
