The DNA sequences encoding β-keratin have been obtained from Marsh Mugger (Crocodylus palustris) and Orinoco Crocodiles (Crocodylus intermedius). Through the deduced amino acid sequence, these proteins are rich in glycine, proline and serine. The central region of the proteins are composed of two beta-folded regions and show a high degree of identity with β-keratins of aves and squamates. This central part is thought to be the site of polymerization to build the framework of β-keratin filaments. It is believed that the β-keratins in reptiles and birds share a common ancestry. Near the C-terminal, these β-keratins contain a peptide rich in glycine-X and glycine-X-X, and the distinctive feature of the region is some 12-amino acid repeats, which are similar to the 13-amino acid repeats in chick scale keratin but absent from avian feather keratin. From our phylogenetic analysis, the β-keratins in crocodile have a closer relationship with avian keratins than the other keratins in reptiles.  相似文献

Patterns of intraspecific variation in functional traits have been widely studied across plant species to find out what general suites of traits provide functional advantage under specific environmental conditions. Much less is known about this variation within tree species and, in particular, about its relationship with performance variables such as photosynthetic rates under water deficit. Nevertheless, this knowledge is fundamental to understand the adaptive potential of drought sensitive tree species to increased aridity as predicted in the context of climate change.Intraspecific variation in photosynthetic performance and other leaf functional traits in response to water availability were examined in a glasshouse experiment using seedlings of six European beech populations. The physiological response of seedlings to a “water stress” treatment was compared to a “control” treatment along an experimental cycle of progressive soil water deficit and recovery. We found evidence of intraspecific variation in beech's photosynthetic performance and other leaf functional traits in response to water availability. We also detected intraspecific variation in leaf-level tolerance of water deficit and phenotypic plasticity to water availability suggesting a pattern shaped by both regional and local scale effects. The Swedish population was particularly sensitive to water deficit, being the only population showing impaired photochemical efficiency under the experimental water deficit. Leaf-level tolerance of water deficit was related to PNUE, but not to other functional traits, such as WUE, SLA or leaf nitrogen content, that have been described to vary across species in adaptation to drought tolerance. Our results support the idea that general trends for variation in functional traits across species do not necessarily reflect a similar pattern when observed at the intraspecific level. The observed functional variation between beech populations reaffirms the importance of local adaptation to water deficit in the context of climate change.  相似文献

The addition of 1.5 mM putrescine to the culture medium facilitated the conversion of protocorm-like bodies (PLBs) of Dendrobium officinale to shoots. The administration of 1.0 mM of the polyamine inhibitor, α-DL-difluoromethylarginine, decreased the conversion of PLBs to shoots. Compared to the control, the PLBs treated with 1.5 mM putrescine had higher levels of total intracellular free polyamines. The enhanced conversion of PLBs to shoots following the addition of putrescine was accompanied by an increase in the ratio of total cytokinins (CTKs) to indole-3-acetic acid (IAA). Analysis of enzyme activities indicated that the increased endogenous level of total CTKs driven by putrescine was associated with the inhibition of CTK decomposition by CTK oxidase, while the decreased endogenous level of IAA was associated with the promotion of IAA decomposition by IAA oxidase. In addition, putrescine increased the ratio of reduced glutathione to oxidized glutathione in the conversion process of PLBs to shoots.  相似文献

Although mechanical isolation mediated by shared pollinators has been considered as a classic model of pollinator-mediated floral isolation in Pedicularis, a superdiverse genus in Hengduan Mountains, southwest China, there has been no empirical study of interspecific pollen flow between closely related species. We examined reproductive barriers at six different stages between Pedicularis cranolopha and Pedicularis tricolor, two sister species. The two sister species were geographically isolated from each other based on our field survey and the records of herbarium specimens. Translocation experiments showed that flowering phenology partly overlapped and bumblebee pollinators did not discriminate between flowers of the two species. Bumblebee interspecific moves could mediate interspecific pollination as traced using fluorescent powder, in which pollen analogs placed on one species were transferred to the stigmas of the other species in experimental plots containing both species. Heterospecific pollen tubes grew in the style as well as conspecific pollen in hand-pollination experiments. Reciprocal hybridization between the two species could produce (partially) viable seeds, suggesting weak post-pollination barriers. Our results showed that geographic isolation was an important barrier of two species, and the total reproductive isolation between two species was incomplete when without geographical isolation. The formation of Big Snow Mountains could introduce an important pre-zygotic reproductive barrier between the two sister species of Pedicularis; such geographical isolation could be responsible for allopatric speciation, giving a clue to understanding the rapid radiation on mountain areas.  相似文献

琴蛙属(Nidirana)是广泛分布于东洋界的两栖动物,目前安徽省琴蛙属仅记录了弹琴蛙 (N. adenopleura)1种。2022年7月在安徽省黄山地区(九龙峰自然保护区和马金岭自然保护区)采集到9号两栖动物标本,其形态特征与孟闻琴蛙(N. mangveni)相似;此外,基于线粒体COI基因片段的分子系统发育分析显示,此次采集到的琴蛙标本与孟闻琴蛙聚为一支,且具有高的支持率(贝叶斯后验概率/最大似然支持率为1.00/98%),而且和孟闻琴蛙遗传距离最近,与模式产地的孟闻琴蛙遗传距离仅有0.04%。因此,结合形态比较和分子系统学分析,鉴定此次采集的琴蛙属标本为孟闻琴蛙,为安徽省两栖动物分布新记录种。这一发现将孟闻琴蛙的分布范围扩展至皖南地区,为琴蛙属和皖南地区的生物地理学和地史研究提供了重要数据。  相似文献

An accurate understanding of species diversity is essential to studies across a wide range of biological subdisciplines. However, species delimitation remains challenging in evolutionary radiations, particularly in those herbaceous plants associated with microendemic, naturally fragmented distribution systems, where genotypic and phenotypic traits likely evolved discordantly. The Primula merrilliana complex, which is endemic to eastern China and has high horticultural value, used to be treated as one species but several clues suggested it might be composed of multiple species. Here we used multiple lines of evidence, including molecular, morphological, reproductive isolation, and geographic data, to assess independently evolving lineages within this complex. Our results indicated that the species diversity in the complex was underestimated previously, and four species (independently evolving lineages) can be recognized, including two new species described here. The extensive variation of the breeding system, especially the floral morph transition from distyled (outcrossing) to homostyled (selfing) multiple times, possibly promoted the rapid speciation within such a small geographic scale. This study case indicated that the phenomenon of genetically highly divergent but morphologically indistinguishable is perhaps shown in herbs with fragmented distributions; the alternative extreme evolutionary phenomenon, in which complete reproductive barriers have been accumulated but with little genetic differentiation, also exists. Thus we highlight the importance of incorporating other characters, such as postzygotic reproductive isolation and geographic data, with commonly used molecular and morphological traits to infer species boundaries through an integrative taxonomic approach in such systems.  相似文献

This study used the mitochondrial DNA cytochrome b and control region sequences to examine the population genetic structure of Microphysogobio brevirostris, an endemic cyprinid species in northern Taiwan. In total, 90 specimens were collected from six populations belonging to four major rivers. All haplotypes were identified as two major clades (Clades N and S), exhibiting a southern and a northern distribution. The results of statistical dispersal-vicariance analysis (S-DIVA) revealed that two vicariance events and one dispersal event shaped the phylogeographic pattern of M. brevirostris. These results suggest that the ancestral populations of M. brevirostris are widely distributed throughout northern Taiwan, north of the Miaoli Plateau. The ancestral populations then diverged into two clades (N and S) by the raising of the Taoyuan Plateau. Finally, two major rivers of the S clade diverged and dispersed into adjacent rivers. Besides, our results revealed that Microphysogobio alticorpus and M. brevirostris colonized Taiwan during the same ice age through different colonization routes.  相似文献

The mitochondrial control regions (CRs) and flanking sequences of Pelodiscus sinensis, Apalone ferox, Palea steindachneri and Carettochelys insculpta were obtained using Long-PCR with gene-specific primers. The CR lengths of the four species were 1843 bp, 1356 bp, 1725 bp, and 969 bp. The base composition percentages of A+T were 60.5%, 60.7%, 65.7%, 64.7%, respectively. Combined with CR sequences of other three soft-shelled turtles published in GenBank (Pelodiscus sinensis, Korea, AY962573; Dogania subplana, AF366350; Lissemys punctata, EF050073), we compared the CR structures and identified three functional domains (TAS, CD and CSB) in which conserved sequence blocks (TAS, CSB -F, CSB-1, CSB-2 and CSB-3) were also successfully identified according to their sequence similarities to those of other turtles. The variable numbers of tandem repeats (VNTRs 1) with 50–52 bp motif were identified at 5′-end of CR among the five soft-shelled turtles P. sinensis (China), P. sinensis (Korea), A. ferox, P. steindachneri, D. subplana. The copy number of the VNTRs varied from 5 to 15. VNTRs 2 with 2–11 bp motif were identified in the 3′- end of CR among all of the six soft-shelled turtles with variable number of motifs from 4 to 29. Moreover, VNTRs 3 with 6 bp motif were identified between CSB-1 and CSB-2 of CR both in P. sinensis (China) and P. sinensis (Korea), in which the number of motifs varied from 19 to 29. The types and distribution of VNTRs of the six soft-shelled turtles were also discussed. With Alligator mississippiensis as an outgroup, combined with the CR sequences (excluding VNTRs) of other five turtles which were published in GenBank, the molecular phylogenetic trees were constructed using PAUP 4.0b10 and MrBayes ver. 3.0. The results strongly supported the monophyly of Carretochelyidae and Carettochelyidae as sister group to an assemblage of Cryptodira. Our research suggested that the earliest phylogenetic tree splits into three separated basal branches; the Pelomedusidira (Pelomedusa subrufa), the Carettochelyidae (C. insculpta), and an assemblage of Cryptodira and the C. insculpta that might be a representation of distinctive suborder.  相似文献
