Actinomycetes are one of the most valuable sources of natural products with industrial and medicinal importance. After more than half a century of exploitation, it has become increasingly challenging to find novel natural products with useful properties as the same known compounds are often repeatedly re-discovered when using traditional approaches. Modern genome mining approaches have led to the discovery of new biosynthetic gene clusters, thus indicating that actinomycetes still harbor a huge unexploited potential to produce novel natural products. In recent years, innovative synthetic biology and metabolic engineering tools have greatly accelerated the discovery of new natural products and the engineering of actinomycetes. In the first part of this review, we outline the successful application of metabolic engineering to optimize natural product production, focusing on the use of multi-omics data, genome-scale metabolic models, rational approaches to balance precursor pools, and the engineering of regulatory genes and regulatory elements. In the second part, we summarize the recent advances of synthetic biology for actinomycetal metabolic engineering including cluster assembly, cloning and expression, CRISPR/Cas9 technologies, and chassis strain development for natural product overproduction and discovery. Finally, we describe new advances in reprogramming biosynthetic pathways through polyketide synthase and non-ribosomal peptide synthetase engineering. These new developments are expected to revitalize discovery and development of new natural products with medicinal and other industrial applications.  相似文献

为鉴定籼粳稻杂交衍生系的苗期抗旱性,以课题组自育的高代抗逆品系ZD15为母本、籼稻品种IR29为父本,以及杂交衍生的重组自交系群体120份为试验材料,利用PEG-6000对各材料苗期进行干旱胁迫处理,测定根长、根冠比、地上部鲜重、地下部鲜重、地上部干重和地下部干重;利用PEG-6000对各材料芽期进行干旱胁迫处理,测定芽鞘长和芽长。采用主成分分析和隶属函数法对各材料的抗旱性进行综合评价,根据综合抗旱D值可将122份材料分成3类,D值在0.201~0.400之间的有33份,属于不抗旱材料;D值在0.401~0.600之间的有79份,属于中等抗旱材料;D值在0.601~0.800之间的有10份,属于抗旱材料。利用D值进行逐步回归分析,结果表明根长、根冠比、地上部鲜重、地下部鲜重、地上部干重、地下部干重、芽鞘长和芽长8个性状均可作为水稻苗期抗旱性的评价指标。本研究筛选出的抗旱材料,可作为育种中间材料进一步培育,或作为育种资源加以利用,以丰富本区水稻育种的资源库。  相似文献

Due to the lack of complete understanding of metabolic networks and reaction pathways, establishing a universal mechanistic model for mammalian cell culture processes remains a challenge. Contrarily, data-driven approaches for modeling these processes lack extrapolation capabilities. Hybrid modeling is a technique that exploits the synergy between the two modeling methods. Although mammalian cell cultures are among the most relevant processes in biotechnology and indeed looks ideal for hybrid modeling, their application has only been proposed but never developed in the literature. This study provides a quantitative assessment of the improvement brought by hybrid models with respect to the state-of-the-art statistical predictive models in the context of therapeutic protein production. This is illustrated using a dataset obtained from a 3.5 L fed-batch experiment. With the goal to robustly define the process design space, hybrid models reveal a superior capability to predict the time evolution of different process variables using only the initial and process conditions in comparison to the statistical models. Hybrid models not only feature more accurate prediction results but also demonstrate better robustness and extrapolation capabilities. For the future application, this study highlights the added value of hybrid modeling for model-based process optimization and design of experiments.  相似文献

Postcopulatory sexual selection may promote evolutionary diversification in sperm form, but the contribution of between‐species divergence in sperm morphology to the origin of reproductive isolation and speciation remains little understood. To assess the possible role of sperm diversification in reproductive isolation, we studied sperm morphology in two closely related bird species, the common nightingale (Luscinia megarhynchos) and the thrush nightingale (Luscinia luscinia), that hybridize in a secondary contact zone spanning Central and Eastern Europe. We found: (1) striking divergence between the species in total sperm length, accompanied by a difference in the length of the mitochondrial sperm component; (2) greater divergence between species in sperm morphology in sympatry than in allopatry, with evidence for character displacement in sperm head length detected in L. megarhynchos; (3) interspecific hybrids showing sperm with a length intermediate between the parental species, but no evidence for decreased sperm quality (the proportion of abnormal spermatozoa in ejaculates). Our results demonstrate that divergence in sperm morphology between the two nightingale species does not result in intrinsic postzygotic isolation, but may contribute to postcopulatory prezygotic isolation. This isolation could be strengthened in sympatry by reinforcement.  相似文献

×Taxodiomeria peizhongiiZ. J. Ye, J. J. Zhang et S. H. Pan was regarded as a new intergeneric hybrid between Taxodium mucronatum Tenore (as the female donor) and Cryptomeria fortunei Hooibrenk ex Otto et Dietr (as the male donor). To confirm the authenticity of the intergeneric hybrid, we analyzed the rbcL gene and the internal transcribed spacer (ITS) of 26S-18S ribosomal RNA gene of the three species using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and arbitrarily primed PCR (AP-PCR), and obtained the following results: i) Taxodiomeria peizhongii had the same RFLP maps of the rbcL gene and the ITS as Taxodium mucronatum, but was different from C. fortunei; ii) a 311-bp PCR amplification product was obtained in C. fortunei by AP-PCR of ITS, but was not found in Taxodiomeria peizhongii. Our results have demonstrated that C. fortunei did not provide any genome for Taxodiomeria peizhongii, implying that T. peizhongii is not an intergeneric hybrid between the two species.  相似文献

Heterosis, or hybrid vigor, has recently been proposed as a factor promoting invasive growth of some non-indigenous aquatic plant species, particularly those capable of spreading rapidly within and among lakes through clonal reproduction. We tested this hypothesis for variable-leaf water milfoil (Myriophyllum heterophyllum), a non-indigenous aquatic plant that has become a major management and conservation concern in New England. Using nuclear ribosomal DNA, we looked for F1 hybrid populations of invasive M. heterophyllum in 25 New Hampshire (NH) lakes. In contrast to a previous study that found F1 hybrid lineages of invasive M. heterophyllum in Connecticut, we did not find hybrids in our study lakes. This result has two implications: (1) pure lineages of M. heterophyllum are also capable of invasive growth, and (2) the distribution of invasive M. heterophyllum lineages (hybrid vs. pure) may be spatially structured across New England. We stress the importance of more detailed distributional and ecological studies for understanding the invasive potential of this species.  相似文献

Two partly modified insect-resistant genes (BtCryI Ac gene [Bt gene toxin against Lepidopterean insects] and API gene [arrowhead proteinase inhibitor]) were transferred to the triploid hybrid of Chinese white poplar ((Populus tomentosa Carr. × Populus bolleana Louche) × Populus tomentosa Carr.) mediated by A. tumefaciens. The survival of Agrobacterium in transgenic plants was examined during the processes of transplanting and subculturing on the nutrient medium. The results suggested that 80% of the plants, which were obtained by repeated selection on media added with 50 mg/L kanamycin and 300 mg/l carbenicillin, showed positive reactions after examination using molecular methods. The ELISA test indicated that the Bt toxoprotein was expressed in seven of the transgenic sub-clones. Leaves, stems, and roots of all the 28 transgenic plants were cultured on the YEB medium added with 50 mg/L kanamycin, and it was found that Agrobacterium survived in three sub-clones (33, 37, 5) and could have existed for 24 months in the bottle. These three transgenic sub-clones were transplanted and cultivated for one month in the room, and then the target Agrobacterium was found in rhizosphere of the sub-clone 33.  相似文献

为解析苹果对炭疽叶枯病的抗性机制,该研究以‘嘎拉’、‘藤牧1号’苹果及其杂交后代等87个苹果品种(系)为试验材料,进行田间调查及人工接种病菌并检测不同品种(系)中病菌生物量,利用SSR分子标记进行基因型鉴定,分析各品种(系)对炭疽叶枯病的抗性差异,利用荧光定量PCR及酶活检测比较分析水杨酸相关基因、抗性酶基因及酶活性水平差异。结果表明:(1)87个苹果品种(系)对苹果炭疽叶枯病的抗性差异明显,‘嘎拉’、‘2 5’、‘19 19’等品种(系)叶片发病严重,表现出对炭疽叶枯病的高感性,‘藤牧1号’、‘40 9’及‘16 16’等品种(系)叶片无病斑或病斑极少,炭疽叶枯病菌含量显著低于感病性品种(系),其抗性显著。(2)SSR标记S0405127和S0304673的基因型鉴定结果与田间表型调查结果相比,准确率分别为93.10%和91.95%,与人工接种结果相比,准确率分别为91.95%和95.40%。(3)SA相关基因的表达模式结果表明,接种炭疽叶枯病菌4 d后,‘藤牧1号’、‘40 9’及‘16 16’等抗性品种(系)中SA合成相关基因MdEDS1、MdPAD4和MdPAL被强烈诱导表达;同时,SA信号转导相关基因MdNPR1、MdPR1、MdPR5的表达显著高于‘嘎拉’等感病品种(系)。(4)接种炭疽叶枯病菌4 d后,‘藤牧1号’、‘40 9’及‘16 16’等抗性品种(系)的MdSODMdPOD酶基因表达水平及酶活性显著高于‘嘎拉’、‘2 5’、‘19 19’等感病品种(系)。研究认为,‘藤牧1号’、‘40 9’及‘16 16’等品种(系)通过调控水杨酸合成和信号转导通路及氧化还原相关反应等提高对炭疽叶枯病的抗性,为挖掘抗性基因以及利用优良种质选育抗病品种奠定了基础。  相似文献

Inorganic–organic hybrid nanoparticles formed by lanthanide-doped nanostructures and organic ligands have been intensively studied, which could greatly increase their photoluminescence performance as a result of the energy transfer process from organic ligands to Ln3+ ions. However, the photoluminescence intensity and excitation spectral width are still quite limited on coordinating with a single type of organic ligand. In this work, Eu3+-doped LaF3 (LaF3:Eu3+) nanoparticles were prepared using a hydrothermal method, and were then hybridized with benzoic acid and thenoyltrifluoroacetone to form the hybrid nanostructures. After that, the hybrid nanostructures were mixed with 2,2′-azobisisobutyronitrile and methyl methacrylate to prepare the composites. The sample obtained by hybridization and composite doping with 5% Eu3+ exhibited the best photoluminescence performance. The excitation peak width and luminescence intensity of the hybrid nanostructures were significantly increased. The excitation spectral width of the inorganic–organic mixed hybrid nanostructures was particularly enhanced, and covered the whole ultraviolet band region of solar light on Earth. The prepared composites exhibited good optical properties.  相似文献
