Advances in salivary bioscience enable the widespread integration of biological measures into the behavioral and social sciences. While theoretical integration has progressed, much less attention has focused on analytical strategies and tactics. The statistical literature warns that common methods for comparing groups and studying associations can have relatively poor power compared to more modern robust techniques. Here we illustrate, in secondary data analyses using the USC Well Elderly II study (n = 460, age 60–95, 66% female), that modern robust methods make a substantial difference when analyzing relations between salivary analyte and behavioral data. Analyses that deal with the diurnal pattern of cortisol and the association of the cortisol awakening response with depressive symptoms and physical well-being are reported. Non-significant results become significant when using improved methods for dealing with skewed distributions and outliers. Analytical strategies and tactics that employ modern robust methods have the potential to reduce the probability of both Type I and Type II errors in studies that compare salivary analytes between groups, across time, or examine associations with salivary analyte levels. 相似文献

The molecular arrangement within a lamellar structure composed of human erythrocyte lipids is determined. The 45 A thick lipid layer, in water, is filled in the interior with a liquid-like configuration of the hydrocarbon chains of phospholipid molecules and is covered on both sides by their hydrophilic polar groups. Cholesterol is located so that part of its steroid nucleus is between the polar groups of the phospholipid molecules while the rest of the molecule extends into the inner hydrocarbon layer. This lipid leaflet would be expected to have the mechanical properties of a purely liquid surface, as other authors have shown for the "black" lipid membranes. Data are presented which demonstrate that the intact erythrocyte membrane is a tough viscoelastic substance with a Young''s modulus of 106–108 dynes/cm2 and a viscosity of 107–1010 poises. The parameters and the kinetics of membrane breakdown are incompatible with the model system of pure lipid. Caution must be exercised in applying various data on the model systems to intact membranes. 相似文献

Most cats surrendered to nonhuman animal shelters are identified as unowned, and the surrender reason for these cats is usually simply recorded as “stray.” A cross-sectional study was conducted with people surrendering cats to 4 Australian animal shelters. Surrenderers of unowned cats commonly gave surrender reasons relating to concern for the cat and his/her welfare. Seventeen percent of noncaregivers had considered adopting the cat. Barriers to assuming ownership most commonly related to responsible ownership concerns. Unwanted kittens commonly contributed to the decision to surrender for both caregivers and noncaregivers. Nonowners gave more surrender reasons than owners, although many owners also gave multiple surrender reasons. These findings highlight the multifactorial nature of the decision-making process leading to surrender and demonstrate that recording only one reason for surrender does not capture the complexity of the surrender decision. Collecting information about multiple reasons for surrender, particularly reasons for surrender of unowned cats and barriers to assuming ownership, could help to develop strategies to reduce the number of cats surrendered. 相似文献

Fluorescent and luminescent gene reporters allow us to dynamically quantify changes in molecular species concentration over time on the single cell level. Coagulation factor VIIa (FVIIa) is a serine protease that, after binding to tissue factor (TF), plays a pivotal role in the initiation of blood coagulation. We used hydrogen exchange monitored by mass spectrometry to visualize the details of FVIIa activation by comparing the exchange kinetics of distinct molecular states, namely zymogen FVII, endoproteolytically cleaved FVIIa, TF-bound zymogen FVII, TF-bound FVIIa, and FVIIa in complex with an active site inhibitor. The hydrogen exchange kinetics of zymogen FVII and FVIIa are identical indicating highly similar solution structures. However, upon tissue factor binding, FVIIa undergoes dramatic structural stabilization as indicated by decreased exchange rates localized throughout the protease domain and in distant parts of the light chain, spanning across 50A and revealing a concerted interplay between functional sites in FVIIa. The results provide novel insights into the cofactor-induced activation of this important protease and reveal the potential for allosteric regulation in the trypsin family of proteases. 相似文献

Explaining cooperation remains a central topic for evolutionary theorists. Many have argued that group selection provides such an explanation: theoretical models show that intergroup competition could have given rise to cooperation that is costly for the individual. Whether group selection actually did play an important role in the evolution of human cooperation, however, is much debated. Recent experiments have shown that intergroup competitions do increase human cooperation, which has been taken as evidence for group selection as a mechanism for the evolution of cooperation. Here we challenge this standard interpretation. Competitions change the payoff structure by creating a threshold effect whereby the group that contributes more earns an additional prize, which creates some incentive for individuals to cooperate. We present four studies that disentangle competition and thresholds, and strongly suggest that it is thresholds – rather than competitions per se – that increase cooperation. Thus, prior intergroup competition experiments provide no evidence of a unique or special role for intergroup competition in promoting human cooperation, and shed no light on whether group selection shaped human evolution. 相似文献

In almost all finite element (FE) studies in dentistry, virtual forces are applied directly to dentures. The purpose of this study was to develop a FE model with non-linear contact simulation using an antagonist as force transmitter and to compare this with a similar model that uses direct force transmission. Furthermore, five contact situations were created in order to examine their influence on the peri-implant bone stresses, which are relevant to the survival rate of implants. Biological variation exists across a nested set of hierarchical levels from nucleotides within genes to populations within
species to lineages within the tree of life. How selection acts across this hierarchy is a long-standing question in evolutionary
biology. Recent studies have suggested that genome size is influenced largely by the balance of selection, mutation and drift
in lineages with different population sizes. Here we use population cage and maternal transmission experiments to identify
the relative strength of selection at an individual and cytoplasmic level. No significant trends were observed in the frequency
of large (L) and small (S) mtDNAs across 14 generations in population cages. In all replicate cages, new length variants were
observed in heteroplasmic states indicating that spontaneous length mutations occurred in these experimental populations.
Heteroplasmic flies carrying L genomes were more frequent than those carrying S genomes suggesting an asymmetric mutation
dynamic from larger to smaller mtDNAs. Mother-offspring transmission of heteroplasmy showed that the L mtDNA increased in
frequency within flies both between and within generations despite sampling drift of the same intensity as occurred in population
cages. These results suggest that selection for mtDNA size is stronger at the cytoplasmic than at the organismal level. The
fixation of novel mtDNAs within and between species requires a transient intracellular heteroplasmic stage. The balance of
population genetic forces at the cytoplasmic and individual levels governs the units of selection on mtDNA, and has implications
for evolutionary inference as well as for the effects of mtDNA mutations on fitness, disease and aging. 相似文献
