We examined the effect of low density lipoprotein (LDL) on histamine release from purified human lung mast cells. LDL inhibited anti-IgE- induced histamine release in a dose-dependent manner, with 100 micrograms/ml LDL-protein inhibiting histamine release by 53 +/- 8% (mean +/- SEM); half-maximal inhibition occurred at 40-80 micrograms/ml. LDL also inhibited calcium ionophore A23187-induced histamine release in a dose-dependent manner, with 1 mg/ml of LDL inhibiting histamine release by 83 +/- 9%; half maximal inhibition occurred at 220-280 micrograms/ml. Inhibition by LDL was time-dependent: half-maximal inhibition of anti-IgE- induced histamine release by LDL occurred at 30-50 minutes of incubation. The inhibitory effect of LDL was independent of buffer calcium concentrations (0-5 mM) or temperature (0-37 degrees C). Bulk water velocities and local relative velocities generated in experimental tanks around and within thalli of free moving Gracilaria conferta were estimated according to the dissolution rate of benzoic acid sticks. Boundary-layer thickness and HCO

-mass-transfer coefficient were derived from the water velocities. Average relative velocities varied between 12 cm s –1 to less than 0.1 cm s –1 as a function of the absolute water flow in the tank, alga shape and location within the thallus. The lower range of velocities was observed at 20% of maximum aeration in the inner part of the plant. In laboratory experiments, photosynthetic rates, as determined in a closed Clark-type O2-electrode system, increased by 30%–50% when water velocity was increased from zero to about 1.5 cm s –1. Another minor increase was obtained between 1.5 cm s –1 and 8 cm s –1 water velocity. This response to water motion was affected by bulk inorganic carbon concentration and by plant condition, as was reflected from the differences in the response in the winter and spring. It might be suggested that under carbon saturation, water velocity above 2 cm s–1 provided almost sufficient flow to saturate carbon uptake. 相似文献

Assimilate translocation has been identified and characterized in Gracilaria cornea under different conditions. Carbon fixation and translocation were carried out by inserting the base part of the thallus into a bicarbonate labeled solution in seawater and exposing its upper part to the air (open system) or to a non-labeled solution above a rubber septum (closed system). After a pulse-chase treatment in the light, three separate sections of each thallus were extracted by DMF (high moleuclar weight photosynthates) or by ethanol (low molecular weight). The results indicate a high rate of active photosynthate translocation which is directly related to inorganic carbon gradients in the thallus, and probably also to sugar gradients in the thallus. Translocation parameters of Gracilaria cornea are lower than of brown algae, as Gracilaria does not contain specific translocation tissues. 相似文献

Summary Cellular subclones of high and low tumorigenicity obtained from a mouse c-Ha-ras-transformed clone, were examined for their sensitivity to tumornecrosis-factor (TNF)-mediated cytotoxicity. Cells of the highly tumorigenic subclones showed a significantly enhanced resistance to the cytotoxic effect of TNF plus cyclohexamide (CHI) as compared to cells of the lowtumorigenic subclones. The enhanced resistance to TNF+CHI was not due to a lower expression of TNF receptors on the cells. The c-Ha-ras-transfected cells were transformed and maintained in culture only (C cells). In vivo passage of cells of the initially low-tumorigenic c-Ha-ras subclones through the mouse significantly enhanced the tumorigenic potential of these CTC cells (culture/tumor/culture). In correlation with their enhanced tumorigenicity, the CTC cells were highly resistant to TNF-mediated cytotoxicity as compared to C cells of the same subclone. Furthermore, the involvement of TNF in determining the tumorigenic phenotype of the c-Ha-ras-transformed cells was demonstrated in a more direct manner. Cells of a c-Ha-ras-transformed low-tumorigenic, highly TNF-sensitive subclone were selected by repeated cycles of in vitro exposure to TNF. As a result, a stable, highly TNF-resistant population of cells emerged. These TNF-resistant cells caused more tumors in mice as compared to their original TNF-sensitive cells. These results show that the resistance to the cytotoxic effect of TNF plus cyclohexamide may be involved, at least partially, in the tumorigenic potential of c-Ha-ras-transformed cells and suggest a possible role for TNF in the enhancement of the tumorigenic potential of these cells in mice. 相似文献

S. granarius was acclimated to high temperatures by exposing adults to 32° for 14 days and by continuously rearing at 30° for four generations. The insects were then exposed for one week to 35° and mortality and number of offspring produced per live female were followed over 4 weeks. S. granarius reared at 30° did not show any signs of acclimation judging by mortality data, nor by reproduction data. Insects acclimated by prior exposure to 32° showed some degree of acclimation in terms of mortality, but not in the number of offspring produced after exposure to 35°. The acclimation procedures themselves did not affect reproduction or survival.
Zusammenfassung S. granarius wurde an hohe Temperaturen akklimatisiert durch 14-tägiges Aussetzen ausgewachsener Tiere einer Temperatur von 32° sowie durch stetiges Züchten, über vier Generationen, bei 30°. Die Insekten wurden dann eine Woche lang einer Temperature von 35° ausgesetzt und anschließend in 26,5° verbracht. Sterblichkeits- und Nachkommenschaftszahlen wurden 4 Wochen lang verfolgt.Nach diesen Daten beurteilt zeigten S. granarius, bei 30° gezüchtet, keine Akklimatisierung, jedoch Insekten die bei 32° ausgesetzt waren, bewiesen einen gewissen Grad von Akklimatisierung durch ihre Sterblichkeitsdaten. Kein Unterschied in der Nachkommenschaftszahl wurde nach Aussetzen auf 35° zwischen akklimatisierten und nicht-akklimatisierten Insekten gefunden.Die Akklimatisierungsart hat Vermehrung und Überleben nicht beeinflusst.相似文献

We determined the effects of various degrees of chemical modification of low-density lipoprotein (LDL) on its interaction with receptors present on human fibroblasts, human monocyte-derived macrophages and rat peritoneal macrophages. We isolated LDL (d = 1.019-1.063 g/ml) and carbamylated different numbers of lysine residues and tested its cell-interactive properties, including binding, degradation, and stimulation of [3H]oleate incorporation into cholesteryl oleate. Small carbamylation of LDL (approximately 1-2% of lysine residues) resulted in a reduced ability (70-80% of control) to displace 125I-labeled LDL from fibroblast receptors. Modification of 12.5-25% of lysine residues resulted in a marked increase in the ability of LDL to interact with scavenger receptors and an almost total loss in the ability to interact with apolipoprotein B-E receptors. Acetylated LDL and malondialdehyde-modified LDL inhibited competitively the degradation of 125I-carbamylated LDL by human macrophages. Thus, the extent of modification plays an important role in recognition of modified LDL by scavenger receptors. There also seems to be a range of modification over which LDL is not yet recognized by the scavenger receptor, but its interaction with the apolipoprotein B-E receptor is markedly reduced. This perhaps explains how a small in vivo modification of LDL can result in an increase in residence time of LDL in the subendothelial tissue which can lead to further local interactions, ultimately increasing the atherogenicity of the LDL particle. 相似文献

Effects of energy intake on the estrous cycle of the desert gerbillid, Sundevall's jird (Meriones crassus; 80 g; n=22) were studied. Females were offered either maintenance or below maintenance levels of millet seeds and ad lib. Atriplex halimus leaves and stems; drinking water was not available. Vaginal smears were used to determine sexual stage. We hypothesized that the estrous cycle ceases at low levels of energy intake and commences when sufficient energy is available. Females lost body mass linearly with a decrease in metabolizable energy intake. Estrous cycle averaged 4.46 d at maintenance energy intake but increased to an average of 7.81 d at 70% of maintenance energy intake. A cessation of the cycle occurred at an energy intake below 70% of maintenance requirements, which resulted in a body mass loss of more than 1% per day. More variability in the length of the different stages of the cycle was found with lower levels of energy intake. When offered ad lib. millet seeds and A. halimus (n=14), recovery to the normal cycle was attained within 10 d by 43% of the females and within 16 d by the rest of the females. Recovery time was longest in females that previously had the lowest energy intake. We concluded that the estrous cycle of M. crassus is sensitive to energy intake. With restricted energy intake, the estrous cycle and reproductive activities cease but can be restored with provision of adequate energy. The ubiquitous members of the aquaporin (AQP) family form transmembrane pores that are either exclusive for water (aquaporins) or are also permeable for other small neutral solutes such as glycerol (aquaglyceroporins). The purpose of this review is to provide an overview of our current knowledge of AQP structures and to describe the structural features that define the function of these membrane pores. The review will discuss the mechanisms governing water conduction, proton exclusion and substrate specificity, and how the pore permeability is regulated in different members of the AQP family. 相似文献
