Studies of the effects of increases in ultraviolet-B (UV-B) radiation on plants and terrestrial ecosystems have been undertaken using a variety of methods including: controlled-environment cabinets, glasshouses, outdoor filtration and outdoor supplementation using fluorescent UV-B lamps. Outdoor supplementation systems provide a method of study which creates only small alterations to the microclimate and the number of such studies has increased during the past 3 years. These supplementation systems differ in their methods of operation, equipment, UV-B exposure regime and experimental design. This essay surveys the systems currently developed, considers problems associated with their use and discusses these in relation to the interpretation of biological effects.  相似文献

In the final stage of radiation damage in cryo-electron microscopy of proteins, bubbles of hydrogen gas are generated. Proteins embedded in DNA bubble sooner than free-standing proteins and DNA does not bubble under the same conditions. These properties make it possible to distinguish protein from DNA. Here we explored the scope of this technique (“bubblegram imaging”) by applying it to bacteriophage T7, viewed as a partially defined model system. T7 has a thin-walled icosahedral capsid, 60 nm in diameter, with a barrel-shaped protein core under one of its twelve vertices (the portal vertex). The core is densely wrapped with DNA but details of their interaction and how their injection into a host bacterium is coordinated are lacking. With short (10 s) intervals between exposures of 17 electrons/Å2 each, bubbling starts in the third exposure, with 1–4 bubbles nucleating in the core: in subsequent exposures, these bubbles grow and merge. A 3D reconstruction from fifth-exposure images depicts a bipartite cylindrical gas cloud in the core. In its portal-proximal half, the axial region is gaseous whereas in the portal-distal half, it is occupied by a 3 nm-wide dense rod. We propose that they respectively represent core protein and an end of the packaged genome, poised for injection into a host cell. Single bubbles at other sites may represent residual scaffolding protein. Thus, bubbling depends on dose rate, protein amount, and tightness of the DNA seal.  相似文献

ObjectiveOur study aims to assess and track work load, working conditions and professional recognition of radiation oncology medical physicists (ROMPs) in the Asia Pacific Region over time.MethodsA structured questionnaire was mailed in 2008, 2011 and 2014 to senior medical physicists representing 23 countries. The questionnaire covers 7 themes: education and training including certification; staffing; typical tasks; professional organisations; resources; research and teaching; job satisfaction.ResultsAcross all surveys the response rate was >85% with the replies representing practice affecting more than half of the world’s population. The expectation of ROMP qualifications (MSc and between 1 and 3 years of clinical experience) has not changed much over the years.However, compared to 2008, the number of medical physicists in many countries has doubled. Formal professional certification is only available in a small number of countries. The number of experienced ROMPs is small in particular in low and middle income countries. The increase in staff numbers from 2008 to 2014 is matched by a similar increase in the number of treatment units which is accompanied by an increase in treatment complexity. Many ROMPs are required to work overtime and not many find time for research. Resource availability has only improved marginally and ROMPs still feel generally overworked, but professional recognition, while varying widely, appears to be improving slowly.ConclusionWhile number of physicists and complexity of treatment techniques and technologies have increased significantly, ROMP practice remains essentially unchanged over the last 6 years in the Asia Pacific Region.  相似文献

PURPOSE: The incidence of anaplastic lymphoma kinase (ALK) rearrangement in pulmonary sarcomatoid carcinoma (PSC) is controversial. In this study, we aimed to reveal the reliable frequency and the clinical-pathologic characteristics of pulmonary sarcomatoid carcinoma (PSC) with ALK rearrangement in Chinese population, and to provide insight into the translatability of anti-ALK treatment in this treatment-refractory disease. METHODS: Immunohistochemistry (IHC) using a Ventana anti-ALK (D5F3) rabbit monoclonal antibody was performed in 141 PSC specimens collected from multiple medical centers. IHC-positive cases were then confirmed using ALK fluorescent in situ hybridization (FISH). The incidence rates and clinical-pathologic characteristics of ALK-rearranged PSC were then analyzed. Response to ALK inhibitor crizotinib in a patient with ALK-rearranged PSC was evaluated according to the response evaluation criteria for solid tumors (RECIST) version 1.1. RESULTS: Five of 141 (3.5%) of PSCs showed ALK rearrangement-positive by IHC and then were confirmed by FISH. Two were carcinosarcomas and the other three were pulmonary pleomorphic carcinoma (PPC). Strong positive ALK rearrangement was observed in both the epithelioid and sarcomatoid components. The median age of ALK-positive patients was younger than that of ALK-negative patients. PSCs in never-smokers were more likely to harbor ALK rearrangement than those in former or current smokers (P < .05). A 40-year-old woman diagnosed with ALK-rearranged PPC experienced a partial response (?32%) to the ALK inhibitor crizotinib. CONCLUSIONS: The incidence rates of ALK rearrangement in PSC in the Chinese population are similar to those of other subtypes of NSCLC. PSCs in younger never-smokers are more often to harbor ALK rearrangement. ALK inhibitors may serve as an effective treatment for ALK-rearranged PSC.  相似文献

PurposeThis study aimed to assess the noise characteristics of ultra-high-resolution computed tomography (UHRCT) with deep learning-based reconstruction (DLR).MethodsTwo different diameters of water phantom were scanned with three different resolution acquisition modes. Images were reconstructed by filtered back projection (FBP), hybrid iterative reconstruction (hybrid-IR), and DLR. Image noise analysis was performed with noise magnitude, peak frequency (fp) of the noise power spectrum (NPS), and the square root of the area under the curve (√AUCNPS) for the NPS curve.ResultsThe noise magnitude was up to 3.30 times higher for the FBP acquired in SHR mode than that for the NR mode.The fp values of the FBP were 0.20–0.21, 0.34–0.36, and 0.34–0.37 cycles/mm for normal resolution (NR), high resolution (HR), and super high resolution (SHR) mode, respectively. The fp of hybrid-IR was 0.16–0.19, 0.21–0.26, and 0.23–0.26 cycles/mm for NR, HR, and SHR mode, respectively. The fp of DLR was 0.21–0.32 and 0.22–0.33 cycles/mm for HR and SHR mode, respectively.√AUCNPS showed that the highest value in FBP images of the SHR mode was up to 1.89 times that of the NR mode. DLR in the HR and SHR modes showed high noise reduction while suppressing fp shift with respect to FBP.ConclusionsThe new DLR algorithm could be a solution to the noise increase due to the high-definition detector elements and the small reconstruction matrix element size.  相似文献

The Pat1 protein of Saccharomyces cerevisiae was identified during a screen for proteins that interact with topoisomerase II. Previously, we have shown that pat1Δ mutants exhibit a slow-growth phenotype and an elevated frequency of both mitotic and meiotic chromosome mis-segregation. Here, we have studied the effects of deleting the PAT1 gene on chromosomal stability, with particular reference to rates of homologous recombination within the rDNA locus. This locus was analyzed because rDNA-specific hyperrecombination is known to occur in conditional top2 mutants. We show that pat1Δ strains mimic top2 mutants in displaying an elevated rate of intrachromosomal excision recombination at the rDNA locus, but not elsewhere in the genome. The elevated rate of recombination is dependent upon Rad52p, but not upon Rad51p or Rad54p. However, pat1Δ strains display additional manifestations of more general genomic instability, in that they show mild sensitivity to UV light and an increased incidence of interchromosomal recombination between heteroalleles. With a development of radiotherapeutic techniques, availability of radiotherapy data on cardiotoxicity, and slowly improving esophageal cancer outcomes, an increasing emphasis is placed on the heart protection in radiation treated esophageal cancer patients. Radiation induced heart complications encompass mainly pericardial disease, cardiomyopathy, coronary artery atherosclerosis, valvular heart disease, and arrhythmias. The most frequent toxicity is pericardial effusion which is usually asymptomatic in the majority of patients. The use of modern radiotherapy techniques is expected to reduce the risk of cardiotoxicity, although this expectation has to be confirmed by clinical data.  相似文献
