Metronidazole (2-methyl-5-nitroimidazole-1-ethanol) at 1–2 mM levels has been shown to be a selective inhibitor of nitrogenase activity in Anabaena. Two constitutive hydrogenases and photosynthesis are insensitive to metronidazole at these same concentrations. At higher concentrations metronidazole inhibits photosynthesis in Anabaena while photoreduction and to a lesser extent photohydrogen production are retarded in Scenedesmus. Respiration is slightly stimulated at high metronidazole levels in both algae. The reductant source for nitrogenase in Anabaena and photohydrogen production and photoreduction electron transport in Scenedesmus are discussed. Due to the activity of metronidazole as a selective inhibitor of ferredoxin-associated processes, it should prove to be useful in N2 fixation studies and in distinguishing between ferredoxin-linked reactions of different sensitivities and other activities not associated with low reduction potential components.  相似文献

In the absence of sufficient combined nitrogen, some filamentous cyanobacteria differentiate nitrogen-fixing heterocysts at approximately every 10th cell position. As cells between heterocysts grow and divide, this initial pattern is maintained by the differentiation of a single cell approximately midway between existing heterocysts. This paper introduces a mathematical model for the maintenance of the periodic pattern of heterocysts differentiated by Anabaena sp. strain PCC 7120 based on the current experimental knowledge of the system. The model equations describe a non-diffusing activator (HetR) and two inhibitors (PatS and HetN) that undergo diffusion in a growing one-dimensional domain. The inhibitors in this model have distinct diffusion rates and temporal expression patterns. These unique aspects of the model reflect recent experimental findings regarding the molecular interactions that regulate patterning in Anabaena. Output from the model is in good agreement with both the temporal and spatial characteristics of the pattern maintenance process observed experimentally.  相似文献

The present study demonstrated the growth of two species of cyanobacteria on wastewater isolated from sewage plant in Aswan, Egypt. We evaluated their efficiency for eliminating nitrogen, phosphorus, chemical oxygen demand (COD) and heavy metals (Fe2+, Pb2+, Cu2+, and Mn2+). The growth of Cyanosarcina fontana has supported wastewater as a growth medium than Anabaena oryzae compared to standard medium. The nutrients concentration such as COD, NO3–N and PO4–P were decreased by the growth of A. oryzae and C. fontana in the wastewater after primary settling and centrate. However, the reduction of COD was less efficient than the other nutrients. The reduction percentage of COD, NO3–N and PO4–P reached 39.3, 84.1 and 90.7% as well as 54.6, 83.1, and 89.8%, in cultures of A. oryzae and C. fontana grown in the wastewater after primary settling, respectively. The reduction amounted to 10.1, 76.8, and 63.0% by A. oryzae and 43.2, 62.1, and 74.8% by C. fontana, grown in the centrate, respectively. Cyanobacteria species have the ability to accumulate the heavy metals from the wastewater to level far than the exceeding metal level in the water. Whereas, the heavy metals biosorption performance of C. fontana was higher in accumulating Fe2+ (93.95%), Pb2+ (81.21%), Cu2+ (63.9%), and Mn2+ (48.49%) compared to A. oryzae. The biosorption ability is dependent on the nature of the adsorbent studied and the type of wastewater treated. Therefore, removal of heavy metals and nutrients by the tested algae is strongly recommended as a powerful technique for the removal of pollutants from wastewater.  相似文献

The photosynthetic bacteria (Rhodospirillum rubrum, Synechococcus and Anabaena variabilis) as well as their fragments embedded in isotropic and anisotropic polymer film were investigated. The orientation of photosynthetic pigments inside these organisms was compared, on the basis of the polarised absorption and fluorescence spectra, with the macroscopic orientation of investigated objects seen under microscope. The anisotropy of fluorescence was much higher than anisotropy of absorption. It showed strong influence of the photoselection by polarised radiation on the various bacterial chromophores exhibiting different orientations in the cells and various yields of fluorescence. The dimensions of cells were investigated on the basis of their photographs and by the scattering of the monochromatic radiation.  相似文献

Alginate-immobilized and free cells ofAnabaena doliolum andChlorella vulgaris were compared for their use in the removal and toxicity bioassays of Cu and Fe. A decrease in toxicity with regard to growth and uptake of NO 3 and NH 4 + was noticed following immobilization of both the organisms. In contrast, immobilized cells had higher uptake rates of Cu and Fe suggesting that immobilization offers protection against metal toxicity. Compared with free cells, the immobilized cells showed greater efficiency for metal removal, even over three repeated cycles, though with a gradual decrease in efficiency in the second and third cycles. This reduction in removal efficiency was, however, more pronounced for Fe withA. doliolum and for Cu withC. vulgaris. The ease in harvesting and potential for repeated use makes the immobilized cells good tools for scavenging heavy metals from metal-contaminated environments.  相似文献

1. This work investigated the consumption of a microalgal (Anabaena spiroides) exopolysaccharide by the cladoceran Simocephalus serrulatus (Cladocera, Daphnidae) and its effect on copper toxicity. 2. Total organic carbon concentration was used to quantify the microalgal exopolysaccharide. Both copper‐complexed exopolysaccharides and copper‐free exopolysaccharides were taken up by S. serrulatus. 3. A reduction of free copper ions was obtained in the presence of A. spiroides exopolysaccharide. Copper toxicity (EC50) and zooplankton concentrations were inversely correlated to exopolysaccharide concentration in experimental media. 4. An increase of free copper ions in experimental media was obtained after exopolysaccharide consumption, suggesting that S. serrulatus excreted copper to the surrounding environment.  相似文献

We acquired high spectral resolution reflectance data in Carter Lake, a eutrophic oxbow on the Iowa–Nebraska border, from April 1995 to April 1996. Chlorophyll a, total seston, sestonic organic matter, Secchi depth, and nephelometric turbidity were determined for each respective spectral measurement. Changes in algal taxonomic structure and abundance coincided with the development and senescence of a midsummer through autumn bloom of Anabaena. Taxonomic structure was more diverse in late winter and spring when Synedra sp. (diatom) and several chlorophytes and dinoflagellates were present. Overall, chlorophyll a varied from about 20 to 280 μg·L?1, Secchi transparency from 18 to 74 cm, and seston dry weight from 11 to 48 mg·L?1 in February and September, respectively. Particulate matter completely dominated lake water light attenuation. Dissolved organic matter had low optical activity. The most sensitive spectral feature to variation in chlorophyll a concentration was the magnitude of the scattering peak near 700 nm. The 700-nm peak correlated to chlorophyll concentration through the relationships between algal pigment absorption near 670 nm and the cell biomass and surface-related scattering signal in the near infrared. An algorithm relating the height of the 700-nm reflectance peak above a reference baseline between 670 and 850 nm to chlorophyll a was accurate and robust despite large variations in optical constituents caused by both strong seasonality in the algal system and short-term variations in seston from wind-induced sediment resuspension. The present algorithms were successfully used in other systems with different seasonality and productivity patterns. The coefficients of the models relating chlorophyll a and spectral reflectance variables appeared to be ecosystem specific: both the intercept and slope for the models in this study were moderately lower than for several other recently published results. We validated our algorithm coefficients with a second, independent dataset. The standard error for chlorophyll a prediction was ±28 μg·L?1.  相似文献

Akinetes of a clonal culture of Anabaena circinalis Rabenhorst from Mt. Bold reservoir (eutrophic), South Australia, were isolated and the effects of light, phosphorus, and nitrogen availability on their germination were investigated. Light was required but there was no significant difference in percentage of germination after 72 h if akinetes were incubated in ASM-1 medium at irradiances of 15, 30, or 50 μmol.m-2.s-1. Maximum akinete germination occurred by 48 h. Nitrogen was not required, as 88% of akinetes germinated in the flasks without combined nitrogen added to the medium and without N2 in the air. NH4+-N at 28 mg N.L-1 completely suppressed germination, whereas 28 mg NO3 N.L-1 had no effect relative to the controls without nitrogen. Phosphorus was required, and at 48 h percentage of germination in the flasks with 0.6 mg P.L-1 added (78%) was significantly greater than in the flasks with 0.06 P.L-1 (58%) and 0 mgP.L-1 (24%) added. Germlings in the 0 mg P.L-1 flasks were only 2–4 cells long and stunted in appearance, whereas germlings at all other P concentrations were 8–16 cells long. It is likely that the isolation process exposed some akinetes to intracellular phosphorus released from lysing vegetative cells, but this was insufficient to allow normal development in the 0 mg P.L-1 flasks. The plot of percentage of germination vs. initial phosphorus concentration, in the medium showed a relationship analogous to Michaelis-Menten nutrient uptake kinetics, suggesting that a specific membrane-bound enzyme system(s) is involved, with phosphorus as the substrate. The half saturation value (KS) for germination was 50 μg P.L-1.  相似文献
