在红肉脐橙幼果期和着色前分2次喷施不同浓度的外源ABA和GA3,研究其对红肉脐橙果肉糖含量的影响.结果表明:10mg.L-1ABA处理显著或极显著提高了果实成熟时的葡萄糖、果糖和总糖含量,50mg.L-1ABA处理极显著提高了果实蔗糖含量,而100mg.L-1ABA处理极显著降低了果实葡萄糖含量;中低浓度的GA3(10、50和250mg.L-1)极显著提高了果实蔗糖含量,10mg.L-1GA3处理对果实葡萄糖和果糖含量无明显影响,但极显著提高了果实总糖含量,50、250和500mg.L-1GA3处理极显著降低了果实葡萄糖、果糖和总糖含量.表明着色前较低浓度的外源ABA处理(10和50mg.L-1)可提高果实中一种或几种糖的含量,而较高浓度的GA3处理(250和500mg.L-1)则严重阻碍了果肉中糖的积累. 相似文献

Impaired germination is common among halophyte seeds exposed to salt stress, partly resulting from the salt-induced reduction of the growth regulator contents in seeds. Thus, the understanding of hormonal regulation during the germination process is a main key: (i) to overcome the mechanisms by which NaCl-salinity inhibit germination; and (ii) to improve the germination of these species when challenged with NaCl. In the present investigation, the effects of ABA, GA3, NO−3, and NH+4 on the germination of the oilseed halophyte Crithmum maritimum (Apiaceae) were assessed under NaCl-salinity (up to 200 mM NaCl). Seeds were collected from Tabarka rocky coasts (N-W of Tunisia). The exogenous application of GA3, nitrate (either as NaNO3 or KNO3), and NH4Cl enhanced germination under NaCl salinity. The beneficial impact of KNO3 on germination upon seed exposure to NaCl salinity was rather due to NO−3 than to K+, since KCl failed to significantly stimulate germination. Under optimal conditions for germination (0 mM NaCl), ABA inhibited germination over time in a dose dependent manner, but KNO3 completely restored the germination parameters. Under NaCl salinity, the application of fluridone (FLU) an inhibitor of ABA biosynthesis, stimulated substantially seed germination. Taken together, our results point out that NO−3 and GA3 mitigate the NaCl-induced reduction of seed germination, and that NO−3 counteracts the inhibitory effect of ABA on germination of C. maritimum. During leaf senescence, resources are recycled by redistribution to younger leaves and reproductive organs. Candidate pathways for the regulation of onset and progression of leaf senescence include ubiquitin‐dependent turnover of key proteins. Here, we identified a novel plant U‐box E3 ubiquitin ligase that prevents premature senescence in Arabidopsis plants, and named it SENESCENCE‐ASSOCIATED E3 UBIQUITIN LIGASE 1 (SAUL1). Using in vitro ubiquitination assays, we show that SAUL1 has E3 ubiquitin ligase activity. We isolated two alleles of saul1 mutants that show premature senescence under low light conditions. The visible yellowing of leaves is accompanied by reduced chlorophyll content, decreased photochemical efficiency of photosystem II and increased expression of senescence genes. In addition, saul1 mutants exhibit enhanced abscisic acid (ABA) biosynthesis. We show that application of ABA to Arabidopsis is sufficient to trigger leaf senescence, and that this response is abolished in the ABA‐insensitive mutants abi1‐1 and abi2‐1, but enhanced in the ABA‐hypersensitive mutant era1‐3. We found that increased ABA levels coincide with enhanced activity of Arabidopsis aldehyde oxidase 3 (AAO3) and accumulation of AAO3 protein in saul1 mutants. Using label transfer experiments, we showed that interactions between SAUL1 and AAO3 occur. This suggests that SAUL1 participates in targeting AAO3 for ubiquitin‐dependent degradation via the 26S proteasome to prevent premature senescence. 相似文献

Most aerial parts of the plant body are products of the continuous activity of the shoot apical meristem (SAM). Leaves are the major component of the aerial plant body, and their temporal and spatial distribution mainly determines shoot architecture. Here we report the identification of the rice gene PLASTOCHRON3 ( PLA3 )/ GOLIATH ( GO ) that regulates various developmental processes including the rate of leaf initiation (the plastochron). PLA3 / GO encodes a glutamate carboxypeptidase, which is thought to catabolize small acidic peptides and produce small signaling molecules. pla3 exhibits similar phenotypes to pla1 and pla2 – a shortened plastochron, precocious leaf maturation and rachis branch-to-shoot conversion in the reproductive phase. However, in contrast to pla1 and pla2 , pla3 showed pleiotropic phenotypes including enlarged embryo, seed vivipary, defects in SAM maintenance and aberrant leaf morphology. Consistent with these pleiotropic phenotypes, PLA3 is expressed in the whole plant body, and is involved in plant hormone homeostasis. Double mutant analysis revealed that PLA1 , PLA2 and PLA3 are regulated independently but function redundantly. Vacuoles have long been suggested to mediate a rise in the cytosolic free Ca2+ during environmental signal transduction. This study addresses the issue of the control of vacuolar calcium release by some of the known signaling molecules such as IP3, cADPR, ABA, ATP, cAMP, cGMP, H2O2 and CaM. Over 30 concentrations and/or combinations of these signaling compounds were studied in a series of electrophysiological experiments involving non-invasive ion flux measurements (the MIFE) and patch-clamp techniques. The root systems of cucumber (Cucumis sativus L.), wheat (Triticum aestivum L.), and barley (Hordeum vulgare L.) plants were subjected to a short-term (6–7-h-long) treatment with low and high temperatures, sodium chloride, and lead nitrate, and the effect of these treatments on the changes in the cold- and heat-tolerance of leaf cells was determined. It was established that chilling of cucumber and wheat seedling roots at 10 and 2°C, respectively, or their heating at 38°C and 40°C, respectively, induced an increase in both cold- and heat-tolerance of leaf cells. An increase in the cold- and heat-tolerance was also observed in roots treated with sodium chloride at concentrations of 0.15 M (cucumber) and 0.2 M (wheat), as well as with lead nitrate at a concentration of 0.1 mM (barley, wheat). The tolerance increase induced by these stress agents was accompanied by a considerable increase in the ABA concentration in leaves. The effect of physical and chemical stress agents is suggested to induce the same nonspecific changes in the aboveground organs. These changes bring about, directly or indirectly, an increase in the cold- and heat-tolerance and are related to an increase in the ABA content. 相似文献
