The oxygen binding behaviour of hemocyanins from Crustacea is regulated by small organic compounds such as urate and -lactate. We investigated the binding characteristics of urate and the related compound caffeine to the 2×6-meric hemocyanin of A. leptodactylus under fully oxygenated conditions employing isothermal titration calorimetry (ITC). An analysis of urate and caffeine binding based on a model of n identical binding sites resulted in approximately four binding sites for caffeine and eight for urate. This result suggests that the binding process for these effectors is more complex than this most simple model. Therefore, we introduced a number of alternative models. Displacement experiments helped to select the appropriate model. Based on these experiments, at least two different types of binding sites for urate and caffeine exist on the 2×6-meric hemocyanin of A. leptodactylus. The two binding sites differ strongly in their specificity towards the two analogues. It can be hypothesized that two different subunit types (β and γ) are responsible for the two types of binding sites.  相似文献

Summary Results of experiments on three passerine species suggest that brood size may be constrained by nest size, since the breeding success of pairs provided with large nestcups was greater than that of those provided with small artificial nestcups. These results may have important implications, e.g. to the design of experiments involving manipulation of clutch and brood size. A small nestcup is requisite for successful hatching during the incubation period, but a large one for successful rearing during the nestling period. In nature this difference may select for types of nesting materials that are elastic, such as mosses and lichens. However, experiments showed that such materials rapidly absorb rainwater but only slowly dry out. In addition, because large nests dry out more slowly than small nests, selection will favour small nests among those open-nesting species that have exposed nests. A further possible nest size constraint on open-nesters is nest predation. However, no difference in the predation rate was found in experiments with small and large artificial nests.  相似文献

In oviparous tetrapods, the nesting-site selection by females is related to the habitat characteristics, which influences nest incubation temperature. Females can directly influence the incubation temperature by choosing certain construction materials or by building nests of different sizes. There are few studies focusing on these aspects in crocodilians that build mound nests. The aim of this study was to determine whether the nest size, its exposure to solar radiation, and the environmental temperature influence the incubation temperature of Morelet's crocodile (Crocodylus moreletii) nests. Artificial nests of two sizes (small and large) were constructed with similar characteristics to natural nests and placed in two locations differing in exposure to solar radiation (shaded and sunny). We used temperature and relative humidity data loggers to record the incubation temperature inside each nest every hour during the species' natural nesting period. Likewise, we recorded the ambient temperature every hour where the experiments were set up with temperature data loggers. We found that nest size and its exposure to solar radiation affected the incubation temperature, with smaller nests in shaded locations having lower incubation temperatures than larger nests in sunny locations. We discuss the importance of nest-site selection and maintenance behaviour of the mound nest by female crocodiles on the incubation temperature of the nest.  相似文献
