The oomycete Albugo candida causes white rust of Brassicaceae, including vegetable and oilseed crops, and wild relatives such as Arabidopsis thaliana. Novel White Rust Resistance (WRR) genes from Arabidopsis enable new insights into plant/parasite co-evolution. WRR4A from Arabidopsis accession Columbia (Col-0) provides resistance to many but not all white rust races, and encodes a nucleotide-binding, leucine-rich repeat immune receptor. Col-0 WRR4A resistance is broken by AcEx1, an isolate of A. candida. We identified an allele of WRR4A in Arabidopsis accession Øystese-0 (Oy-0) and other accessions that confers full resistance to AcEx1. WRR4AOy-0 carries a C-terminal extension required for recognition of AcEx1, but reduces recognition of several effectors recognized by the WRR4ACol-0 allele. WRR4AOy-0 confers full resistance to AcEx1 when expressed in the oilseed crop Camelina sativa.  相似文献

Storing the eggs of T. pisiformis for periods of 0 to 9 days at a temperature of 36–38°C and relative humidity of 87–93 % revealed at least 4 stages in the ageing process. Firstly, a stage in which eggs hatched and activated in vitro and were also fully infective for rabbits. Secondly, a stage in which eggs hatched and activated in vitro but underwent only partial development in rabbits. Thirdly, a stage in which eggs hatched and activated in vitro, and presumably therefore in vivo, but no evidence of infection could be found when they were fed to rabbits. Fourthly, a stage in which eggs either did not hatch, or if they did hatch, the oncosphere was rapidly digested. In vitro techniques for assessing viability of T. pisiformis eggs were not a reliable guide to their infectivity for rabbits. ‘Senescent’ eggs, that is eggs which produced evidence of early infection in rabbits but did not complete their development to cysticerci, failed to produce immunity to challenge infection with T. pisiformis eggs.  相似文献

Interleukin-10 (IL-10) is recognized as an anti-inflammatory cytokine that downmodulates inflammatory immune responses at multiple levels. In innate cells, production of this cytokine is usually triggered after pathogen recognition receptor (PRR) engagement by pathogen-associated molecular patterns (PAMPs) or damage-associated molecular patters (DAMPs), as well as by other soluble factors. Importantly, IL-10 is frequently secreted during acute bacterial infections and has been described to play a key role in infection resolution, although its effects can significantly vary depending on the infecting bacterium. While the production of IL-10 might favor host survival in some cases, it may also result harmful for the host in other circumstances, as it can prevent appropriate bacterial clearance. In this review we discuss the role of IL-10 in bacterial clearance and propose that this cytokine is required to recover from infection caused by extracellular or highly pro-inflammatory bacteria. Altogether, we propose that IL-10 drives excessive suppression of the immune response upon infection with intracellular bacteria or in non-inflammatory bacterial infections, which ultimately favors bacterial persistence and dissemination within the host. Thus, the nature of the bacterium causing infection is an important factor that needs to be taken into account when considering new immunotherapies that consist on the modulation of inflammation, such as IL-10. Indeed, induction of this cytokine may significantly improve the host’s immune response to certain bacteria when antibiotics are not completely effective.  相似文献

Guinea pigs acquired resistance to Dermacentor andersoni larvae. Administration of the immunosuppressant, Methotrexate, prevented the acquisition of resistance. Resistant hosts allowed relatively very few larvae to engorge, and showed marked infiltration of the infested skin by basophil leucocytes. Particular high concentrations of basophils occurred in vesicles in the hyperplastic epidermis beneath the mouth-parts of attached larvae. These changes appear similar to Cutaneous Basophil Hypersensitivity reactions.  相似文献

Oysters, Crassostrea virginica, from two populations, one from a coastal pond experiencing repeated dinoflagellate blooms (native), and the other from another site where blooms have not been observed (non-native), were analyzed for cellular immune system profiles before and during natural and simulated (by adding cultured algae to natural plankton) blooms of the dinoflagellate Prorocentrum minimum. Significant differences in hemocytes between the two oyster populations, before and after the blooms, were found with ANOVA, principal components analysis (PCA) and ANOVA applied to PCA components. Stress associated with blooms of P. minimum included an increase in hemocyte number, especially granulocytes and small granulocytes, and an increase in phagocytosis associated with a decrease in aggregation and mortality of the hemocytes, as compared with oysters in pre-bloom analyses. Non-native oysters constitutively had a hemocyte profile more similar to that induced by P. minimum than that of native oysters, but this profile did not impart increased resistance. The effect of P. minimum on respiratory burst was different according to the origin of the oysters, with the dinoflagellate causing a 35% increase in the respiratory burst of the native oysters but having no effect on that of the non-native oysters. Increased respiratory burst in hemocytes of native oysters exposed to P. minimum in both simulated and natural blooms may represent an adaptation to annual blooms whereby surviving native oysters protect themselves against tissue damage from ingested P. minimum.  相似文献

用ELISA方法测定95例SARS患者急性期、恢复期血清细胞因子水平,用流式细胞仪检测该组患者急性期、恢复期淋巴细胞亚群,并分析细胞因子水平和淋巴细胞亚群变化的关系。结果发现,IL-10和TGF—β在观察期(急性期和恢复期)持续升高,急性期CD4^+和CD8^+T细胞显著减少,恢复期患者外周血CD8^+记忆T细胞减少36.78%。IL-10和TGF-β升高与淋巴细胞变化具有统计学相关。结果提示,SARS病毒感染抑制宿主的细胞免疫功能,引起急性期CD4^+和CD8^+T细胞显著降低和恢复期的免疫记忆细胞减少。而IL-10和TGF—β过表达可能在SARS免疫病理中起重要作用。  相似文献

A group of interesting molecules called defensins exhibit multiple functions but have been primarily recognized to possess a broad spectrum of antimicrobial activities. Studies have reported two different types of defensins (α and β) from human and animals, a cyclic θ defensin from rhesus, and several defensin-like peptides from plants. There is no amino acid sequence homology between these peptides, but they all contain three Cys-Cys disulfide linkages while the connectivities are different. Human β-defensin-3 (HβD-3) is the most recently discovered member of the host-defense peptide family that has attracted much attention. This molecule is expressed either constitutively or induced upon a challenge, and a growing evidence indicates the involvement of such molecules in adaptive immunity as well. It has been shown to exhibit antibacterial activities towards Gram-negative and Gram-positive bacteria as well as an ability to act as a chemo-attractant. Analysis of NMR structural data suggested a symmetrical dimeric form of this peptide in solution, which consists of three β strands and a short helix in the N-terminal region. While the disulfide linkages are known to provide the structural stability and stability against proteases, the biological relevance of this dimeric form was contradicted by another biological study. Since there is considerable current interest in developing HβD-3 for possible pharmaceutical applications, studies to further our understanding on the determinants of antibacterial activities and immunomodulatory function of HβD-3 are considered to be highly significant. The knowledge of its biosynthetic regulation will also help in understanding the role of HβD-3 in immunity. This article presents an overview of the expression and regulation of HβD-3 in humans, and the structure-function correlations among HβD-3 and its modified peptides are discussed emphasizing the functional importance. The future scope for studies on HβD-3 and design of short potent antimicrobial peptides, based on the native HβD-3 molecule, that do not interfere in the immunomodulatory function is also outlined.  相似文献

Stress may have consequences for the evolution of condition-dependentsexual traits. For example, stress may be related to sexualtraits through immune function, and sexual traits can reflecthow individuals bear the costs of stress-mediated immunosuppression.However, male traits may be directly associated with stress,and such traits would then indicate stress tolerance. Here,we present initial results for the relationship between physiologicalstress estimated by the levels of heat shock proteins (HSP60and HSP70) and heterophil/lymphocyte ratio and the elaborationof sexual traits, such as forehead and wing patch size and songfeatures in the collared flycatcher Ficedula albicollis. Malesproducing longer and more versatile songs had significantlyhigher levels of HSP70, but other traits were unrelated to stress.In general, effect sizes for the relationship between stressand sexual traits had broad confidence intervals and variedbetween being small and medium effects. Immunoglobulin levels,leukocyte abundance, haemoparasite prevalence, male age, anddate and time effects did not affect the relationship betweenstress and sexual traits. These preliminary results, servinga basis for further experimental studies indicate that the relationshipbetween sexual traits and stress does not seem to be strong,but stress may partially constrain the expression of some sexualtraits.  相似文献
